Selective Inhibition of Osteoclast Vacuolar H+-ATPase
Carlo Farina,Stefania Gagliardi +1 more
TL;DR: Four independent approaches that did not start from bafilomycin have led to the discovery of a different class of V-ATPase inhibitors, among which 4-(2,6-dichlorobenzoyl)amino-2-trifluoromethyl(benzoimidazol-1-yl)acetyl morpholine was the most effective in preventing bone resorption in an ovariectomized rat model of osteoporosis.
Journal ArticleDOI
Reaction of Grignard reagents with carbonyl compounds under continuous flow conditions
Elena Riva,Stefania Gagliardi,Marisa Martinelli,Daniele Passarella,Daniele Vigo,Anna Rencurosi +5 more
TL;DR: In this paper, a continuous flow reactor was used to react carbonyl compounds with Grignard reagents at room temperature in an efficient and safe manner, and the results showed that a small collection of secondary and tertiary alcohols were obtained using the set-up protocol.
Journal ArticleDOI
5-(5,6-Dichloro-2-indolyl)-2-methoxy-2,4-pentadienamides: novel and selective inhibitors of the vacuolar H+-ATPase of osteoclasts with bone antiresorptive activity.
Stefania Gagliardi,Guy Marguerite Marie Gerard Nadler,Emanuela Consolandi,Carlo Parini,Marcel Morvan,Marie-Noelle Legave,Pietro Belfiore,Andrea Zocchetti,Geoffrey D. Clarke,Ian E. James,Ponnal Nambi,Maxine Gowen,Carlo Farina +12 more
TL;DR: The validity of the osteoclast proton pump as a useful molecular target to produce novel inhibitors of bone resorption, potentially useful as antiosteporotic agents is supported.
Journal ArticleDOI
Cinnamic Anilides as New Mitochondrial Permeability Transition Pore Inhibitors Endowed with Ischemia-Reperfusion Injury Protective Effect in Vivo
Daniele Fancelli,Agnese Abate,Raffaella Amici,Paolo Bernardi,Marco Ballarini,Anna Cappa,Giacomo Carenzi,Andrea Colombo,Cristina Contursi,Fabio Di Lisa,Giulio Dondio,Stefania Gagliardi,Eva Milanesi,Saverio Minucci,Saverio Minucci,Gilles Pain,Pier Giuseppe Pelicci,Alessandra Saccani,Mariangela Storto,Florian Thaler,Mario Varasi,Manuela Villa,Simon Plyte +22 more
TL;DR: In vitro and in vivo data are presented for (E)-3-(4-fluoro-3-hydroxy-phenyl)-N-naphthalen-1-yl-acrylamide 22, one of the most interesting compounds, able to attenuate opening of the mPTP and limit reperfusion injury in a rabbit model of acute myocardial infarction.
Journal ArticleDOI
Synthesis and structure-activity relationships of bafilomycin A1 derivatives as inhibitors of vacuolar H+-ATPase
Stefania Gagliardi,P A Gatti,Pietro Belfiore,Andrea Zocchetti,Geoffrey D. Clarke,Carlo Farina +5 more
TL;DR: 31 derivatives of bafilomycin A1 were synthesized and tested for their ability to inhibit differentially the V-ATPase-driven proton transport in membrane vesicles derived from chicken osteoclasts and bovine chromaffin granules.
