Publisher Summary This chapter summarizes current data concerning the ultrastructural localization of transcription sites and the subsequent distribution of newly synthesized ribonucleic acid (RNA) within the cell nucleus. It discusses the roles of ribonucleoprotein (RNP)-containing nuclear structures with respect to the synthesis and processing of nucleolar and extranucleolar RNA. RNA transcription in the nucleolar, as well as extranucleolar, areas takes place predominantly within the regions situated on the border of condensed chromatin. Perichromatin fibrils, considered as the first in situ detected structures containing newly transcribed extranucleolar RNA, originates in these perichromatin regions and the nucleolar fibrillar components are in intimate contact with intranucleolar chromatin areas. DNA replication sites also appear predominantly within this nuclear region. This particular nuclear area consequently plays a key role in the expression of chromatin functions and these findings lead to the conclusion that the layer of chromatin decondensing on the periphery of dense chromatin areas represents the most active chromatin fraction in the nucleus.

Publisher Summary Chemical and physical agents can lead to selective cytological lesions if the treatment is suitably chosen to dissociate specific target effects from general cytotoxicity involving the whole cell. This chapter discusses the nucleus in the context of ultrastructural modifications taking place after the treatment of cells with various antimetabolites. The specificity of lesions induced by substances having similar molecular action explains the normal structural support by which nucleic acids and proteins are formed, stored, and transported from the nucleus to the cytoplasm. The chapter describes a variety of nuclear inclusions following treatment with 4-nitroquinoline-N-oxide and actinomycin D. The inclusions contain RNA and a large number of proteins; they appear to be formed by a rearrangement of the interchromatin RNP network. Cellular intoxication with heavy metals, such as lead, can also produce nuclear inclusions of a different type. More research should be undertaken on the structural arrangement of the nucleus to investigate its complex functional machinery and the apparent lack of organized framework. The use of models, such as drug-induced lesions and combined structure-function studies are likely to provide more information in the near future.

The nucleus: action of chemical and physical agents.

The structure of nuclei and nucleoli of hepatic cells after short-term ethionine administration was investigated with the electron microscope. By 1½ hr after the injection, a distinct alteration occurred in the nucleoli which was characterized by the appearance of electron-opaque masses in the nucleolonema. After 6–8 hr, the nucleoli showed partial fragmentation into small, dense masses. Large aggregates of interchromatinic granules appeared in the nucleoplasm. Condensation of chromatin became prominent in the nucleoplasm particularly along the nuclear membrane. By 12 hr almost complete fragmentation of nucleoli had occurred. The administration of adenine or methionine at 4 hr prevented the development of nucleolar changes. Also, adenine administration at 8 hr after ethionine completely reversed the nucleolar lesion by 12 hr. After methionine administration at 8 hr, many nucleoli showed incomplete reconstruction with many twisted ropelike structures when viewed 4 hr later. Identical structures were found when adenine was given at 8 hr, and animals were sacrificed 2 hr later. On the basis of this observation, the simplified structures of nucleoli found 2 hr after adenine or 4 hr after methionine appeared to be precursors of the nucleolonema. It is suggested that nucleoli show at least two basic reaction patterns to inhibitors of RNA synthesis, one typified by actinomycin D and one by ethionine.

Dense cultures of mouse fibroblast cell line 3T6 were prepared for light microscopic study using either one of two physical substrates, glass slides or a matrix of collagencoated cellulose sponge. Whether propagated on glass or in matrix, cultures receiving a supplement of 250 μg per ml of ascorbic acid formed reticular fibers in less than 10 days. Reticular fibers were more abundant in the matrix cultures than on glass. Fibrous collagen was not found with light or electron microscopy in cultures fed on the control medium, i.e. without the ascorbic acid supplement.
