Modification of collagen with a natural derived cross-linker, alginate dialdehyde

Collagen, a prominent biopolymer, which is famous for its excellent biological activity, has been used extensively for tissue engineering applications. In this study, a novel solvent system for collagen was developed with an ionic liquid, 1-ethyl-3-methylimidazolium acetate ([EMIM][Ac]), solvent system. A series of sodium salts were introduced into this solvent system to enhance collagen's dissolution procedure. The results show that the solubility of collagen was significantly influenced by the temperature and sodium salts. The solubility reached up to approximately 11% in the [EMIM][Ac]/Na2HPO4 system at 45°C. However, the structure of the regenerated collagen (Col-regenerated) may have been damaged. Hence, we focused on the structural integrity of the collagen regenerated from the [EMIM][Ac] solvent system by the methods of sodium dodecyl sulfate–polyacrylamide gel electrophoresis, Fourier transform infrared spectroscopy, ultrasensitive differential scanning calorimetry, atomic force microscopy, X-ray diffraction, and circular dichroism because its signature biological and physicochemical properties were based on its structural integrity. Meanwhile, a possible dissolution mechanism was proposed. The results show that the triple-helical structure of collagen regenerated from the [EMIM][Ac] solvent system below 35°C was retained to a large extent. The biocompatibility of Col-regenerated was first characterized with a fibroblast adhesion and proliferation model. It showed that the Col-regenerated had almost the same good biological activity as nature collagen, and this indicated the potential application of [EMIM][Ac] in tissue engineering. © 2013 Wiley Periodicals, Inc. This review provides a report on properties and recent advances in the application of collagen in cosmetics. Collagen is a structural protein found in animal organisms where it provides for the fundamental structural support. Most commonly it is extracted from mammalian and fish skin. Collagen has attracted significant academic interest as well as the attention of the cosmetic industry due to its interesting properties that include being a natural humectant and moisturizer for the skin. This review paper covers the biosynthesis of collagen, the sources of collagen used in the cosmetic industry, and the role played by this protein in cosmetics. Future aspects regarding applications of collagen-based materials in cosmetics have also been mentioned.

The invention discloses a collagen-polyvinyl alcohol-chitosan blending medical fiber and the preparation method thereof. The invention is characterized by the following steps: 25-50 parts of collagen are dissolved in acetic acid solution with the concentration of 0.2-0.5mmol/L for preparing a solution; 20-40 parts of polyvinyl alcohol, and 16-40 parts of chitosan are respectively dissolved in distilled water at 85-95 DEG C and 40-60 DEG C for preparing solutions with different concentrations; the solutions are mixed in proportion and are uniformly stirred into spinning raw liquor; a wet-spinning machine carries out spinning and consolidation forming, and the collagen-polyvinyl alcohol-chitosan blending medical fiber can be obtained by processing after modification by adopting carbodiimide or aldehyde system.

The invention discloses an acellular dermal matrix complex film material and a preparation method thereof. The method is characterized in that: 18 to 28 weight portions of collagen is dissolved into a solution by an acetic acid aqueous solution with a concentration between 0.2 and 0.5mmol/L; 10 to 24 weight portions of polyvinyl alcohol and 10 to 40 weight portions of chitosan are respectively dissolved into solutions with different concentrations by distilled water at a temperature of between 85 and 95 DEG C and at a temperature of between 40 and 60 DEG C, and the collagen solution and the polyvinyl alcohol solution and/or the chitosan solution are mixed according to the proportion and then are evenly stirred to prepare a mixed liquid; the mixed liquid is evenly coated on the surface of an acellular dermal matrix material to ensure that the thickness of a film reaches between 0.5 and 2.5cm and the film is quickly frozen, and then the acellular dermal matrix complex film material is produced through freeze drying; and finally carbodiimide is adopted to modify so as to prepare an acellular dermal matrix composite collagen-polyvinyl alcohol and/or chitosan film material.
