In order to estimate the production of charcoal and the atmospheric emissions of trace gases volatilized by burning we have estimated the global amounts of biomass which are affected by fires. We have roughly calculated annual gross burning rates ranging between about 5 Pg and 9 Pg (1 Pg = 1015 g) of dry matter (2–4 Pg C). In comparison, about 9–17 Pg of above-ground dry matter (4–8 Pg C) is exposed to fires, indicating a worldwide average burning efficiency of about 50%. The production of dead below-ground dry matter varies between 6–9 Pg per year. We have tentatively indicated the possibility of a large production of elemental carbon (0.5–1.7 Pg C/yr) due to the incomplete combustion of biomass to charcoal. This provides a sink for atmospheric CO2, which would have been particularly important during the past centuries. From meager statistical information and often ill-documented statements in the literature, it is extremely difficult to calculate the net carbon release rates to the atmosphere from the biomass changes which take place, especially in the tropics. All together, we calculate an overall effect lof the biosphere on the atmospheric carbon dioxide budget which may range between the possibilities of a net uptake or a net release of about 2 Pg C/yr. The release of CO2 to the atmosphere by deforestation projects may well be balanced by reforestation and by the production of charcoal. Better information is needed, however, to make these estimates more reliable.

Biodegradation of lignin in a compost environment: a review

Culture parameters influencing metabolism of synthetic14C-lignins to14CO2 in defined media have been studied in shallow batch cultures of the ligninolytic wood-destroying HymenomycetePhanerochaete chrysosporium Burds. Study of the effect of O2 concentration in the gas phase above non-agitated cultures indicated essentially complete absence of attack on the lignin polymer at 5% O2 in N2, and a 2- to 3-fold enhancement by 100% O2 as compared to air (21% O2). Agitation of the cultures resulting in the formation of mycelial pellets greatly suppressed lignin decomposition. The optimum culture pH for lignin decomposition was 4 to 4.5, with marked suppression above 5.5 and below 3.5. The source of nutrient nitrogen (NO

 , amino acids) had little influence on lignin decomposition, but the concentration of nitrogen was critical; decomposition at 24 mM was only 25–35% of that at 2.4 mM N. Thiamine was the only vitamin required for growth and lignin decomposition. Under the optimum conditions developed, decomposition of 5 mg of synthetic lignin was accompanied by utilization of approximately 100 mg of glucose. The influence of the various culture parameters was analogous for metabolism of synthetic lignin labeled in the ring-,side chain-, and methoxyl carbon atoms.

Influence of culture parameters on lignin metabolism by Phanerochaete chrysosporium

The white rot fungus Phanerochaete chrysosporium degraded DDT [1,1,-bis(4-chlorophenyl)-2,2,2-trichloroethane], 3,4,3',4'-tetrachlorobiphenyl, 2,4,5,2',-4',5'-hexachlorobiphenyl, 2,3,7,8-tetrachlorodibenzo-p-dioxin, lindane (1,2,3,4,5,6-hexachlorocylohexane), and benzo[a]pyrene to carbon dioxide. Model studies, based on the use of DDT, suggest that the ability of Phanerochaete chrysosporium to metabolize these compounds is dependent on the extracellular lignin-degrading enzyme system of this fungus.

Oxidation of persistent environmental pollutants by a white rot fungus.

Lignin is the most abundant renewable source of aromatic polymer in nature, and its decomposition is indispensable for carbon recycling. It is chemically recalcitrant to breakdown by most organisms because of the complex, heterogeneous structure. The white-rot fungi produce an array of extracellular oxidative enzymes that synergistically and efficiently degrade lignin. The major groups of ligninolytic enzymes include lignin peroxidases, manganese peroxidases, versatile peroxidases, and laccases. The peroxidases are heme-containing enzymes with catalytic cycles that involve the activation by H2O2 and substrate reduction of compound I and compound II intermediates. Lignin peroxidases have the unique ability to catalyze oxidative cleavage of C-C bonds and ether (C-O-C) bonds in non-phenolic aromatic substrates of high redox potential. Manganese peroxidases oxidize Mn(II) to Mn(III), which facilitates the degradation of phenolic compounds or, in turn, oxidizes a second mediator for the breakdown of non-phenolic compounds. Versatile peroxidases are hybrids of lignin peroxidase and manganese peroxidase with a bifunctional characteristic. Laccases are multi-copper-containing proteins that catalyze the oxidation of phenolic substrates with concomitant reduction of molecular oxygen to water. This review covers the chemical nature of lignin substrates and focuses on the biochemical properties, molecular structures, reaction mechanisms, and related structures/functions of these enzymes.

Structure and Action Mechanism of Ligninolytic Enzymes

A definitive assay for microbiological and biochemical research on the biodegradation of lignin was developed using radioactive synthetic lignins specifically labeled in the side chains, aromatic rings or in the methoxyl groups. The [14C]lignins were prepared by oxidative polymerization with peroxidase and H2O2 Of specifically labeled coniferyl alcohol (4-hydroxy-3-methyoxycinnamyl alcohol). The synthetic polymers were shown by spectroscopic and chemical methods to contain the same intermonomer linkages found in natural lignins. Incubation of the [14C]lignins with known lignin-degrading fungi and with a forest soil resulted in 14CO2 evolution.

Preparation and microbial decomposition of synthetic [14C]ligins.

Lignin biodegradation in a variety of natural materials was examined using specifically labeled synthetic 14C-lignins. Natural materials included soils, sediments, silage, steer bedding, and rumen contents. Both aerobic and anaerobic incubations were used. No 14C-labeled lignin biodegradation to labeled gaseous products under anaerobic conditions was observed. Aerobic 14C-labeled lignin mineralization varied with respect to type of natural material used, site, soil type and horizon, and temperature. The greatest observed degradation occurred in a soil from Yellowstone National Park and amounted to over 42% conversion of total radioactivity to 14CO2 during 78 days of incubation. Amounts of 14C-labeled lignin mineralization in Wisconsin soils and sediments were significantly correlated with organic carbon, organic nitrogen, nitrate nitrogen, exchangeable calcium, and exchangeable potassium.
