The role of the lateral dimension of graphene oxide in the regulation of cellular responses

The different behaviours of dissolved silver and silver nanoparticles under ICP-MS single particle detection conditions have been used to differentiate directly between both forms of silver in aqueous samples. Suspensions containing silver nanoparticles at number concentrations below 109 L−1 and/or dissolved Ag(I) are introduced into the ICP-MS by conventional pneumatic nebulization and measured with a time resolution of 5 ms. Each silver nanoparticle is converted in the ICP into a packet of ions, which are detected as a single pulse, whose intensity is proportional to the number of silver atoms in the nanoparticle, whereas dissolved silver produces pulses of averaged constant intensity. The frequency plots with respect to the intensity measured for each pulse show independent distributions for dissolved silver and silver nanoparticles, whose profiles are also different (Poisson and lognormal, respectively). Size limits of detection for pure Ag nanoparticles of 18 nm, equivalent to a silver mass of 32 ag, were obtained. Number concentration limits of detection of 1 × 104 L−1 can be achieved. A methodological approach for identification, characterization and determination of mass and number concentration of dissolved Ag(I) and silver nanoparticles at environmentally relevant concentrations is presented.

Selective identification, characterization and determination of dissolved silver(I) and silver nanoparticles based on single particle detection by inductively coupled plasma mass spectrometry

Accurate in vitro assessment of nanoparticle cytotoxicity requires a careful selection of the test systems. Due to high adsorption capacity and optical activity, engineered nanoparticles are highly potential in influencing classical cytotoxicity assays. Here, four common in vitro assays for oxidative stress, cell viability, cell death and inflammatory cytokine production (DCF, MTT, LDH and IL-8 ELISA) were assessed for validity using 24 well-characterized engineered nanoparticles. For all nanoparticles, the possible interference with the optical detection methods, the ability to convert the substrates, the influence on enzymatic activity and the potential to bind proinflammatory cytokines were analyzed in detail. Results varied considerably depending on the assay system used. All nanoparticles tested were found to interfere with the optical measurement at concentrations of 50 μg cm−2 and above when DCF, MTT and LDH assays were performed. Except for Carbon Black, particle interference could be prevented by altering assay protocols and lowering particle concentrations to 10 μg cm−2. Carbon Black was also found to oxidize H2DCF-DA in a cell-free system, whereas only ZnO nanoparticles significantly decreased LDH activity. A dramatic loss of immunoreactive IL-8 was observed for only one of the three TiO2 particle types tested. Our results demonstrate that engineered nanoparticles interfere with classic cytotoxicity assays in a highly concentration-, particle- and assay-specific manner. These findings strongly suggest that each in vitro test system has to be evaluated for each single nanoparticle type to accurately assess the nanoparticle toxicity.

Interference of engineered nanoparticles with in vitro toxicity assays.

The discovery of fullerenes in 1985 has ushered in an explosive growth in the applications of engineered nanomaterials and consumer products. Nanotechnology and engineered nanomaterials (ENMs) are being incorporated into a range of commercial products such as consumer electronics, cosmetics, imaging and sensors. Nanomaterials offer new possibilities for the development of novel sensing and monitoring technologies. Nanosensors can be classified under two main categories: (i) Nanotechnology-enabled sensors or sensors that are themselves nanoscale or have nanoscale materials or components, and (ii) Nanoproperty-quantifiable sensors or sensors that are used to measure nanoscale properties. The first category can eventually result in lower material cost, reduced weight and energy consumption. The second category can enhance our understanding of the potential toxic effects of emerging pollutants from nanomaterials including fullerenes, dendrimers, and carbon nanotubes. Despite the enormous literatures and reviews on Category I sensors, there are few sensors to measure nanoscale properties or sensors belonging to Category II. This class of nanosensors is an area of critical interest to nanotoxicology, detection and risk assessment, as well as for monitoring of environmental and/or biological exposure. This article discusses emerging fields of nanotoxicology and nanomonitoring including the challenges of characterizing engineered nanomaterials and the potentials of combining existing analytical techniques with conventional cytotoxicity methods. Two case studies are provided for development of Category II nanosensors for fullerene nanoparticles and quantum dots. One highlights the uniqueness of a portable, dissolved oxygen electrochemical sensor arrays capable of detecting the ENMs as well as provide rapid nanotoxicological information. This review has shown that addressing the complex and critical issues surrounding the environmental transformation and toxicity of ENMs must be accompanied by the creation of new approaches or further developments of existing instrumentation.

Sensors as tools for quantitation, nanotoxicity and nanomonitoring assessment of engineered nanomaterials

A portable biosensor system includes at least one capillary tube extending longitudinally along a major axis between a proximal inlet end and a distal end. The at least one capillary tube has an interior surface coated with a capture material and forms a waveguide. At least one collimated light emitting diode is disposed proximate and perpendicular to the major axis of the at least one capillary tube and is positioned relative to the at least one capillary tube so that energy enters the at least one capillary tube from its exterior along the entire length of the at least one capillary tube to project a line of energy along substantially the entire longitudinal extent of the at least one capillary tube. A photosensor is disposed proximate the distal end of the at least one capillary tube for receiving emissive radiation therefrom. The photosensor generates an output voltage (or, optical output) representative of the emissive radiation, and a means for measuring the output voltage is provided. Also disclosed is a method of detecting target molecules in a sample using the biosensor of the present invention.

Capillary biosensor system and its method of use

This paper presents the first development of a mass-sensitive nanosensor for the isolation and quantitative analyses of engineered fullerene (C₆₀) nanoparticles, while excluding mixtures of structurally similar fullerenes. Amino-modified beta-cyclodextrin (β-CD-NH₂) was synthesized and confirmed by ¹HNMR as the host molecule to isolate the desired fullerene C₆₀. This was subsequently assembled onto the surfaces of gold-coated quartz crystal microbalance (QCM) electrodes using N-dicyclohexylcarbodiimide/N-hydroxysuccinimide (DCC/NHS) surface immobilization chemistry to create a selective molecular configuration described as (Au)-S-(CH₂)²-CONH-beta-CD sensor. The mass change on the sensor configuration on the QCM was monitored for selective quantitative analysis of fullerene C₆₀ from a C₆₀/C₇₀ mixture and soil samples. About ~10¹⁴-10¹⁶ C₆₀ particles/cm² were successfully quantified by QCM measurements. Continuous spike of 200 μL of 0.14 mg C₆₀ /mL produced changes in frequency (-Δf) that varied exponentially with concentration. FESEM and time-of-flight secondary-ion mass spectrometry confirmed the validity of sensor surface chemistry before and after exposure to fullerene C₆₀. The utility of this sensor for spiked real-world soil samples has been demonstrated. Comparable sensitivity was obtained using both the soil and purified toluene samples. This work demonstrates that the sensor has potential application in complex environmental matrices.
