Promises and challenges of targeting Bcl-2 anti-apoptotic proteins for cancer therapy.

TL;DR: A substantial number of small molecules that bind to BCL-2 or BCL -XL are now available for pre-clinical testing; in turn, basic studies with these reagents should yield new insights about optimal strategies to disrupt B CL-2 survival functions.
Journal Article

Small-molecule inhibitors of Bcl-2.

TL;DR: Two Bcl-2 small-molecule inhibitors are currently undergoing phase I/II clinical trials and several other compounds are in preclinical development, and ongoing studies with these investigational drugs should provide new insights into optimal strategies to disrupt B cl-2 survival functions to selectively kill cancer cells.
