Prevalence and identification of tick species (Ixodidae) on domestic animals in district Multan, Punjab Pakistan

Ticks and tick-borne diseases (TTBDis) are a major constraint to the health and production of small ruminants in Pakistan. Despite being the subject of intermittent studies over the past few decades, comprehensive information on the epidemiology and control of TTBDis is lacking. Herein, we have systematically reviewed the current knowledge on TTBDis of small ruminants in Pakistan. Critical appraisal of the selected 71 articles published between 1947 to 2020 revealed that morphological examination had been the most widely used method for the identification of TTBDis in Pakistan. Tick fauna comprise at least 40 species, mainly belonging to Haemaphysalis, Hyalomma and Rhipicephalus. The prevalence of ticks is the highest in summer (June-September) and it is also higher in goats than sheep. Anaplasma, Babesia and Theileria spp. are the major tick-borne pathogens (TBPs), and their prevalence is usually higher in sheep than goats. Spatio-temporal distribution, genetic diversity and control of ticks and TBPs of small ruminants as well as the competence of tick vectors for various TBPs remain to be explored. Therefore, coordinated and focused investigations are required to fill knowledge gaps in these areas to maximise the health, production and welfare of small ruminants and minimise economic losses associated with TTBDis in Pakistan.

Systematic Review of Ticks and Tick-Borne Pathogens of Small Ruminants in Pakistan

Theileria ovis and Anaplasma marginale are intracellular pathogens affecting a wide range of animals, causing huge economic losses worldwide. The present study reports the molecular evidence of Theileria ovis and Anaplasma marginale in sheep blood samples (N = 218) collected from Layyah district in Punjab (Pakistan), where economy heavily relies on livestock. A 520 base pair fragment specific for 18S ribosomal RNA gene of Theileria ovis was PCR amplified in 23/218 (10.6%) sheep blood samples, while for Anaplasma marginale, a 265 base pair fragment specific for msp1b gene was generated in 15/218 (6.9%) sheep blood samples. Two blood samples were found co-infected (0.9%) with both parasites. Amplified PCR products of both parasites were confirmed by DNA sequencing and submitted to GenBank. Prevalence of both Theileria ovis (p = 0.3) and Anaplasma marginale (p = 0.4) varied non-significantly among the investigated sheep breeds. Tick burden on dogs present with sheep herds was found associated with Theileria ovis infection in sheep (p = 0.05). It was observed that lambs (p = 0.009), sheep in small herds (p = 0.04), and tick burden on dogs present with sheep herds (p = 0.01) were associated with Anaplasma marginale infection in sheep during the present study. In conclusion, we are reporting a higher prevalence of Theileria ovis than Anaplasma marginale in blood samples of sheep collected from Layyah district. Tick-infested dogs were found to be risk factors for the transmission of both pathogens in sheep, and tick control strategies should be extended to dogs associated with sheep herds in this area.

Molecular detection and prevalence of Theileria ovis and Anaplasma marginale in sheep blood samples collected from Layyah district in Punjab, Pakistan.

Background: Ticks are ectoparasites that transmit a variety of pathogens that cause many diseases in livestock which can result in skin damage, weight loss, anemia, reduced production of meat and milk, and mortality. Aim: The aim of this study was to identify tick species and the distribution on livestock hosts (sheep, goat, dairy cattle, and buffalo) of Punjab, Khyber Pakhtunkhwa Province and Islamabad from October 2019 to November 2020. Materials and Methods: Surveillance was performed to calculate the prevalence of ticks on livestock. Tick prevalence data (area, host, breed, gender, age, and seasonal infestation rate) was recorded and analyzed. Results: A total of 2080 animals were examined from selected farms, and, of these, 1129 animals were tick-infested. A total of 1010 male tick samples were identified to species using published keys. Haemaphysalis punctata, Haemaphysalis sulcata, Hyalomma anatolicum, Hyalomma detritum, Hyalomma dromedarii, Hyalomma excavatum, Hyalomma marginatum, Hyalomma rufipes, Rhipicephalus decoloratus Rhipicephalus microplus, and Rhipicephalus sanguineus were collected from goats, sheep, buffalo, and cattle. The overall rates of tick infestation on livestock were 34.83% (buffalo), 57.11% (cattle), 51.97% (sheep) and 46.94% (goats). Within each species, different breeds demonstrated different proportions of infestation. For cattle breeds, infestation proportions were as follows: Dhanni (98.73%), Jersey (70.84%) and the Australian breed of cattle (81.81%). The Neeli Ravi breed (40%) of buffalo and the Beetal breed (57.35%) of goats were the most highly infested for these species. Seasonally, the highest prevalence of infestation (76.78%) was observed in summer followed by 70.76% in spring, 45.29% in autumn, and 20% in winter. The prevalence of tick infestation in animals also varied by animal age. In goats, animals aged 4–6 years showed the highest prevalence (90%), but in cattle, the prevalence of ticks was highest (68.75%) in 6 months–1-year-old animals. 1–3 years old buffalo (41.07%) and 6 months–1 year sheep (65.78%) had the highest prevalence rate. Females had significantly higher infestation rates (61.12%, 55.56% and 49.26%, respectively) in cattle, sheep, and goats. In buffalo, males showed a higher prevalence (38.46%) rate. Conclusions: This study showed tick diversity, infestation rate, and numerous factors (season, age, and gender of host) influencing tick infestation rate in different breeds of cattle, sheep, goats, and buffalo in Punjab Province, Khyber Pakhtunkhwa Province, and Islamabad, Pakistan. Higher tick burdens and rates of tick-borne disease reduce production and productivity in animals. Understanding tick species’ prevalence and distribution will help to develop informed control measures.

Epidemiology, Distribution and Identification of Ticks on Livestock in Pakistan

Tropical theileriosis, caused by the apicomplexan hemoparasite of the genus Theileria, is a major constraint to livestock production in various parts of world, including South Asia. Several studies have been carried out over the last five decades; however, comprehensive information in this region regarding the diversity and distribution of Theileria is lacking. Therefore, keeping in mind the economic importance of theileriosis, we have systematically reviewed the current knowledge about Theileria spp. diversity and distribution affecting cattle, water buffaloes, goats and sheep in three countries included India, Pakistan and Bangladesh of the Indian sub-continent. The data collected indicated that the microscopic method is the widely used method for evaluating Theileria species in the three countries from 1970 to 2021. This is the first study in this region to compile a comprehensive knowledge about the diversity and distribution of Theileria. Our study revealed the existence of 11 different species of Theileria, including Theileria spp. Theleria annulata, T. orientalis, T. mutans, T. velifera circulating in cattle and buffalo while T. annulata, T. lestoquardi, T. luwenshuniT. ovis, Theileria spp. and T. lestoquardi-like spp., were infecting goats and sheep from various regions of India, Pakistan and Bangladesh. We find that T. annulata can be found in both small and large ruminants and is widely distributed in the different regions of India, Pakistan and Bangladesh. In addition, our analysis revealed that the existence of possible tick vectors of the genera Hyalomma, Haemophysalis, Rhipicephalus and Amblyomma may be responsible for the diverse and wide distribution of different Theileria species. However, the competence of these tick vectors for different Theileria species still need to be explored. Therefore, further studies are needed to bridge this gap and to improve the health and production of livestock and reduce economic losses due to theileriosis in India, Pakistan and Bangladesh. Furthermore, we selected representative 18S rRNA sequences for T. annulata from the different regions to infer phylogenetic relationship. Phylogenetic analysis of the selected isolates clustered in different clades which might be due to the variation in a hypervariable region of 18S rRNA. The outcome of this analysis is expected to provide a coherent and integrated framework about the different Theileria species prevailing in these countries and contribute to improving the surveillance and control plans of various Theileria species in the region.

Diversity and Distribution of Theileria Species and Their Vectors in Ruminants from India, Pakistan and Bangladesh

The current study was carried out to check the prevalence of tick infestation in sheep and goats in district Multan, Punjab, Pakistan. For this purpose, a total of 45 animals (25 sheep and 20 goats) were screened out. The various body parts like ears, udder and tails of animals were checked for tick collection. Out of total, 17(68.00%) sheep and 08(40.00%) goats were found infested with tick species. Three tick species such as Rhipicephalus sanguineus, Hyalomma anatolicum and Hyalomma marginatum were identified during the present study. The maximum infestation rate was in sheep as compared to goats. Mostly tick was collected from ears that are suitable site for tick infestation. The maximum tick infestation was in summer months (June-July) because these months are favorable for Tick development and reproduction
