Aim: Skin burn is a health problem that requires fast and accurate treatment. If not well-treated, the burn will cause various damaging conditions for the patient. The leaf extract of Madeira vine (Anredera cordifolia (Ten.) Steenis), or popularly known as Binahong in Indonesia, has been used to treat various diseases. The purpose of this research is to determine the effects of leaf extracts of Madeira vine (A. cordifolia (Ten.) Steenis) on skin burn healing process in rats as an animal model.
Materials and Methods: In this research, there were four treatment groups: G0, G1, G2, and G3, each consisting of five rats. All these rats were given skin burns, using hot metal plates. Then, sulfadiazine was given to G0, 2.5% leaf extract of Madeira vine was given to G1, 5% extract was given to G2, and 10% extract was given to G3, for straight 14 days topically, 3 times a day. At the end of the treatment period, skin excisions were conducted, and histopathological examination was carried out.
Result: Microscopic observation on the wound healing process on the collagen deposition, polymorphonuclear infiltration, angiogenesis, and fibrosis showed that G2 had a significant difference with G0, G1, and G3 (p<0.05), while group G0 was significantly different from G1 and G3 (p<0.05). The better burn healing process on G2 allegedly because of the activity of flavonoid, saponin, and tannin, contained in the Madeira vine, which have the antioxidant, anti-inflammatory, and antibacterial effects.
Conclusion: The ointment from the 5% leaf extract of Madeira vine (A. cordifolia (Ten.) Steenis) has been proven to be effective to be used for topical burn therapy.

Effects of herbal ointment containing the leaf extracts of Madeira vine (Anredera cordifolia (Ten.) Steenis) for burn wound healing process on albino rats

Endophytes, especially those that are found from ethnopharmacologically noteworthy medicinal plants have attracted attention due to their diverse bioactive metabolites of pharmacological importance. This study aimed at isolating endophytic bacterium from the leaves of Anredera cordifolia CIX1 for its bioactive metabolites. The endophytic isolates were identified by 16S rRNA sequence and investigated for antibiotic sensitivity using different antibiotics. The secondary metabolites were evaluated for antibacterial activity against four bacterial strains. The 2-diphenyl-1-picrylhydrazyl (DPPH) and 2, 2′-azinobis (3- ethylbenzothiazoline-6-sulfonic acid) (ABTS) methods were used to assess their scavenging activities. The chemical components were analysed by gas chromatography-mass spectrometry (GC-MS). Out of 13 isolates, Isolate 1 was identified as Pseudomonas aeruginosa CP043328.1. It was resistant to clindamycin, ertapenem, penicillin G, amoxicillin, cephalothin and kanamycin but sensitive to imipenem, meropenem, and gentamycin. Its extract demonstrated antibacterial activity with minimum inhibitory concentration value of 0.098 against Bacillus cereus (ATCC 10102) and Staphylococcus aureus (ATCC 25925) and 0.391 mg/ml against Escherichia coli (ATCC 25922) and Proteus mirabilis (ATCC 25933). The extract revealed DPPH and ABTS scavenging activities with half maximal inhibitory concentration value of 0.650 mg/ml and 0.15 mg/ml, respectively. The GC-MS revealed a total of 15 compounds with diisooctyl phthalate (50.51%) and [1, 2, 4] oxadiazole, 5-benzyl-3 (10.44%) as major components. P. aeruginosa CP043328.1 produced secondary metabolites with antibacterial and antioxidant activities.

Isolation of endophytic bacteria from the leaves of Anredera cordifolia CIX1 for metabolites and their biological activities

Background: Wound caused by trauma poses challenging daily clinical problems for physicians in hospitals and other health services. It has high risk for mortality and morbidity. Binahong [Anredera Cordifolia (Ten.) Steenis] plant originates from China and can be found abundantly in Indonesia. The objective of this research is to investigate the effect of topical application of Binahong leaf paste on wound healing process. Methods : A laboratory experimental study was conducted at the Pharmacology Laboratory, Universitas Padjadjaran, from October to December 2012 by using 27 Mus Muscularis mice which were divided into 3 groups: group 1 (sodium chloride 0.9%), group 2 (Binahong) and group 3 (povidone iodine 5%). Binahong leaves were crushed and were mixed with water to make a paste. The application was given for 12 days. On day zero, 1.0 cm2 of wound was incised at the back of the mice, and further on day 1, 4, 8 and 12, the percentage of wound contraction was measured. The data was analyzed using Mann-Whitney test. Results : The study indicated that the percentage of wound contraction in group 2 on day 4 (1.17%), day 8 (23.7%) and day 12 (76.14%) is the highest among all groups. The result showed that the comparison between binahong and NaCl was highly significant on day 12, p= 0.001 (p < 0.05) and the data for comparison between Binahong and povidone iodine on day 12 was significant with p= 0.003 (p < 0.05). Conclusions : Topical application of Binahong leaf paste shows better result in wound healing process of Mus muscularis mice. From this study it can be concluded that binahong promotes wound healing and can be used for home remedies for wound care and as an alternative traditional way to treat wound.[AMJ.2014;1(1):6–11] Keywords: Binahong [Anredera Cordifolia (Ten.) Tujuan dari penelitian untuk mengetahui efek dari aplikasi pasta daun Binahong secara topikal terhadap proses penyembuhan luka. Metode: Penelitian eksperimental ini dilakukan di Laboratorium Farmakologi, Universitas Padjadjaran, mulai Oktober–Desember 2012, menggunakan tikus jenis MusMuscularis sebanyak 27 ekor, dibagi menjadi 3 kelompok: kelompok 1 (natrium klorida 0,9%), kelompok 2 (pasta daun Binahong) dan kelompok 3 (povidone iodine 5%). Daun Binahong dihancurkan dan ditambahkan sedikit air sehingga menjadi bentuk pasta. Perlakuan diberikan selama 12 hari. Pada hari 0 dilakukan insisi pada punggung tikus untuk membuat luka sebesar 1,0 cm2. Persentase kontraksi luka akan dinilai pada hari ke-1, 4, 8, dan 12. Kontraksi luka adalah pengurangan luas pada area luka. Hasil penilaian tersebut akan dibandingkan pada tiap kelompok untuk melihat efektivitas aplikasi topikal pasta daun Binahong. Data akan dianalisis dengan uji Mann-Whitney. Hasil : Hasil penelitian menunjukkan bahwa persentase kontraksi luka kelompok 2 pada hari ke-4 (1,17%), hari ke-8 (23,7%), dan hari ke-12 (76,14%) adalah lebih tinggi dibandingkan dengan kelompok lainnya. Hasil juga menunjukkan bahwa terdapat perbedaan sangat signifikan antara perbandingan Binahong dan natrium klorida pada hari ke-12, dengan p=0,001 (p<0,05). Perbandingan antara Binahong dan povidone iodine pada hari ke-12 juga menunjukkan hasil signifikan dengan p=0,003 (p<0,05). Simpulan : Aplikasi pasta daun Binahong secara topikal menunjukkan hasil yang lebih baik dalam proses penyembuhan luka pada tikus jenis Musmuscularis. Dari penelitian ini dapat disimpulkan bahwa Binahong mempercepat penyembuhan luka serta dapat digunakan untuk perawatan luka di rumah dan sebagai cara alternatif yang bersifat tradisional untuk mengobati luka. Effect of Topical Application of Binahong [Anredera cordifolia (Ten.) Steenis] Leaf Paste in Wound Healing Process in Mice

Anredera cordifolia (Tenore) Steenis is widely planted as an ornamental and medicinal plant in Indonesia. On the other hand, in some other countries this plant is classified as a noxious weed. As a harmful weed, A. cordifolia is reported to have the ability to smother all native vegetation, collapse canopies of tall trees, cultivate as a ground cover and disrupt native seedling development. There is no available information about the involvement of any allelochemicals from A. cordifolia related to these issues. The present study evaluated the allelopathic effect by isolating and identifying the allelopathic substance from A. cordifolia leaf extract. The allelopathic potency of A. cordifolia was determined by a series of bioassays of shoot and root growth on some selected test plants. Separation and purification of the active substances was achieved through several chromatography processes. Finally, the substances with allelopathic activity were identified through high-resolution electrospray ionization mass spectrometry (HRESIMS) analysis and determined by the specific rotation of compound, proton and carbon NMR spectroscopies. The results show that A. cordifolia possesses allelopathic properties which affect other plant species. The isolated compound from the plant material, 3-hydroxy-alpha-ionone, may contribute to the allelopathic effects of A. cordifolia.

Allelopathic Potency and an Active Substance from Anredera cordifolia (Tenore) Steenis.

Saponin has various types; it can be bond with glycosides that form soapy lathers when mixed and agitated with water, and have been used to treat diabetes; liver, hepatitis, cardiovascular as high blood pressure, high cholesterol, and physical stress. In this study describe of saponins compound in Binahong, one of medicinal plant investigated from these plant probably contribute to the effectiveness the above plant for treatment several diseases. The sample were dried by oven (60 o C) to be powder and extracted with solvents. After that used qualitative and quantitative test. The result of Binahong plant contains saponins compound on all a part of the Binahong plant, are positive indicate of presence of saponins triterpenoid and steroid. . Moreover crude of saponins substances from Leaves (28.14±0.22), Stems (3.65±011) and Tubers (43.15±0.10) of mg/g. The research showed of the Binahong plant, importance of the distribution of determination of saponins compound from plant, has a potential to treatment for several diseases
