This study describes a comparison between LC-UV and LC–MS method for the simultaneous analyses of a few disease-modifying agents of multiple sclerosis. Quantitative determination of fampridine (FAM), teriflunomide (TFM) and dimethyl fumarate (DMF) was performed in human plasma with the recovery values in the range of 85–115%. A reversed-phase high-performance liquid chromatography (HPLC) with UV as well as MS detection is used. The method utilizes an XBridge C18 silica column and a gradient elution with mobile phase consisting of ammonium formate and acetonitrile at a flow rate of 0.5 mL min−1. The method adequately resolves FAM, TFM and DMF within a run time of 15 min. Owing to low molecular weights, the estimation of DMF and FAM is more versatile in UV than MS detection. With LC-UV, the detection limits of FAM, TFM and DMF were 0.1, 0.05, 0.05 μg and the quantification limit for all the analytes was 1 μg. With LC–MS, the detection and quantification limits for all of the analytes were 1 and 5 ng, respectively. The two techniques were completely validated and shown to be reproducible and sensitive. Role of dimethyl fumarate in oxidative stress of multiple sclerosis: A review.

A method for the determination of dimethyl fumarate (DMF) in desiccant and antimould sachets, employed for protecting consumer products from humidity and mould, has been developed. The method is based on a solid–liquid extraction followed by HPLC-UV analysis. The method was validated with respect to recovery, linearity, limits of detection and quantitation and precision. The recovery was 98%. The correlation coefficient value (r) was equal to 0.94. Both intra- and inter-day precisions were studied at several concentration levels, being satisfactory in all cases (RSD < 5). Limits of detection and quantification values were in the low microgram per gram level, thus allowing the determination of DMF at concentrations below the limit established (0.1 mg/kg) by the recent EU Directive (Decision 2009/251/EC). The proposed procedure was applied for the determination of the target compound in 41 desiccant and antimould samples. DMF was detected in 39.0% of samples and its content in many samples exceeded the legal limits. The results of our analysis highlight the high risk of exposure to this powerful allergic sensitizer for consumers. Copyright © 2011 John Wiley & Sons, Ltd.

Determination of dimethyl fumarate in desiccant and antimould sachets by reversed‐phase liquid chromatography

Near infrared spectroscopy (NIRS) has been proved to be a powerful analytical tool in different fields. However, because of the low sensitivity in near infrared region, it is a significant challenge to detect trace analytes with normal NIRS technique. A novel enrichment technique called fluidized bed enrichment has been developed recently to improve sensitivity of NIRS which allows a large volume solution to pass through within a short time. In this paper, fluidized bed enrichment method was applied in the determination of trace dimethyl fumarate in milk. Macroporous styrene resin HZ-816 was used as adsorbent material, and 1 L solution of dimethyl fumarate was run to pass through the material for concentration. The milk sample was pretreated to remove interference matters such as protein, fat, and then passed through the material for enrichment; after that, diffuse reflection NIR spectra were measured for the analyte concentrated on the material directly without any elution process. The enrichment and spectral measurement procedures were easy to operate. NIR spectra in 900–1,700 nm were collected for dimethyl fumarate solutions in the concentration range of 0.506–5.060 μg/mL and then used for multivariate calibration with partial least squares (PLS) regression. Spectral pretreatment methods such as multiplicative scatter correction, first derivative, second derivative, and their combinations were carried out to select the optimal PLS model. Root mean square error of cross-validation calculated by leave-one-out cross-validation is 0.430 μg/mL with ten PLS factors. Ten samples in an independent test set were predicted by the model with the mean relative error of 5.33 %. From the results shown in this work, it can be concluded that the NIR technique coupled with on-line enrichment method can be expanded for the determination of trace analytes, and its applications in real liquid samples like milk and juice may also be feasible.

Fast determination of trace dimethyl fumarate in milk with near infrared spectroscopy following fluidized bed enrichment

A simple method for the determination of dimethyl fumarate (DMFu) in silica gel pouches has been developed. The gas chromatographic behaviour of DMFu was investigated; the instrumental method, based on EI mass spectrometry coupled with an Ion Trap Detector operating whether in full scan mode or in MS/MS mode was also investigated. Several factors have been evaluated and optimized during the development process: solvent and temperature of extraction, type of stationary phase in capillary column. The analytical procedure consists of two steps as follows: (a) the sample preparation using 10 g of sample extracted with acetonitrile in an ultrasonic, heated bath and (b) the determination by gas chromatography-ion trap mass spectrometry. Mass spectrometry in conjunction with chromatographic separation is a very powerful combination for identification of an analyte in the extract in spite of selective detectors employed with GC, such as ECD, FPD and NPD, that offer only limited specificity. Blank samples show no interfering peaks in the areas of interest, so the specificity of the method was assured under the investigated GC/MS conditions. The method has been validated in terms of recovery, repeatability, linearity, detection limits and measurement uncertainty. The results obtained meet both the method validation criteria and requirements of the European/National legislation. The method was verified to be accurate with 97 % mean recoveries at 0.05 mg/kg and 1.00 mg/kg levels; the repeatability (expressed as RSD %) was found to be better than 15 %. Good linearity was found in the range between 0.05 μg/mL and 5.0 μg/mL and a value of R² > 0.9998 was calculated. The procedure ensures high specificity and a good sensitivity with detection limit (ld) of 0.02 mg/kg and quantification limit (LOQ) of 0.05 mg/kg. This procedure has been successfully applied for the analysis of several hundred of real samples collected during a monitoring control plan started last year in our country. All samples exceeding the maximum allowed level of DMFu (0, 1 mg/kg) were confirmed by GC/MS/MS (ITD) for a higher degree of confidence in identification. The main advantages of this method include: rapidity, simplicity (few, simple steps), reliability, cheapness, no need for long and difficult clean-up and evaporation steps, high specificity by using GC/MS and GC/MS/MS, availability for routine monitoring.
