Increasing interest in the health benefits of tea has led to the inclusion of tea extracts in dietary supplements and functional foods. However, epidemiologic evidence regarding the effects of tea consumption on cancer and cardiovascular disease risk is conflicting. While tea contains a number of bioactive chemicals, it is particularly rich in catechins, of which epigallocatechin gallate (EGCG) is the most abundant. Catechins and their derivatives are thought to contribute to the beneficial effects ascribed to tea. Tea catechins and polyphenols are effective scavengers of reactive oxygen species in vitro and may also function indirectly as antioxidants through their effects on transcription factors and enzyme activities. The fact that catechins are rapidly and extensively metabolized emphasizes the importance of demonstrating their antioxidant activity in vivo. In humans, modest transient increases in plasma antioxidant capacity have been demonstrated following the consumption of tea and green tea catechins. The effects of tea and green tea catechins on biomarkers of oxidative stress, especially oxidative DNA damage, appear very promising in animal models, but data on biomarkers of in vivo oxidative stress in humans are limited. Larger human studies examining the effects of tea and tea catechin intake on biomarkers of oxidative damage to lipids, proteins, and DNA are needed.

Protection against ionizing radiation by antioxidant nutrients and phytochemicals

The development of radioprotective agents has been the subject of intense research in view of their potential for use within a radiation environment, such as space exploration, radiotherapy and even nuclear war. However, no ideal, safe synthetic radioprotectors are available to date, so the search for alternative sources, including plants, has been on going for several decades. In Ayurveda, the traditional Indian system of medicine, several plants have been used to treat free radical-mediated ailments and, therefore, it is logical to expect that such plants may also render some protection against radiation damage. A systematic screening approach can provide leads to identifying potential new candidate drugs from plant sources, for mitigation of radiation injury. This article reviews some of the most promising plants, and their bioactive principles, that are widely used in traditional systems of medicine, and which have rendered significant radioprotection in both in vitro and in vivo model systems. Plants and their constituents with pharmacological activities that may be relevant to amelioration of radiation-mediated damage, including antiemetic, antiinflammatory, antioxidant, cell proliferative, wound healing and haemopoietic stimulatories are also discussed.

Radioprotection by plant products: present status and future prospects.

Cinnamon has been used as a spice and as traditional herbal medicine for centuries. The available in vitro and animal in vivo evidence suggests that cinnamon has anti-inflammatory, antimicrobial, antioxidant, antitumor, cardiovascular, cholesterol-lowering, and immunomodulatory effects. In vitro studies have demonstrated that cinnamon may act as an insulin mimetic, to potentiate insulin activity or to stimulate cellular glucose metabolism. Furthermore, animal studies have demonstrated strong hypoglycemic properties. However, there are only very few well-controlled clinical studies, a fact that limits the conclusions that can be made about the potential health benefits of cinnamon for free-living humans. The use of cinnamon as an adjunct to the treatment of type 2 diabetes mellitus is the most promising area, but further research is needed before definitive recommendations can be made.

History and development of radiation-protective agents

Garlic extract was evaluated in the mouse bone marrow micronucleus test for its possible protective effects against gamma-radiation-induced chromosomal damage. Together with this, biochemical assays were carried out to determine the changes in sulfhydryl content and glutathione S-transferase activities. Three doses of freshly prepared garlic extract (125, 250 and 500 mg/kg b.w.) were orally administered for 5 consecutive days, and the animals were irradiated 2 h after the final feeding. The results of the micronucleus test demonstrated that pre-treatment with garlic extract can lead to significant dose-related reductions in the frequencies of gamma-radiation-induced (2 Gy) micronucleated polychromatic erythrocytes. The anticlastogenic effect of garlic extract was observed against lower radiation doses of 0.5 and 1 Gy, but not 0.25 Gy. Significant increases in the sulfhydryl content and glutathione S-transferase activity were observed after either pre-treatment with garlic extract or irradiation. However, the irradiated garlic-extract pre-treated animals showed a significant reduction in sulfhydryl content and glutathione S-transferase activities.

In vivo radioprotection with garlic extract

Antigenotoxic effects and changes in glutathione S-transferase (GST) activity were assessed in mice after oral co-administration of urethane (URE) with aqueous extracts of dietary vegetables (carrot, spinach and cabbage), spices (cinnamon, pepper, cumin, clove and cardamom), tea and coffee. The results of the genotoxicity assay (micronucleus test) demonstrated dose-related antigenotoxic effects after URE was co-administered with aqueous extracts of vegetables, spices, tea and coffee. Inhibition of GST activity was observed 4 h after treatment with URE alone. Co-administration of URE with extracts of vegetables, coffee and spices resulted in dose-related attenuation of the inhibitory effect of URE on GST activity. However, tea had no effect on inhibition of GST activity by URE. Hence an association between antigenotoxicity and GST activity could not be established.
