Nanochemistry and Nanomedicine for Nanoparticle-based Diagnostics and Therapy

The design and use of materials in the nanoscale size range for addressing medical and health-related issues continues to receive increasing interest. Research in nanomedicine spans a multitude of areas, including drug delivery, vaccine development, antibacterial, diagnosis and imaging tools, wearable devices, implants, high-throughput screening platforms, etc. using biological, nonbiological, biomimetic, or hybrid materials. Many of these developments are starting to be translated into viable clinical products. Here, we provide an overview of recent developments in nanomedicine and highlight the current challenges and upcoming opportunities for the field and translation to the clinic.

Gold Nanomaterials at Work in Biomedicine

Nowadays, magnetic hyperthermia constitutes a complementary approach to cancer treatment. The use of magnetic particles as heating mediators, proposed in the 1950s, provides a novel strategy for improving tumor treatment and, consequently, patient's quality of life. This review reports a broad overview about several aspects of magnetic hyperthermia addressing new perspectives and the progress on relevant features such as the ad hoc preparation of magnetic nanoparticles, physical modeling of magnetic heating, methods to determine the heat dissipation power of magnetic colloids including the development of experimental apparatus and the influence of biological matrices on the heating efficiency.

Fundamentals and advances in magnetic hyperthermia

We present a critical review of the state of the art of magnetic particle hyperthermia (MPH) as a minimal invasive tumour therapy. Magnetic principles of heating mechanisms are discussed with respect to the optimum choice of nanoparticle properties. In particular, the relation between superparamagnetic and ferrimagnetic single domain nanoparticles is clarified in order to choose the appropriate particle size distribution and the role of particle mobility for the relaxation path is discussed. Knowledge of the effect of particle properties for achieving high specific heating power provides necessary guidelines for development of nanoparticles tailored for tumour therapy. Nanoscale heat transfer processes are discussed with respect to the achievable temperature increase in cancer cells. The need to realize a well-controlled temperature distribution in tumour tissue represents the most serious problem of MPH, at present. Visionary concepts of particle administration, in particular by means of antibody targeting, are far from clinical practice, yet. On the basis of current knowledge of treating cancer by thermal damaging, this article elucidates possibilities, prospects, and challenges for establishment of MPH as a standard medical procedure.

Magnetic particle hyperthermia—a promising tumour therapy?

Magnetic nanoparticles are interesting materials for a lot of medical and technical applications. A less experimentally investigated question is the influence of particle packing density on magnetic properties due to magnetic interactions between single particles. For this, magnetic nanoparticles of iron oxides prepared as fine dry powder by laser deposition are investigated with respect to their structural and magnetic properties as function of packing density. The particles are nearly spherically shaped single crystals in the magnetic single domain size range with a mean diameter of 21 nm occasionally exhibiting spinel growth facets. Powders of these particles are diluted by nonmagnetic silicon oxide particles in a range of volume concentrations from 0.2 % up to 68 % of the bulk density of magnetite. The concentration dependence of remanence, coercivity and hysteresis losses is determined by measurements of minor loops in a vibrating sample magnetometer. Results which are discussed in the frame of present theoretical models may be understood in terms of the cubic anisotropy of magnetite distorted by a small uniaxial shape contribution.
