Lignin, a complex aromatic polymer in terrestrial plants, contributes significantly to biomass recalcitrance to microbial and/or enzymatic deconstruction. To reduce biomass recalcitrance, substantial endeavors have been exerted on pretreatment and lignin engineering in the past few decades. Lignin removal and/or alteration of lignin structure have been shown to result in reduced biomass recalcitrance with improved cell wall digestibility. While high lignin content is usually a barrier to a cost-efficient application of bioresources to biofuels, the direct correlation of lignin structure and its concomitant properties with biomass remains unclear due to the complexity of cell wall and lignin structure. Advancement in application of biorefinery to production of biofuels, chemicals, and bio-derived materials necessitates a fundamental understanding of the relationship of lignin structure and biomass recalcitrance. In this mini-review, we focus on recent investigations on the influence of lignin chemical properties on bioprocessability-pretreatment and enzymatic hydrolysis of biomass. Specifically, lignin-enzyme interactions and the effects of lignin compositional units, hydroxycinnamates, and lignin functional groups on biomass recalcitrance have been highlighted, which will be useful not only in addressing biomass recalcitrance but also in deploying renewable lignocelluloses efficiently.

Current Understanding of the Correlation of Lignin Structure with Biomass Recalcitrance

Lignin is a complex aromatic biopolymer that strengthens and waterproofs plant secondary cell walls, enabling mechanical stability in trees and long-distance water transport in xylem. Lignin removal is a key step in paper production and biomass conversion to biofuels, motivating efforts to re-engineer lignin biosynthesis. However, the physical nature of lignin’s interactions with wall polysaccharides is not well understood. Here we show that lignin self-aggregates to form highly hydrophobic and dynamically unique nanodomains, with extensive surface contacts to xylan. Solid-state NMR spectroscopy of intact maize stems, supported by dynamic nuclear polarization, reveals that lignin has abundant electrostatic interactions with the polar motifs of xylan. Lignin preferentially binds xylans with 3-fold or distorted 2-fold helical screw conformations, indicative of xylans not closely associated with cellulose. These findings advance our knowledge of the molecular-level organization of lignocellulosic biomass, providing the structural foundation for optimization of post-harvest processing for biofuels and biomaterials. The interactions of lignin with polysaccharides in plant secondary cell walls are not well understood. Here the authors employ solid-state NMR measurements to analyse intact stems of maize, Arabidopsis, switchgrass and rice and observe that lignin self-aggregates and forms highly hydrophobic microdomains that make extensive surface contacts to xylan.

Common processes drive the thermochemical pretreatment of lignocellulosic biomass

Structural changes across multiple length scales associated with hydrothermal pretreatments of biomass were investigated by using small- and wide-angle X-ray and neutron scattering on oriented specimens. Isotropic and anisotropic scattering components were numerically separated and then interpreted as contributions from matrix and cellulose components, respectively. Equatorial diffraction peaks present in the isotropic component became sharper after hydrothermal treatments or ammonia treatment. Before pretreatment the wet cell wall was found to be homogeneous in the 10–100 nm range and scattering below Q = 0.5 (nm−1) was dominated by surface scattering from the lumen. After pretreatment with acid or steam at 160 or 180 °C, density fluctuation developed in the cell wall at length scales above 10 nm, most likely due to lateral coalescence of microfibrils that partially co-crystallize to give larger apparent crystal sizes. A density fluctuation up to about 100 nm appeared in the isotropic component after acid and steam pretreatments due to morphological changes in the hemicellulose and lignin matrix.
