Preparation of isolated rat liver cells.

A new technique employing continuous recirculating perfusion of the rat liver in situ, shaking of the liver in buffer in vitro, and filtration of the tissue through nylon mesh, results in the conversion of about 50% of the liver into intact, isolated parenchymal cells. The perfusion media consist of: (a) calcium-free Hanks' solution containing 0.05% collagenase and 0.10% hyaluronidase, and (b) magnesium and calcium-free Hanks' solution containing 2 mM ethylenediaminetetraacetate. Biochemical and morphologic studies indicate that the isolated cells are viable. They respire in a medium containing calcium ions, synthesize glucose from lactate, are impermeable to inulin, do not stain with trypan blue, and retain their structural integrity. Electron microscopy of biopsies taken during and after perfusion reveals that desmosomes are quickly cleaved. Hemidesmosome-containing areas of the cell membrane invaginate and appear to pinch off and migrate centrally. Tight and gap junctions, however, persist on the intact, isolated cells, retaining small segments of cytoplasm from formerly apposing parenchymal cells. Cells which do not retain tight and gap junctions display swelling of Golgi vacuoles and vacuoles in the peripheral cytoplasm. Cytoplasmic vacuolization in a small percentage of cells and potassium loss are the only indications of cell injury detected. By other parameters measured, the isolated cells are comparable to normal hepatic parenchymal cells in situ in appearance and function.

HIGH-YIELD PREPARATION OF ISOLATED RAT LIVER PARENCHYMAL CELLS: A Biochemical and Fine Structural Study

50 years ago Peter Mitchell proposed the chemiosmotic hypothesis for which he was awarded the Nobel Prize for Chemistry in 1978. His comprehensive review on chemiosmotic coupling known as the first "Grey Book", has been reprinted here with permission, to offer an electronic record and easy access to this important contribution to the biochemical literature. This remarkable account of Peter Mitchell's ideas originally published in 1966 is a landmark and must-read publication for any scientist in the field of bioenergetics. As far as was possible, the wording and format of the original publication have been retained. Some changes were required for consistency with BBA formats though these do not affect scientific meaning. A simple device is described for the rapid centrifugal separation of isolated hepatocytes or similar tissue fragments from the suspending medium. Rapid separation is essential if meaningful information on the concentrations of cell constituents and their distribution between cell and suspension medium is to be obtained. The usefulness of the technique is illustrated by measurements of hepatocyte constituents (glutamate, aspartate, alanine, K+) which show large concentration gradients against the extracellular medium.

Tissue lipogenesis is variably controlled by substrate supply and hormones. The possibility that nitric oxide (NO) might regulate lipogenesis derives from the action of NO on coenzyme A (CoA) to produce metabolically inactive S-nitrosoCoA. The effect of the nitric oxide donor S-nitrosoglutathione (GSNO) on long chain fatty acid and cholesterol synthesis was measured in isolated cultured rat hepatocytes. [1-14C] Butyrate was used as substrate to measure 14C incorporation into lipids as butyrate is twice as effective as acetate in hepatic lipogenesis and is ketogenic via the Lynen cycle. NO very significantly (P < 0.01) impaired long chain fatty acid and cholesterol synthesis an observation dependent upon time of exposure (3 h pre-incubation or 6 h continuous exposure) and concentration of GSNO (500 μM to 2.0 mM). Decrease in hepatic lipogenesis was paralleled by decrease in ketogenesis. ATP levels remained unchanged following short term exposure to GSNO. Exposure of hepatocytes to GSNO together with 2.0 mM glutathione significantly diminished the inhibition of lipogenesis induced by GSNO alone. Impairment of lipogenesis by GSNO appears not to be limited by energy supply and now adduced, but not proven, to be operative via the degree of inactivation of cytosolic CoA. NO control of lipogenesis could be clinically important where NO production is increased as in demyelinating diseases, chronic arthritis or colitis and in wasting diseases such as AIDS. Inhibition of hepatocyte lipogenesis by nitric oxide donor: could nitric oxide regulate lipid synthesis?

1. Kidney-cortex slices and the perfused livers of vitamin B12-deficient rats removed propionate from the incubation and perfusion media at 33 and 17% respectively of the rates found with tissues from rats receiving either a normal or a vitamin B12-supplemented diet. There was a corresponding fall in the rates of glucose synthesis from propionate in both tissues. 2. The addition of hydroxocobalamin or dimethylbenzimidazolylcobamide coenzyme to kidney-cortex slices from vitamin B12-deficient rats in vitro failed to restore the normal capacity for propionate metabolism. 3. Although the vitamin B12-deficient rat excretes measurable amounts of methylmalonate, no methylmalonate production could be detected (probably because of the low sensitivity of the method) when kidney-cortex slices or livers from deficient rats were incubated or perfused with propionate. 4. The addition of methylmalonate (5mm) to kidney-cortex slices from rats fed on a normal diet inhibited gluconeogenesis from propionate by 25%. 5. Methylmalonate formation is normally only a small fraction of the flux through methylmalonyl-CoA. This fraction increases in vitamin B12-deficient tissues (as shown by the urinary excretion of methylmalonate) presumably because the concentration of methylmalonyl-CoA rises as a result of low activity of methylmalonyl-CoA mutase (EC 5.4.99.2). Slow removal of methylmalonyl-CoA might depress propionate uptake owing to the reversibility of the steps leading to methylmalonyl-CoA formation.

Enthalpies of hydrolysis of glutamine and asparagine and of ionization of glutamic and aspartic acids

Hepatocytes were prepared from rats fed a chow diet (control-fed) and from rats fed a similar diet in which the drinking water contained 10% (wt/vol) fructose (fructose-fed). Both types of hepatocyte preparations were cultured for < or = 48 hours in supplemented Waymouth's medium containing increasing concentrations of bovine insulin (0 to 780 nmol/L). During the first 24 hours of culture, hepatocytes from fructose-fed rats secreted more very-low-density lipoprotein (VLDL) triacylglycerol (TAG) than hepatocytes from control-fed rats. This difference persisted at all concentrations of insulin. There was no difference in the rate of secretion of apolipoprotein B (apo B). In both control-fed and fructose-fed animals, the inhibitory effect of insulin on the secretion of VLDL was greater on the second versus the first day of culture. Under these conditions, hepatocytes from fructose-fed groups were less sensitive to insulin inhibition as compared with those from the control-fed group. This was evidenced by the following: (1) the decreased inhibitory effect of insulin on the secretion of both total and newly synthesized VLDL TAG, (2) the attenuated inhibitory effect of insulin on the secretion of VLDL apo B, (3) the decreased potency of insulin in suppressing the secretion of VLDL TAG in TAG-depleted hepatocytes from fructose-fed as compared with control-fed animals, and (4) the larger proportion of newly synthesized TAG secreted as VLDL in hepatocytes from fructose-fed rats as compared with controls. This difference was exacerbated at higher concentrations of insulin.(ABSTRACT TRUNCATED AT 250 WORDS)
