In complex media such as white paint and biological tissue, light encounters nanoscale refractive-index inhomogeneities that cause multiple scattering. Such scattering is usually seen as an impediment to focusing and imaging. However, scientists have recently used strongly scattering materials to focus, shape and compress waves by controlling the many degrees of freedom in the incident waves. This was first demonstrated in the acoustic and microwave domains using time reversal, and is now being performed in the optical realm using spatial light modulators to address the many thousands of spatial degrees of freedom of light. This approach is being used to investigate phenomena such as optical super-resolution and the time reversal of light, thus opening many new avenues for imaging and focusing in turbid media

Controlling waves in space and time for imaging and focusing in complex media

Optical imaging through and inside complex samples is a difficult challenge with important applications in many fields. The fundamental problem is that inhomogeneous samples such as biological tissue randomly scatter and diffuse light, preventing the formation of diffraction-limited images. Despite many recent advances, no current method can perform non-invasive imaging in real-time using diffused light. Here, we show that, owing to the ‘memory-effect’ for speckle correlations, a single high-resolution image of the scattered light, captured with a standard camera, encodes sufficient information to image through visually opaque layers and around corners with diffraction-limited resolution. We experimentally demonstrate single-shot imaging through scattering media and around corners using spatially incoherent light and various samples, from white paint to dynamic biological samples. Our single-shot lensless technique is simple, does not require wavefront-shaping nor time-gated or interferometric detection, and is realized here using a camera-phone. It has the potential to enable imaging in currently inaccessible scenarios. Diffraction-limited imaging in a variety of complex media is realized based on analysis of speckle correlations in light captured using a camera phone.

Non-invasive single-shot imaging through scattering layers and around corners via speckle correlations

Optical forces can be used to manipulate biological and colloidal material in a non-contact manner. This forms the foundation of a wealth of exciting science, particularly in the fields of physics, biology and soft condensed matter. Although the standard Gaussian single-beam trap remains a very powerful tool, shaping the phase and amplitude of a light field provides unusual light patterns that add a major new dimension to research into particle manipulation. This Review summarizes the impact and emerging applications of shaped light in the field of optical manipulation.

Shaping the future of manipulation

Light-sheet microscopy facilitates rapid, high-contrast, volumetric imaging with minimal sample exposure. However, the rapid divergence of a traditional Gaussian light sheet restricts the field of view (FOV) that provides innate subcellular resolution. We show that the Airy beam innately yields high contrast and resolution up to a tenfold larger FOV. In contrast to the Bessel beam, which also provides an increased FOV, the Airy beam's characteristic asymmetric excitation pattern results in all fluorescence contributing positively to the contrast, enabling a step change for light-sheet microscopy.

Optical imaging relies on the ability to illuminate an object, collect and analyse the light it scatters or transmits. Propagation through complex media such as biological tissues was so far believed to degrade the attainable depth, as well as the resolution for imaging, because of multiple scattering. This is why such media are usually considered opaque. Recently, we demonstrated that it is possible to measure the complex mesoscopic optical transmission channels that allow light to traverse through such an opaque medium. Here, we show that we can optimally exploit those channels to coherently transmit and recover an arbitrary image with a high fidelity, independently of the complexity of the propagation.

In any optical system, distortions to a propagating wavefront reduce the spatial coherence of a light field, making it increasingly difficult to obtain the theoretical diffraction-limited spot size. Such aberrations are severely detrimental to optimal performance in imaging, nanosurgery, nanofabrication and micromanipulation, as well as other techniques within modern microscopy. We present a generic method based on complex modulation for true in situ wavefront correction that allows compensation of all aberrations along the entire optical train. The power of the method is demonstrated for the field of micromanipulation, which is very sensitive to wavefront distortions. We present direct trapping with optimally focused laser light carrying power of a fraction of a milliwatt as well as the first trapping through highly turbid and diffusive media. This opens up new perspectives for optical micromanipulation in colloidal and biological physics and may be useful for various forms of advanced imaging.
