Es wird der Versuch unternommen, wichtige Entwicklungen auf dem Gebiet der Organischen Synthese in den letzten 25 Jahren zu skizzieren und in die Zukunft zu projizieren. — Die wichtigsten Motive fur die Durchfuhrung einer Naturstoffsynthese existieren nicht mehr: Statt der Zielstruktur als solcher steht heute deren Funktion und Wirksamkeit im Mittelpunkt des Interesses. Inhibitoren mit Affinitat zu allen wichtigen Enzymen und Rezeptoren der belebten Natur werden zu Zielmolekulen. — Neue Synthesemethoden werden am ehesten auf dem Gebiet der biologisch-chemischen und der metallorganischen Chemie entdeckt werden. Enzyme, ganze Mikroorganismen oder Zellkulturen werden schon heute im Forschungslabor wie auch in der Industrie zur enantioselektiven Herstellung bestimmter Verbindungen verwendet. Durch den Entwurf geeigneter Analoga von Ubergangszustanden und Zwischenprodukten wird es zusatzlich moglich sein, unter Zuhilfenahme des Saugetier-Immunsystems und der Gentechnologie katalytisch wirksame monoklonale Antikorper fur praktisch beliebige Reaktionen herzustellen und vor allem im industriellen Masstab einzusetzen. Wirklich neue Reaktionen wird es wohl nur noch auf dem Gebiet der ubergangsmetallorganischen Chemie geben, die uns in den nachsten Jahren weitere „Wunderreagentien” bescheren wird. Bei den Derivaten der Hauptgruppenelemente („elementorganische” Chemie) werden schrittweise Verbesserungen in der Versuchsfuhrung und die Anwendung spezieller Techniken zu ungeahnter Effizienz und Selektivitat bekannter Prozesse fuhren. Das Schwergewicht wird bei allen Synthesemethoden auf der Suche nach katalytischen und enantioselektiven Varianten liegen: Es wird nicht lange dauern, bis fur alle Standardreaktionen, die von achiralen Edukten zu chiralen Produkten fuhren, entsprechende Modifikationen zur Verfugung stehen werden. Analytik, Spektroskopie, Strukturanalyse, Theorie und elektronische Datenverarbeitung sind fur den Fortschritt auf dem Gebiet der Synthese unentbehrlich. Nur durch diese „Hilfsmittel” werden immer grosere und kompliziertere Systeme mit den Methoden der Chemie, d. h. auf molekularer oder supramolekularer Ebene, erfasbar. — Zu allen erwahnten Themen werden Beispiele angefuhrt; eine sehr umfangreiche Literaturliste11 soll das weitere Studium erleichtern. — Wenn dieser Artikel dazu fuhrt, das weniger Kollegen als bisher die Organische Chemie als eine reife Wissenschaft ansehen und das der Nachwuchs erkennt, wie lebendig und kraftvoll die Organische Synthese sich neuen Zielen zuwendet und alte Traume Wirklichkeit werden last, hat er seinen Zweck erfullt.

Organische Synthese — wohin?

Cyclic enaminones were synthesized in high yields from amino acids in two steps via Wolff rearrangement. The cyclization represents a rare 6-exo-dig cyclization involving a ketene as an electrophile. No racemization was observed during this reaction.

The construction of a highly functionalized adamantane core of plukenetione-type polycyclic polyprenylated acylphloroglucinols (PPAPs) is described. The method features the construction of the bicyclo[3.3.1]nonane core (3) by successive Michael reactions and the construction of the adamantane core of plukenetione-type PPAPs by acid-catalyzed cyclization of a bicyclo[3.3.1]nonane precursor (2).

3-Azabicyclic Ketones from 1,3-Dialkoxy-2-azapropenylium Salts and Enamines of Cyclic Ketones



1,3-Diethoxy-2-azapropenylium salts 1 are powerful biselectrophiles and react diastereoselectively with enamines of five-membered ring ketones 2 to yield novel 3-azabicyclo[3.2.1]oct-2-en-8-one 5 after hydrolysis of the intermediate iminium salts 3, 4. Some of the iminium salts 3 can be hydrolyzed further to provide the diketo imines 6. With enamines of six-membered ring ketones 7 the 3-azabicyclo[3.3.1]non-2-en-9-ones 10, 11 are obtained after hydrolytic workup. Mixtures of diastereomers 10/11 are found in the case of 7a, b, whereas with 7g — j the endo-phenyl derivatives 10 are formed exclusively. With the unsymmetrical enamine 7b only one regioisomer is observed. The decalone enamine 12 reacts with 1a to yield the tricyclic iminium salt 13. — The configuration of the bicyclic products has been determined by using NMR spectroscopic techniques (COSY, polarisation transfer by long range couplings); an X-;Ray analysis of compound 10a is used as basis for the NMR assignments. General rules for the determination of the stereochemistry of these azabicyclic systems by 1H-NMR shift arguments are derived.

3-Azabicyclische Ketone durch Reaktionen von 1,3-Diethoxy-2-azapropenylium-Salzen mit Enaminen cyclischer Ketone

Chemical, spectroscopic and X-ray crystallographic evidence has confirmed that the three isomers of 6-hydroxy-7,9-dimethyl-6-phenyladamantane-2,4-dione, obtained by the reaction of crotonoyl chloride with the pyrrolidine enamine of 4-benzoyl-4-methylcyclohexanone, have the 9α-methyl-6α-phenyl (4), 9β-methyl-6α-phenyl (2), and 9β-methyl-6β-phenyl (3) configurations. The mechanism of their formation has been investigated and the evidence available indicates that the stereochemistry of the C-6 and C-9 chiral centres is determined largely by the stereochemistry of the [3,3] sigmatropic rearrangement leading to them. A chair-like transition state for the rearrangement, occurring from the same side of the cyclohexene ring as the benzoyl group (chair-syn) in the intermediate N-acylated enamine (9a) must lead to the 9α-Me,6α-Ph configuration as in (4), whereas the same process from the opposite side (chair-anti) followed by inversion of the substituted ring carbon could lead to the 9β-Me,6β-Ph configuration as in (2) and the 9β-Me,6α-Ph configuration as in (3). However, the main isomer (3), having the more crowded 9β-Me,6β-Ph configuration, is more likely to be derived by a [3,3] sigmatropic rearrangement having a boat-like transition state, but again occurring from the same side of the cyclohexene ring as the benzoyl group (boat-syn). An X-ray crystallographic analysis of 6β- hydroxy-7,9α-dimethyl-6α-phenyladamantane-2,4-dione (4) and of 3-(5-benzoyl-5-methyl-2-oxocyclohexyl)-butanoic acid (34), isolated at an intermediate stage in the above reaction, has been carried out. The corresponding reaction of αβ-unsaturated acid chlorides with enamines of 4-benzoyl-4-phenyl-cyclohexanone (9c) and ethyl 1 -benzoyl-4-oxocyclohexane-1 -carboxylate (9b) has been carried out and shown to lead to 6-hydroxy-6,7-diphenyladamantane-2,4-diones and ethyl 2-hydroxy-4,6-dioxo-2-phenyladamantane-1 -carboxylates respectively, as a mixture of stereoisomers. A similar multiplicity in the stereochemistry of the transition state of the [3,3] sigmatropic rearrangement is evident from the structures of the adamantane derivatives produced.
