Traditional Chinese herbal medicine as a source of molecules with antiviral activity.

Background Chinese herbal medicine has a long history of human use. A novel herbal formula, antiasthma herbal medicine intervention (ASHMI), has been shown to be an effective therapy in a murine model of allergic asthma. Objective This study was undertaken to compare the efficacy, safety, and immunomodulatory effects of ASHMI treatment in patients with moderate-severe, persistent asthma with prednisone therapy. Methods In a double-blind trial, 91 subjects underwent randomization. Forty-five subjects received oral ASHMI capsules and prednisone placebo tablets (ASHMI group) and 46 subjects received oral prednisone tablets and ASHMI placebo capsules (prednisone group) for 4 weeks. Spirometry measurements; symptom scores; side effects; and serum cortisol, cytokine, and IgE levels were evaluated before and after treatment. Results Posttreatment lung function was significantly improved in both groups as shown by increased FEV 1 and peak expiratory flow findings ( P P 2 -bronchodilators, and serum IgE levels were reduced significantly, and to a similar degree in both groups ( P H 2 cytokine levels were significantly reduced in both treated groups ( P P P P Conclusion Antiasthma herbal medicine intervention appears to be a safe and effective alternative medicine for treating asthma. In contrast with prednisone, ASHMI had no adverse effect on adrenal function and had a beneficial effect on T H 1 and T H 2 balance.

Efficacy and tolerability of antiasthma herbal medicine intervention in adult patients with moderate-severe allergic asthma

Viral diseases, such as acquired immunodeficiency syndrome (AIDS), respiratory diseases, and hepatitis, are the leading causes of death in humans worldwide, despite the tremendous progress in human medicine. The lack of effective therapies and/or vaccines for several viral infections, and the rapid emergence of new drug-resistant viruses have urged a growing need for developing new and effective chemotherapeutic agents to treat viral diseases. Recent advances in the understanding of both the cellular and molecular mechanisms of virus replication have provided the basis for novel therapeutic strategies. Several hundred natural products have been isolated for screening and identifying antiviral activity, and some have been shown to have great medicinal value in preventing and/or ameliorating viral diseases in preclinical and clinical trials. There are innumerable potentially useful medicinal plants and herbs waiting to be evaluated and exploited for therapeutic applications against genetically and functionally diverse virus families. This review focuses on several selected pathogenic viruses, including the human immunodeficiency virus (HIV), influenza virus, hepatitis B and C viruses and herpes viruses, and antiviral natural compounds from medicinal plants (herbs), while paying particular attention to promising compounds in preclinical and clinical trials. We also focused our attention on the need to develop effective screening systems for antiviral activity.

As the leading cause of death worldwide, viruses significantly affect global health. Despite the rapid progress in human healthcare, there are few viricidal and antiviral therapies that are efficient enough. The rapid emergence of resistance, and high costs, as well as the related side effects of synthetic antiviral drugs, raise the need to identify novel, effective, and safe alternatives against viral diseases. Nature has been of the most exceptional help and source of inspiration for developing novel multi-target antiviral compounds, affecting several steps of the viral life cycle and host proteins. For that matter and due to safety and efficacy limitations, as well as high resistance rate of conventional therapies, hundreds of natural molecules are preferred over the synthetic drugs. Besides, natural antiviral agents have shown acceptable antiviral value in both preclinical and clinical trials.This is the first review regarding molecular and cellular pathways of the virus life cycle, treatment strategies, and therapeutic targets of several viral diseases with a particular focus on anthocyanins as promising natural compounds for significant antiviral enhancements. Clinical applications and the need to develop nano-formulation of anthocyanins in drug delivery systems are also considered.

The Signaling Pathways, and Therapeutic Targets of Antiviral Agents: Focusing on the Antiviral Approaches and Clinical Perspectives of Anthocyanins in the Management of Viral Diseases.

To evaluate the effects of radix Sophorae flavescentis for chronic hepatitis B, a systematic review of randomized clinical trials was conducted. Randomized trials comparing extract of radix Sophorae flavescentis versus placebo, no intervention, non-specific treatment, other active medicines, or interferon for chronic hepatitis B were identified by electronic and manual searches. Trials of Sophorae herb plus other drugs versus other drugs alone were also included. No blinding and language limitations were applied. The methodological quality of trials was assessed by the Jadad scale plus allocation concealment. Meta-analysis was performed where data was available. Twenty-two randomized trials (n = 2409) were included. Methodological quality of the trials was generally low. The combined results showed that matrine (aqueous extract of Sophorae flavescentis) had antiviral activity, positive liver biochemical effects, and improved symptoms and signs compared with non-specific treatment and other herbal medicines. The combination of matrine and interferon-α (IFN-α), thymosin, or basic treatment showed better effects on viral and liver biochemical responses. The antiviral and biochemical responses were not significantly different between matrine and IFN-α. No serious adverse event was reported. Based on the review, Sophorae flavescentis extract (matrine) may have antiviral activity and positive effects on liver biochemistry in chronic hepatitis B. However, the evidence is not sufficient to recommend matrine for routine clinical use due to the generally low methodological quality of the studies. Further rigorous trials are needed.
