Disclosed are methods and compositions for the detection, diagnosis, prognosis, and therapy of hematological malignancies, and in particular, B cell leukemias, lymphomas and multiple myelomas. Disclosed are compositions, methods and kits for eliciting immune and T cell responses to specific malignancy-related antigenic polypeptides and antigenic polypeptide fragments thereof in an animal. Also disclosed are compositions and methods for use in the identification of cells and biological samples containing one or more hematological malignancy-related compositions, and methods for the detection and diagnosis of such diseases and affected cell types. Also disclosed are diagnostic and therapeutic kits, as well as methods for the diagnosis, therapy and/or prevention of a variety of leukemias and lymphomas.

Compositions and methods for the detection, diagnosis and therapy of hematological malignancies

A single injection of 10 microliters of antiserum to total brain ganglioside onto (and into) the sensorimotor cortex of the rat resulted in recurrent spiking in the cortical electroencephalogram, lasting from 7 to 17 days. Absorption of antibody with pure monosialoganglioside (GM1) completely abosished the effect. Spiking was reactivated after 4 weeks by intramuscular injection of pentylenetetrazole.

— Gangliosides G1 to G5 were isolated from human brain by means of TLC and tested with respect to their specificity to antisera against normal brain and Tay-Sachs brain gangliosides by agar double diffusion analysis. Gangliosides G2 and G4 gave precipitation reactions with antisera to normal human gangliosides (NHG) while only ganglioside G6 reacted with antisera to Tay-Sachs gangliosides (TSG). Additional specificity information was also obtained by use of the enzyme neuraminidase for the removal of specific sialic acid (NANA) residues. It was concluded from these data that the specificity of the anti-NHG antibodies is determined by the presence of a galactose (β1, 3) N-acetyl galactos-amine–while that of anti-TSG antibodies is due to a N-acetyl galactosamine (β1, 4) galactose-end sequence. By means of natural compounds of known structure it was found that both the sequence of carbohydrate residues and position of NANA residues in the molecule played a critical role in the formation of precipitation bands with NHG-antisera. This information was utilized to distinguish one isomeric form of disialoganglioside from another, i.e. G2 from G3 and to confirm the structure of the trisialoganglioside, G1. The immunochemical method appears to be a useful one for elucidating structural differences in ganglioside molecules.

SummaryGangliosides from normal human, Tay-Sachs' and beef brain tissues administered intramuscularly with Freund's adjuvant, caused the production of precipitating antibodies in 15 out of 16 rabbits. Agar double diffusion analysis showed the presence of 2–3 or more precipitin bands in the reaction of each antigen with its homologous antiserum. Antiserum to normal human brain gangliosides formed only one precipitin band with gangliosides from Tay-Sachs' brain, while antiserum to gangliosides from Tay-Sachs' brain did not cross react with the gangliosides from normal brain. A purified monosialoganglioside from Tay-Sachs' brain gave one precipitin band only with the antiserum to gangliosides extracted from Tay-Sachs' brain.
