The invention relates to a method for determining the content of vincamine acid by a non-aqueous titration method, which is applied to the field of chemical analysis. The vincamine acid is dissolved quantitatively with a mixed solvent of anhydrous formic acid, glacial acetic acid and acetic anhydride, a perchloric acid solution is taken as a titrant, and the titration is carried out by a potentiometric titration method or an indicator titration method; the potentiometric titration method means that a potentiometric indicator is used for determining a titration end point and calculating the content of the vincamine acid; the indicator titration method means that crystal violet is taken as an indicator for determining a titration end point and calculating the content of the vincamine acid; the volume of the mixed solvent is directly equal to the sum of the volumes of all the components for preparing the mixed solvent; in the method of quantitatively dissolving the vincamine acid, the vincamine acid is dissolved first with the anhydrous formic acid, and then the glacial acetic acid and the acetic anhydride are added to the solution; and the perchloric acid solution takes anhydrous glacial acetic acid as a solvent. The method has the advantages of small amount of samples, simplicity in instrument equipment, simplicity and convenience in operation, saving of time and materials, easy availability of raw materials, low cost, easiness of grasping, high precision, accurate content and good repeatability.

Method for determining content of vincamine acid by non-aqueous titration method

The invention discloses a method for measuring content of total sugars in total flavonoids of herba desmodii styracifolii. According to an embodiment, the method comprises steps as follows: (1) the total flavonoids of herba desmodii styracifolii are extracted with water or ethanol, and a total sugar extracting solution is obtained; (2) a 4% phenol reagent and sulfuric acid are added to the total sugar extracting solution, and a to-be-measured solution is obtained; (3) the to-be-measured solution is detected under the wavelength of 487 nm with an ultraviolet spectrophotometry, the absorbancy isobtained, and the content of total sugars in total flavonoids of herba desmodii styracifolii is calculated with a standard curve method. Accordingly, the content of total sugars in total flavonoids of herba desmodii styracifolii can be rapidly and accurately measured with the method.

PURPOSE:To quantify vincamine acid of an extremely small amount in ingredients of a living body by extracting a sample, purifying and differentiating the same later, and measuring the fluorescence thereof. CONSTITUTION:Urine is diluted with distilled water, 0.01M disodium phosphate of 1.5ml per 1ml of the diluted urine is added thereto, saturation thereof with common salt is conducted, and later pH thereof is adjusted to be 1.5 with hydrochloric acid. Then, a mixture liquid of isopropyl alcohol and chloroform (1:19) is added and, after extraction, an organic layer is dried with anhydrous sodium sulfate. The layer is put in a column for which silica gel is used, cleansed by a mixture liquid of ethyl alcohol and chloroform (1:1) and elutriated later with a mixture liquid of 5ml of ethyl alcohol containing 0.08N triethanolamine and of chloroform (4:1). This elution liquid is destilled, a liquid of 4ml wherein ethyl alcohol and water are mixed equally is added to the residue, the fluorescent intensity thereof is measured, hydrochloric acid is then added thereto, the same measurement is conducted, and the difference between the former and the latter is taken, whereby the value of fluorescence is corrected. When measurement is conducted by this method after the vincamine of 10mg is given orally to a person to be inspected, the result as shown in the figure is obtained. From the figure it is clear that other effects are separated therefrom.
