The invention relates to a method for producing pancreatin with reduced viral and microbial contamination, comprising the steps of (a) providing the pancreatin in solid form with a residual moisture of 0.5 weight % or less, down to almost zero, based on the pancreatin provided; (b) subjecting the pancreatin provided in step (a) to a heat treatment at a temperature of 84° C., preferably 80° C. and below; wherein, the biological activity of the pancreatin obtained in step (b) corresponds to at least 50% of the biological activity of the pancreatin provided in step (a); and the viral infectiousness of the pancreatin obtained in step (b) has been reduced by a factor of more than 1 log10 in comparison with the viral infectiousness of the pancreatin provided in step (a), as well as a pancreatin produced according to this method and its use for producing a medicine or a nutritional supplement.

Pancreatin and method for reducing the viral and microbial contamination of pancreatin

A method for preparing a high concentration protease formulation comprising the following steps: a) grinding a frozen pancreas gland; b) autolysis of the mixture of pancreas gland obtained in a); c) separating the insoluble fibres in order to obtain a filtrate; d) mixing the filtrate with isopropanol in order to obtain a precipitate; e) purifying the precipitate by washing it with isopropanol and f) isolating the precipitate of step e) by filtration. The process allows the preparation of a formulation having a content of protease having at least 100,000 USP U/g of residual protease activity. This formulation may be used for the treatment of pancreatic disorders.

Pancreatin with high apparent density can be obtained by autolysis of an aqueous suspension of pancreatic tissue, removing fibres, water and fats, precipitating it and drying the precipitate, and by the precipitated pancreatin being adjusted by stirring with isopropyl alcohol or acetone to a concentration between 70 and 85% isopropanol or 80 to 95% acetone, being removed by filtration with suction or centrifugation, and dried, treating with dry air or nitrogen.
