Host fecundity reduction: a strategy for damage limitation?

The transmission success of free-living larval stages of endohelminths is generally modulated by a variety of abiotic and biotic environmental factors. Whereas the role of abiotic factors (including anthropogenic pollutants) has been in focus in numerous studies and summarized in reviews, the role of biotic factors has received much less attention. Here, we review the existing body of literature from the fields of parasitology and ecology and recognize 6 different types of biotic factors with the potential to alter larval transmission processes. We found that experimental studies generally indicate strong effects of biotic factors, and the latter emerge as potentially important, underestimated determinants in the transmission ecology of free-living endohelminth stages. This implies that biodiversity, in general, should have significant effects on parasite transmission and population dynamics. These effects are likely to interact with natural abiotic factors and anthropogenic pollutants. Investigating the interplay of abiotic and biotic factors will not only be crucial for a thorough understanding of parasite transmission processes, but will also be a prerequisite to anticipate the effects of climate and other global changes on helminth parasites and their host communities.

The role of biotic factors in the transmission of free-living endohelminth stages.

Anoplocephala perfoliata is the commonest tapeworm parasite of horses and is incriminated as a significant cause of clinical disease (e.g., ileocaecal intussusception, caeco-caecal intussusception and/or caecal perforation), particularly in horses chronically infected with large numbers of worms. The high prevalence (approximately 20-80%) of the parasite in some countries suggests an increased risk of clinical cases. In spite of research, there is still a paucity of information regarding the pathogenesis of the disease, the epidemiology of the parasite in different geographical regions and there are significant limitations with the diagnosis of infection. The present article provides an account of the biology, epidemiology and pathogenic effects of A. perfoliata, the diagnosis of infection and treatment. It highlights some gaps in knowledge of the parasite and the disease it causes, and suggests opportunities for future research and prospects for improved diagnosis, prevention and control.

The mean parasite burden of a population of Tribolium confusum is shown to rise to a plateau as the exposure density of infective eggs of Hymenolepis diminuta increases. The level of this plateau is shown to be dependent on the nutritional status of the host population, being depressed from approximately 18 cysticeroids/beetle in hosts which have been starved prior to experimentation, to approximately 2 cysticercoids/beetle in satiated hosts. A simple model is used to describe the shape of this infection functional response in terms of the predator-prey interaction between hosts (T. confusum) and parasite infective stages (H. diminuta eggs). The distribution of successful infections/host is shown to be over-dispersed, even when hosts are exposed to infective stages arranged in a uniform spatial pattern. The over-dispersion of parasite numbers/host is shown to become more severe as the spatial pattern of infective stages changes from under-dispersed, through random, to over-dispersed. Experimental results are discussed in relation to the dynamics of parasite-host interactions, in which infection takes place by host ingestion of a free-living infective stage.

The dynamics of infection of Tribolium confusum by Hymenolepis diminuta: the influence of infective-stage density and spatial distribution.

The hatching of Hymenolepis diminuta eggs and penetration of the emergent hexacanths in adult and larval Tenebrio molitor beetles had been described. Hatching was shown to be a two-stage process involving rupture of the eggshell by the insect's mandibles and digestion of the remaining egg-layers in the midgut lumen. Penetration of hatched embryos was examined by histological methods that demonstrated the rate of migration and the route taken by hexacanths entering the haemocoel of adult beetles. Differences in the arrangement of the midgut tissues in adult and larval T. molitor were detected and shown to account for the failure of hexacanths to complete migration in the larvae. The role of the embryonic hooks and penetration gland secretions during hatching and penetration has also been discussed.I should like to thank Professor T. R. E. The hatching of Hymenolepis diminuta eggs and penetration of the hexacanths in Tenebrio molitor beetles

1. It has been shown that the protein eggshell of Hymenolepis diminuta eggs was not significantly stabilized by sulphur or electrovalent linkages but was resistant to attack by proteolytic enzymes. Amino acid analysis demonstrated a marked preponderance of aromatic and heterocyclic acids and it has been suggested that these groups act as cross-linking agents in the shell protein. 2. The role of the lipid-containing subshell ‘membrane’ in protecting the enclosed embryo against fluctuations in pH and osmolarity in the external environment and infiltration of enzymes and toxic compounds has been demonstrated. 3. The cytoplasmic layer was shown to consist of a glucosamine-containing mucoprotein present in a dehydrated or semi-dehydrated state in the intact egg.In mechanically damaged eggs the cytoplasmic layer was rapidly dispersed by some proteolytic enzymes. 4. The embryophore enclosing the hexacanth embryo was composed of a sulphurrich protein that was weakened by some proteolytic enzymes and completely destroyed by others. The chemical composition and some properties of the egg layers in Hymenolepis diminuta eggs

The results of several experiments that included variation of the pH and concentration of the extracts and analysis of the role of enzymes separated from midgut fluid by electrophoresis indicated that hatching involved two or more proteolytic enzymes that digested the cytoplasmic layer and embryophore. Amylases from T. molitor and other sources were shown to play no part in hatching.
 A defined medium based on analyses of midgut fluid and containing trypsin, chymotrypsin and peptidases was shown to digest the embryophore and effect the release of hexacanths in a time comparable to that obtained with T. molitor extracts. The specificity of the enzymes attacking the cytoplasmic layer remains unknown.
