The origin of plant cell and tissue culture can be found in a treatise published during the mid-18th century, entitled La Physique des Arbes, that describes the formation of callus tissue following the for mation of a ring of cortex from elm trees. Over the next two centuries, the discovery of plant growth hormones, in particular auxins and cytokinins, and detailed analyses on the nutritional requirements of plants, led to the formulation of media that could maintain actively dividing cultures derived from gymnosperms, and both dicotyledon ous and monocotyledonous angiosperms. However, much of the prog ress and technological development in the in vitro propagation of plant cells, tissues, and organs has occurred during the last 25 years. Recently, plant tissue culture techniques have been used as basic tools in the rapidly expanding field of plant biotechnology for the development and clonal propagation of new and/or improved plant varieties. Plant tissue culture is used for the micropropagation of commercially valuable cultivars that include ornamentals, oil palm, Glycyrrhiza, Pyrethrum, pine, Eucalyptus, sugar cane, and potatoes. Cultured plant tissue is also used for the selection of cells and, ul timately, the regeneration of plants that are tolerant to physical stresses such as pathogens, drought, and temperature extremes, and to chemical stress agents such as salinity, herbicides, proteins, and pyrethrins. In addition, new plants have been produced by the fusion of protoplasts prepared from cultured cells of different species in cluding sunflower and french bean, tomato and potato, and various cultivars of Datura. Finally, bacterial vectors and various mechanical methods have been used to introduce foreign genes into cultured plant tissues. Genetic transformation can result in profound changes in the phenotype and/or biochemical profile of the regenerated trans genic plants that are not characteristic of the wild type. An impressive variety of technologies in tissue culture, genetic manipulation, and molecular biology have been developed for nu merous plant species. Many of these techniques, sometimes referred to as plant biotechnology, have been extensively summarized and compiled in a well-balanced collection of easy-to-follow protocols presented in three separate, but complimentary, volumes. Plant Cell, Tissue and Organ Culture consists of 22 chapters (with 86 figures) and 5 appendices. The chapters cover critical aspects of (a) the es sential requirements for the operation of a plant tissue culture lab oratory; (b) the basic procedures of micropropagation, regeneration, and somatic embryogenesis; (c) some specific applications of organ culture systems such as embryo rescue and culture, and anther and microspore culture for haploid and double haploid production; (d) elementary transformation technology; and (e) useful microtechnique and analytical protocols specifically adapted to cultured tissues and cells. The appendices provide a convenient summary of media for mulations and commercial suppliers for the materials described in the text.

Diabetes mellitus is one of the major health problems in the world, the incidence and associated mortality are increasing. Inadequate regulation of the blood sugar imposes serious consequences for health. Conventional antidiabetic drugs are effective, however, also with unavoidable side effects. On the other hand, medicinal plants may act as an alternative source of antidiabetic agents. Examples of medicinal plants with antidiabetic potential are described, with focuses on preclinical and clinical studies. The beneficial potential of each plant matrix is given by the combined and concerted action of their profile of biologically active compounds.

Antidiabetic Potential of Medicinal Plants and Their Active Components.

Natural products (NPs) have been the centre of attention of the scientific community in the last decencies and the interest around them continues to grow incessantly. As a consequence, in the last 20 years, there was a rapid multiplication of various databases and collections as generalistic or thematic resources for NP information. In this review, we establish a complete overview of these resources, and the numbers are overwhelming: over 120 different NP databases and collections were published and re-used since 2000. 98 of them are still somehow accessible and only 50 are open access. The latter include not only databases but also big collections of NPs published as supplementary material in scientific publications and collections that were backed up in the ZINC database for commercially-available compounds. Some databases, even published relatively recently are already not accessible anymore, which leads to a dramatic loss of data on NPs. The data sources are presented in this manuscript, together with the comparison of the content of open ones. With this review, we also compiled the open-access natural compounds in one single dataset a COlleCtion of Open NatUral producTs (COCONUT), which is available on Zenodo and contains structures and sparse annotations for over 400,000 non-redundant NPs, which makes it the biggest open collection of NPs available to this date.

Review on natural products databases: where to find data in 2020

Orchids are nature's most extravagant group of flowering plants distributed throughout the world from tropics to high alpine. They exhibit incredible range of diversity in size, shape and color of their flowers. Though orchids are grown primarily as ornamentals, many are used as herbal medicines, food, and other have cultural value by different cultures and tribes in different parts of the world. Orchids have been used in many parts of the world in traditional healing system as well as in the treatment of a number of diseases since the ancient time. Though Orchidaceae is regarded as a largest family of plant kingdom, few studies have been done regarding their medicinal properties. Linking of the indigenous knowledge of medicinal orchids to modern research activities provides a new reliable approach, for the discovery of novel drugs much more effectively than with random collection. Many of these orchids face the extreme danger of extinction due to over-exploitation and habitat loss. Plant tissue culture could be one of the most suitable alternative tools to minimize the pressure on natural population of medicinal orchids and their sustainable utilization.

Medicinal orchids and their uses: Tissue culture a potential alternative for conservation

Magnolia sirindhorniae Noot. & Chalermglin is an endangered species with high ornamental and commercial value that needs to be urgently protected and judiciously commercialized. In this study, a protocol for efficient regeneration of this species is standardized. The lateral buds of the M. sirindhorniae plant were used as an explant. Half-strength Murashige and Skoog (MS) medium supplemented with 2.0 mg/L 6-benzyladenine (BA), 0.1 mg/L α-naphthaleneacetic acid (NAA), and 2.0 mg/L gibberellic acid (GA3) was found to be the optimal medium for shoot induction. The maximum shoot multiplication rate (310%) was obtained on Douglas-fir cotyledon revised medium (DCR) fortified with 0.2 mg/L BA, 0.01 mg/L NAA, and additives. The half-strength DCR medium supplemented with 0.5 mg/L NAA and 0.5 mg/L indole-3-butyric acid (IBA) supported the maximum rate (85.0%) of in vitro root induction. After a simple acclimatization process, the survival rate of plantlets in a substrate mixture of sterile perlite and peat soil (1:3; v/v) was 90.2%. DNA markers were used for assessment of genetic uniformity, confirming the genetic uniformity and stability of regenerated plants of M. sirindhorniae. Thus, the described protocol can safely be applied for large scale propagation of this imperative plant.

Bamboo shoots are considered as one of the useful health foods because of their rich contents of proteins, carbohydrates, vitamins, fibres, and minerals and very low fat. Though bamboo shoots provide lots of health benefits, their consumption is confined mostly to Southeast Asian and East Asian countries. The acceptability of bamboo shoots as popular vegetable crop is very less due to their high pungent smell and bitter acidic taste. The use of bamboo as food in India is mainly restricted to Northeastern part of the country where they form an indispensable part of several traditional speciality dishes. The different ethnic communities take fresh or fermented bamboo shoot as one of most preferred traditional food items. Some of the important bamboo based traditional foods are ushoi, soibum, rep, mesu, eup, ekhung, hirring, and so forth. Bamboo shoots should be properly processed before they are consumed as freshly harvested shoots have high content of toxic cyanogenic glycosides which may pose serious health problems. The prospect of bamboo shoot industry in Northeast India is bright due to its rich genetic resources of bamboos. However, habitat destruction and extensive use of bamboos for food, handicraft, and construction purposes have resulted in severe depletion of natural bamboo resources. This review stresses upon the high nutritive values and health benefits of bamboo shoots and their usage as important traditional foods in Northeast India. The bamboo market potential of the region and use of in vitro plant micropropagation methods as effective means of bamboo conservation are also emphasized in this paper.

The Nutritional Facts of Bamboo Shoots and Their Usage as Important Traditional Foods of Northeast India

In vitro rapid propagation of Cymbidium aloifolium (L.) Sw.: a medicinally important orchid via seed culture.

Melocanna baccifera (Roxb.) Kurz is an economically important bamboo of North-East India experiencing population depletion in its natural habitats. Genetic variation studies were conducted in 7 populations sampled from 5 districts of Manipur using ISSR molecular markers. The investigation was carried out as a primary step towards developing effective conservation strategies for the protection of bamboo germplasm. ISSR marker analysis showed significant level of genetic variation within the populations as revealed by moderately high average values of Nei's genetic diversity (H 0.1639), Shannon's diversity index (I 0.2563), percentage of polymorphic bands (PPB 59.18), total genetic variation (Ht 0.1961), and genetic diversity within population (Hs 0.1639). The study also divulged a high genetic variation at species level with Shannon's diversity index (I), Nei's genetic diversity (H), and percentage of polymorphic band (PPB%) recorded at 0.3218, 0.1939, and 88.37, respectively. Genetic differentiation among the populations (Gst) was merely 19.42% leaving 80.58% of genetic variation exhibited within the populations. The low genetic diversity between populations was consistent with AMOVA. The low genetic differentiation among populations coupled with existence of significantly high genetic diversity at species level indicated the urgent necessity of preserving and protecting all the existing natural bamboo populations in the region.
