Numerous bacteria secrete low molecular weight compounds termed siderophores that have a high affinity for iron ions. Siderophores have a well-documented role as iron-scavenging chemicals, chelating iron ions in the environment whereupon the ferrisiderophores reenter the bacterial cells by means of specific cell-surface receptors. The iron is then released for incorporation into bacterial proteins. Here we show that in addition to its role as an iron-scavenger, the siderophore pyoverdine that is secreted by Pseudomonas aeruginosa regulates the production of at least three virulence factors (exotoxin A, an endoprotease, and pyoverdine itself), which are major contributors to the ability of this bacterium to cause disease. Regulation occurs through a transmembrane signaling system that includes an outer membrane receptor for ferripyoverdine, a signal-transducing protein that is predicted to extend from the periplasm into the cytoplasm, and a sigma factor. Expression of genes that form part of the regulon is triggered by pyoverdine so that this siderophore acts as a signaling molecule to control the production of secreted products. Recognition that a siderophore acts as a signaling molecule has important implications for the understanding of interactions between bacterial cells.

Siderophore-mediated signaling regulates virulence factor production in Pseudomonas aeruginosa

Production of exopolysaccharides (EPS) can be used as a criteria for the isolation of stress tolerant microorganisms. In the present study, EPS-producing fluorescent pseudomonads were isolated from alfisols, vertisols, inseptisols, oxisols, and aridisols of different semiarid millet growing regions of India and were screened in vitro for drought tolerance in trypticase soy broth supplemented with different concentrations of polyethylene glycol (PEG6000). Out of the total 81 isolates, 26 could tolerate maximum level of stress (−0.73 MPa) and were monitored for the amount of EPS produced under maximum level of water stress. The strain GAP-P45, isolated from alfisol of sunflower rhizosphere, showed the highest level of EPS production under water stress conditions, was identified as Pseudomonas putida on the basis of 16S rDNA sequence analysis, and was used as seed treatment to study its effect in alleviating drought stress effects in sunflower seedlings. Inoculation of Pseudomonas sp. strain GAP-P45 increased the survival, plant biomass, and root adhering soil/root tissue ratio of sunflower seedlings subjected to drought stress. The inoculated bacteria could efficiently colonize the root adhering soil and rhizoplane and increase the percentage of stable soil aggregates. Scanning electron microscope studies showed the formation of biofilm of inoculated bacteria on the root surface and this, along with a better soil structure, might have protected the plants from the water stress.

Alleviation of drought stress effects in sunflower seedlings by the exopolysaccharides producing Pseudomonas putida strain GAP-P45

Rhizobia described so far belong to three distinct phylogenetic branches within the alpha-2 subclass of Proteobacteria. Here we report the discovery of a fourth rhizobial branch involving bacteria of the Methylobacterium genus. Rhizobia isolated from Crotalaria legumes were assigned to a new species, "Methylobacterium nodulans," within the Methylobacterium genus on the basis of 16S ribosomal DNA analyses. We demonstrated that these rhizobia facultatively grow on methanol, which is a characteristic of Methylobacterium spp. but a unique feature among rhizobia. Genes encoding two key enzymes of methylotrophy and nodulation, the mxaF gene, encoding the alpha subunit of the methanol dehydrogenase, and the nodA gene, encoding an acyltransferase involved in Nod factor biosynthesis, were sequenced for the type strain, ORS2060. Plant tests and nodA amplification assays showed that "M. nodulans" is the only nodulating Methylobacterium sp. identified so far. Phylogenetic sequence analysis showed that "M. nodulans" NodA is closely related to Bradyrhizobium NodA, suggesting that this gene was acquired by horizontal gene transfer.

In present study Bacillus spp. screened for drought tolerance could tolerate minimal water potential (-0.73 MPa) were evaluated for plant growth promoting properties at –0.73 MPa. Drought stress affected growth of isolates as indicated by increased intracellular free amino acids, proline, total soluble sugars, and exopolysaccharides. Drought-tolerant plant growth promoting Bacillus spp.: effect on growth, osmolytes, and antioxidant status of maize under drought stress

A mutant, named 11B, hypersusceptible to aminoglycosides, tetracycline, and erythromycin was isolated after Tn 501 insertion mutagenesis of Pseudomonas aeruginosa PAO1. Cloning and sequencing experiments showed that 11B was deficient in an, at that time, unknown active efflux system that contains homologs of MexAB. This locus also contained a putative regulatory gene, mexZ , transcribed divergently from the efflux operon. Introduction of a recombinant plasmid that carries the genes of the efflux system restored the resistance of 11B to parental levels, whereas overexpression of these genes strongly increased the MICs of substrate antibiotics for the PAO1 host. Antibiotic accumulation studies confirmed that this new system is an energy-dependent active efflux system that pumps out aminoglycosides. Furthermore, this system appeared to function with an outer membrane protein, OprM. While the present paper was being written and reviewed, genes with a sequence identical to our pump genes, mexXY of P. aeruginosa , have been reported to increase resistance to erythromycin, fluoroquinolones, and organic cations in Escherichia coli hosts, although efflux of aminoglycosides was not examined (Mine et al., Antimicrob. Agents Chemother. 43:415–417, 1999). Our study thus shows that the MexXY system plays an important role in the intrinsic resistance of P. aeruginosa to aminoglycosides. Although overexpression of MexXY increased the level of resistance to fluoroquinolones, disruption of the mexXY operon in P. aeruginosa had no detectable effect on susceptibility to these agents.
