The (2,4-dinitrophenyl)hydrazones of carbonyls are separated by liquid chromatography and detected by ultraviolet spectroscopy (diode array detector) and by atmospheric pressure negative chemical ionization mass spectrometry. Results are presented for 78 carbonyls including 18 1-alkanals (from formaldehyde to octadecanal), 16 other saturated aliphatic carbonyls (5 C4−C7 aldehydes and 11 C3−C9 ketones), 16 unsaturated aliphatic carbonyls (9 C3−C11 aldehydes and 7 C4−C9 ketones), 13 aromatic carbonyls (including hydroxy- and/or methoxy-substituted compounds), 10 C2−C10 aliphatic dicarbonyls, 3 aliphatic carbonyl esters, and 2 other carbonyls. Isomers were observed for α,β-unsaturated ketones and saturated carbonyls that bear other oxygen-containing substituents, e.g. methoxyacetone, 2-furaldehyde, and the 3 carbonyl esters. Liquid Chromatography Analysis of Carbonyl (2,4-Dinitrophenyl)hydrazones with Detection by Diode Array Ultraviolet Spectroscopy and by Atmospheric Pressure Negative Chemical Ionization Mass Spectrometry.

Ion trap multiple fragmentation mass spectrometry (MSn) combined with high-performance liquid chromatography (HPLC) has been used for the structure elucidation and identification of 2,4-dinitrophenylhydrazone derivatives of carbonyl compounds in ambient air samples. Atmospheric pressure chemical ionization in the negative ion mode was the most suitable detection method. Different measures are described to decrease the MS background originating from the HPLC system. Low-picogram quantities were detectable in extracted mass chromatograms generated from full-scan records. Fragment ions produced by MS/MS allowed identification of substructures of the carbonyls. Detailed fragmentation paths were studied by MS3 to MS4 using reference compounds. A fragmentation scheme was established which enabled a structure confirmation and identification with 1−10 ng by HPLC/MS/MS. The identification of a compound coeluting with n-pentanal-DNPH and of a dimerization byproduct are given as examples.

Structure Elucidation of 2,4-Dinitrophenylhydrazone Derivatives of Carbonyl Compounds in Ambient Air by HPLC/MS and Multiple MS/MS Using Atmospheric Chemical Ionization in the Negative Ion Mode.

The wounding and drying of plant material during crop harvest could be a significant source of volatile organic compounds (VOCs) that enter the atmosphere. Here, we show that these primarily oxygenated VOCs can be measured using proton-transfer chemical-ionization mass spectrometry (PT-CIMS), a method that allows online and simultaneous monitoring of oxygenated VOC levels. For clover, alfalfa, and corn, leaf wounding and in particular drying were shown to lead to strongly enhanced emissions of a series of C6 aldehydes, alcohols, and esters derived from (Z)-3-hexenal. Additionally, for the forage crops clover and alfalfa, enhanced emissions of methanol, acetaldehyde, acetone, and butanone were observed. The identities of the measured carbonyl compounds were confirmed using high-pressure liquid chromatography. Aldehydes and ketones readily react with 2,4-dinitrophenylhydrazine (2,4-DNPH) to form the corresponding hydrazones. This reaction has been frequently used for the quantification of airborne carbonyl compounds. Since unsymmetrical aldehydes and ketones are known to form isomeric 2,4-dinitrophenylhydrazones (syn/ anti-isomers), the influence of isomerization on the practicability and accuracy of the 2,4-DNPH-method using 2,4-dinitrophenylhydrazine-coated solid sorbent samplers has been studied with three ketones (methyl ethyl ketone (MEK), methyl isopropyl ketone (MIPK), and methyl isobutyl ketone (MIBK)). With all three ketones the reaction with 2,4-DNPH resulted in mixtures of the isomeric hydrazones which were separated by HPLC and GC and identified by mass spectroscopy and (1)H nuclear magnetic resonance spectroscopy. The isomers show similar chromatographic behaviour in HPLC as well as in GC, thus leading to problems in quantification and interpretation of chromatographic results.
