Journal ArticleDOI
Discovery of new Gram-negative antivirulence drugs: structure and properties of novel E. coli WaaC inhibitors.
François Moreau,Nicolas Desroy,Jean-Marie Genevard,Vanida Vongsouthi,Vincent Gerusz,G. Le Fralliec,Chrystelle Oliveira,Stéphanie Floquet,Alexis Denis,Sonia Escaich,K. Wolf,M. Busemann,A. Aschenbrenner +12 more
TL;DR: The approach to the identification of the first micromolar inhibitors of WaaC and the preliminary SAR generated from this family of 2- Daryl-5-methyl-4-(5-aryl-furan-2-yl-methylene)-2,4-dihydro-pyrazol-3-ones identified by virtual screening are reported.
Journal ArticleDOI
The MUT056399 Inhibitor of FabI Is a New Antistaphylococcal Compound
Sonia Escaich,L. Prouvensier,M. Saccomani,L. Durant,M. Oxoby,Vincent Gerusz,François Moreau,Vanida Vongsouthi,Kirsty Maher,Ian Morrissey,C. Soulama-Mouze +10 more
TL;DR: In vivo, MUT056399, administered subcutaneously, protected mice from a lethal systemic infection induced by MSSA, MRSA, and vancomycin-intermediate S. aureus strains (50% effective doses ranging from 19.3 mg/kg/day to 49.6 mg/ kg/day), and support MUT56399 as a very promising candidate for a novel drug to treat severe staphylococcal infections.
Journal ArticleDOI
Towards Gram-negative antivirulence drugs: new inhibitors of HldE kinase.
Nicolas Desroy,François Moreau,Sophia Briet,Géraldine Le Fralliec,Stéphanie Floquet,Lionel Durant,Vanida Vongsouthi,Vincent Gerusz,Alexis Denis,Sonia Escaich +9 more
TL;DR: Development of a biochemical assay suitable for high-throughput screening allowed the discovery of inhibitors 1 and 2 of HldE kinase, which led to highly potent compounds.
Journal ArticleDOI
Systematic Synthesis of Inhibitors of the Two First Enzymes of the Bacterial Heptose Biosynthetic Pathway: Towards Antivirulence Molecules Targeting Lipopolysaccharide Biosynthesis
Maxime Durka,Abdellatif Tikad,Régis Périon,Michael Bosco,Mounir Andaloussi,Stéphanie Floquet,Elodie Malacain,François Moreau,Mayalen Oxoby,Vincent Gerusz,Stéphane P. Vincent +10 more
TL;DR: This study describes the first inhibitors of GmhA ever reported, and paves the way to the design of a second generation of molecules targeting the bacterial virulence.
Journal ArticleDOI
From Triclosan toward the Clinic: Discovery of Nonbiocidal, Potent FabI Inhibitors for the Treatment of Resistant Bacteria
Vincent Gerusz,Alexis Denis,Fabien Faivre,Yannick Bonvin,Mayalen Oxoby,Sophia Briet,Géraldine LeFralliec,Chrystelle Oliveira,Nicolas Desroy,Cédric Raymond,Laëtitia Peltier,François Moreau,Sonia Escaich,Vanida Vongsouthi,Stéphanie Floquet,Elodie Drocourt,Armelle Walton,Laure Prouvensier,Marc Saccomani,Lionel Durant,Jean-Marie Genevard,Vanessa Sam-Sambo,Coralie Soulama-Mouze +22 more
TL;DR: Some elements of the optimization program to decouple triclosan's specific FabI effect from its nonspecific cytotoxic component delivered highly specific, potent, and nonbiocidal new FabI inhibitors.
