Simple Summary Cashmere goats are one of the most important animal breeds in northern China. Cashmere is characterised by good quality and high yield and represents an excellent textile material. However, the molecular mechanism underlying the development of cashmere hair follicles is poorly understood, which limits the further development of the cashmere industry. Here, we used whole-transcriptome sequencing to analyse goat embryonic skin samples and carried out experimental verification. We found that fibroblast growth factor 10 and the interacting non-coding RNA miR-184 play important roles in dermal papilla cells of cashmere goat secondary hair follicles. This discovery not only further improves the theoretical understanding of the development of cashmere wool follicles but also has significant implications for functional research on fibroblast growth factor 10 and miR-184. Abstract Cashmere, a keratinised product of secondary hair follicles (SHFs) in cashmere goats, holds an important place in international high-end textiles. However, research on the complex molecular and signal regulation during the development and growth of hair follicles (HFs), which is essential for the development of the cashmere industry, is limited. Moreover, increasing evidence indicates that non-coding RNAs (ncRNAs) participate in HF development. Herein, we systematically investigated a competing endogenous RNA (ceRNA) regulatory network mediated by circular RNAs (circRNAs), microRNAs (miRNAs), and messenger RNAs (mRNAs) in skin samples of cashmere goat embryos, using whole-transcriptome sequencing technology. We obtained 6468, 394, and 239 significantly differentially expressed mRNAs, circRNAs, and miRNAs, respectively. These identified RNAs were further used to construct a ceRNA regulatory network, mediated by circRNAs, for cashmere goats at a late stage of HF development. Among the molecular species identified, miR-184 and fibroblast growth factor (FGF) 10 exhibited competitive targeted interactions. In secondary HF dermal papilla cells (SHF-DPCs), miR-184 promotes proliferation, inhibits apoptosis, and alters the cell cycle via the competitive release of FGF10. This study reports that FGF10 and its interaction with ncRNAs significantly affect SHF-DPCs, providing a reference for research on the biology of HFs in cashmere goats and other mammals.

Effect of Fibroblast Growth Factor 10 and an Interacting Non-Coding RNA on Secondary Hair Follicle Dermal Papilla Cells in Cashmere Goats’ Follicle Development Assessed by Whole-Transcriptome Sequencing Technology

Simple Summary In this study, we investigated the effects of Moringa oleifera leaf powder (MOLP) on growth performance, carcass characteristics, and cecum micro-organisms of broiler chickens by using different levels of MOLP instead of canola cake. The results showed that 5% MOLP had an improving effect on broiler growth performance and carcass characteristics in the early and late stages. The sequence of cecal microbiota from broiler chickens via 16S rRNA revealed that 5% MOLP is likely to enhance the growth performance and carcass characteristics of broiler chickens by regulating the relative abundance of intestinal flora. The results of this study can provide some reference for the application of MOLP in livestock and poultry farming. Abstract Currently, the lack of protein source feed has become a pressing issue. Moringa oleifera leaf powder (MOLP) has good potential for the development of protein-derived feeds due to its good protein quality and abundance, but little is known about its effects on broiler growth performance and cecal microbiota. In this study, the chickens were fed different rates of MOLP (1%, 3%, 5%, 7%, and 9%) instead of the rape seed cake, and the effects of different levels of MOLP on growth performance, carcass characteristics, and cecal microbiota of the broilers were evaluated at two different growth stages (day 28 and day 56). In terms of growth performance, the best results were obtained at the 3% MOLP level in the early stages (p < 0.05). In terms of carcass characteristics, in the early stage, the level of 5% MOLP had the best effect; in the later stage, 5% MOLP also had the best effect. In terms of cecal microbial changes, the alpha diversity analysis revealed that 5% MOLP enhanced the richness and diversity of broiler intestinal flora. At the phylum level, the addition of 5% MOLP adjusted the relative abundance of Firmicutes and Bacteroidetes to a level close to that of the A1 group on day 28, while 5% MOLP significantly reduced the relative abundance of Bacteroidetes (p < 0.05) compared to the A2 group on day 56, and the relative abundance of Firmicutes was still higher in the D2 group than in the A2 group (p < 0.05). At the genus level, MOLP addition consistently and significantly increased the relative abundance of Bacteroides (p < 0.05), except for 3% on day 28 and 1% on day 56. For Oscillospira, increasing MOLP levels in the pre- and post-period resulted in a significant decrease in the relative abundance of Oscillospira (p < 0.05). In conclusion, MOLP helps to enhance growth performance and carcass characteristics and improve the cecal microbial structure of broilers. The recommended rate of MOLP addition for broilers is 5% in both the early and late stages.

Moringa oleifera Leaf Powder as New Source of Protein-Based Feedstuff Improves Growth Performance and Cecal Microbial Diversity of Broiler Chicken

Background Artificial insemination (AI) is an effective reproductive technique to improve the performance of cashmere goats and prevent the spread of diseases, and the quality of the semen determines the success of AI. The potential of Moringa oleifera leaf powder (MOLP) and Moringa oleifera leaf ethanolic extract (MOLE) to improve semen quality has been reported, but the underlying mechanisms remain unclear. For the purpose, 18 mature male cashmere goats were randomly assigned into three groups: the control (CON), MOLP, and MOLE groups. The CON group received distilled water orally; the MOLP group was orally treated with 200 mg/kg body weight (BW) MOLP; and the MOLE group was orally treated with 40 mg/kg BW MOLE. Results Results showed that MOLE contained long-chain fatty acids and flavonoids. Treatment with MOLP and MOLE increased the activities of the serum catalase, superoxide dismutase, and glutathione peroxidase (P < 0.05), enhanced the total antioxidant capacity (P < 0.05), and reduced the serum malondialdehyde level (P < 0.05). At the same time, MOLE increased the contents of serum gonadotropin releasing hormone and testosterone (P < 0.05). Moreover, MOLE significantly increased sperm concentration, motility, and viability (P < 0.05). Meanwhile, MOLE raised the Chao1 index (P < 0.05) and altered the composition of the rumen microbiota; it also raised the relative abundance of Treponema (P < 0.05) and Fibrobacter (P < 0.05) and reduced the relative abundance of Prevotella (P < 0.1). Correlation analysis revealed the genus Prevotella was significantly negatively correlated with sperm concentration, as well as sperm motility and viability. Furthermore, MOLE significantly increased the rumen levels of the steroid hormones testosterone and dehydroepiandrosterone (P < 0.05), as well as the polyunsaturated fatty acids (PUFAs) alpha-Linolenic acid, gamma-Linolenic acid, docosapentaenoic acid, and 9-S-Hydroperoxylinoleicacid (P < 0.05). Conclusions Oral MOLE supplementation can improve semen quality by increasing the antioxidant capacity and altering the rumen microbiota and metabolites of cashmere goats. Moreover, the MOLP supplementation could enhance the antioxidant capacity of cashmere goats. Cashmere goat has a typical characteristic in seasonal growth of cashmere. Studies have shown that one of the main factors affecting the cyclical growth of the cashmere is the photoperiod, however, its molecular mechanism remains unclear. Inner Mongolia Arbas cashmere goat was used to reveal the mRNA-microRNA regulatory mechanisms of cashmere growth in different photoperiod. Skin samples from cashmere goats under light control (short photoperiod) and normal conditions (long photoperiod) were collected. Sequencing was performed after RNA extraction. The differentially expressed miRNA and mRNA expression profiles were successfully constructed. We found 56 significantly differentially expressed known mRNAs (P<0.01) and 14 microRNAs (P<0.05). The association analysis of the microRNAs and mRNAs showed that two differentially expressed miRNAs might be targeted by six differentially expressed genes. Targeting relationships of these genes and miRNAs are revealed and verified. In all, the light control technology provides a new way to promote cashmere growth. Our results provide some references in the cashmere growth and development.

Transcriptomes reveal microRNAs and mRNAs in different photoperiods influencing cashmere growth in goat

Cysteamine (CS), as a feed supplement, can increase the level of growth hormone (GH) in the blood, promote animal growth. However, little attention has been paid to the effects of CS on the rumen microbiome and metabolic profile in cashmere goats. This study aimed to assess the effects of rumen microbiota, metabolites, and plasma antioxidative capacity induced by CS supplementation in cashmere goats. We selected 30 Inner Mongolia white cashmere goat ewes (aged 18 months), and randomly separate the goats into three groups (n = 10 per group) to experiment for 40 days. Oral 0 (control group, CON), 60 (low CS, LCS), or 120 mg/kg BW−1 (high CS, HCS) coated CS hydrochloride every day. Using 16S and internal transcribed spacer (ITS) rRNA gene amplicon sequencing, we identified 12 bacterial and 3 fungal genera with significant changes among the groups, respectively. We found a significant increase in rumen NH3-N and total volatile fatty acid (TVFA) concentrations in the LCS and HCS groups compared with the CON. With untargeted LC–MS/MS metabolomics, we screened 59 rumen differential metabolites. Among the screened metabolites, many unsaturated and saturated fatty acids increased and decreased with CS treatment, respectively. CS supplementation increased the levels of plasma total antioxidant capacity (T-AOC), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), GH, and insulin-like growth factor-1(IGF-1). Spearman correlation analysis revealed that the abundance of U29-B03, Lactococcus, and Brochothrix were positively associated with the levels of δ2-THA, TVFA and antioxidant capacity. In conclusion, CS significantly affected rumen microbiota and fermentation parameters, and ultimately inhibited the biohydrogenation of rumen metabolites, enhanced plasma antioxidant capacity, and regulated some hormones of the GH–IGF-1 axis. This study provides an overall view into the CS application as a strategy to improve health production in cashmere goats.
