First introduced in the 1980s, laser speckle contrast imaging is a powerful tool for full-field imaging of blood flow. Recently laser speckle contrast imaging has gained increased attention, in part due to its rapid adoption for blood flow studies in the brain. We review the underlying physics of speckle contrast imaging and discuss recent developments to improve the quantitative accuracy of blood flow measures. We also review applications of laser speckle contrast imaging in neuroscience, dermatology and ophthalmology.

Laser speckle contrast imaging in biomedical optics

X-ray sources are developing rapidly and their coherent output is growing correspondingly. The increased coherent flux from modern X-ray sources is being matched with an associated development in experimental methods. This article reviews the literature describing the ideas that utilize the increased brilliance from modern X-ray sources. It explores how ideas in coherent X-ray science are leading to developments in other areas, and vice versa. The article describes measurements of coherence properties and uses this discussion as a base from which to describe partially coherent diffraction and X-ray phase-contrast imaging, with applications in materials science, engineering and medicine. Coherent diffraction imaging methods are reviewed along with associated experiments in materials science. Proposals for experiments to be performed with the new X-ray free-electron lasers are briefly discussed. The literature on X-ray photon-correlation spectroscopy is described and the features it has in common with other coherent X-ray methods are identified. Many of the ideas used in the coherent X-ray literature have their origins in the optical and electron communities and these connections are explored. A review of the areas in which ideas from coherent X-ray methods are contributing to methods for the neutron, electron and optical communities is presented.

Coherent methods in the X-ray sciences

When laser light illuminates a diffuse object, it produces a random interference effect known as a speckle pattern. If there is movement in the object, the speckles fluctuate in intensity. These fluctuations can provide infor- mation about the movement. A simple way of accessing this information is to image the speckle pattern with an exposure time longer than the shortest speckle fluctuation time scale—the fluctuations cause a blurring of the speckle, leading to a reduction in the local speckle contrast. Thus, velocity distributions are coded as speckle con- trast variations. The same information can be obtained by using the Doppler effect, but producing a two-dimen- sional Doppler map requires either scanning of the laser beam or imaging with a high-speed camera: laser speckle contrast imaging (LSCI) avoids the need to scan and can be performed with a normal CCD- or CMOS-camera. LSCI is used primarily to map flow systems, especially blood flow. The development of LSCI is reviewed and its lim- itations and problems are investigated. © The Authors. When a diffuse object is illuminated with coherent laser light, the backscattered light will form an interference pattern on the detector. This pattern of bright and dark areas is called a speckle pattern. When there is movement in the object, the speckle pattern will change over time. Laser speckle contrast techniques use this change in speckle pattern to visualize tissue perfusion. We present and review the contribution of laser speckle contrast techniques to the field of perfusion visualization and discuss the development of the techniques.

Review of laser speckle contrast techniques for visualizing tissue perfusion.

Laser Speckle Contrast Imaging (LSCI) is a minimally invasive full field optical technique used to generate blood flow maps with high spatial and temporal resolution. The lack of quantitative accuracy and the inability to predict flows in the presence of static scatterers such as an intact or thinned skull have been the primary limitation of LSCI. We present a new Multi-Exposure Speckle Imaging (MESI) instrument that has potential to obtain quantitative baseline flow measures. We show that the MESI instrument extends the range over which relative flow measurements are linear. We also present a new speckle model which can discriminate flows in the presence of static scatters. We show that in the presence of static scatterers the new model used along with the new MESI instrument can predict correlation times of flow consistently to within 10% of the value without static scatterers compared to an average deviation of more than 100% from the value without static scatterers using traditional LSCI. We also show that the new speckle model used with the MESI instrument can maintain the linearity of relative flow measurements in the presence of static scatterers.

Robust flow measurement with multi-exposure speckle imaging.

We describe a multispeckle dynamic light scattering technique capable of resolving the motion of scattering sites in cases that this motion changes systematically with time. The method is based on the visibility of the speckle pattern formed by the scattered light as detected by a single exposure of a digital camera. Whereas previous multispeckle methods rely on correlations between images, here the connection with scattering site dynamics is made more simply in terms of the variance of intensity among the pixels of the camera for the specified exposure duration. The essence is that the speckle pattern is more visible, i.e., the variance of detected intensity levels is greater, when the dynamics of the scattering site motion is slow compared to the exposure time of the camera. The theory for analyzing the moments of the spatial intensity distribution in terms of the electric-field autocorrelation is presented. It is tested for two well-understood samples, a colloidal suspension of Brownian particles and a coarsening foam, where the dynamics can be treated as stationary and hence can be benchmarked by traditional methods. However, our speckle-visibility method is particularly appropriate for samples in which the dynamics vary with time, either slowly or rapidly, limited only by the exposure time fidelity of the camera. Potential applications range from soft-glassy materials, to granular avalanches, to flowmetry of living tissue.

Speckle-visibility spectroscopy: A tool to study time-varying dynamics

Bio Xu Mao has been an associate professor in the College of Engineering at China Agricultural University since 2021. He received his PhD from the Mechanical Engineering Department at Texas A&M University, then joined the Mechanical and Industrial Engineering Department at Northeastern University, where he was a postdoctoral fellow in 2019. His research interests focus on ultrafast laser dynamics, intelligent measurement and control, and multimodal sensing, imaging and fusion, with applications to advanced manufacturing, agricultural products, and smart agricultural devices. C Steve Suh is faculty member in the Mechanical Engineering Department at Texas A&M University, and an ASME Fellow. His interests in nonlinear timefrequency control, advanced manufacturing, engineering design theory, ultrafast laser dynamics, and complex networks have resulted in exceeding 180 scientific publications, ten book volumes and book chapters.
