Plumb's Veterinary Drug Handbook

Forty three dogs with non-neoplastic canine gastric and/or duodenal ulcers were diagnosed at the University of Tennessee Veterinary Teaching Hospital (UTVTH) and reviewed in conjunction with dogs reported in the literature. No age, sex, or breed predilection was found. Most of the 43 UTVTH dogs presented with clinical signs referable to gastrointestinal disease, but evidence of hemorrhage was not always present. Nonregenerative anemia was a common (33/43 dogs) finding. Diagnosis of ulcer disease was made by contrast radiography, with clinical evidence of gastrointestinal hemorrhage, or surgery, endoscopy, or necropsy. Treatment with nonsteroidal, anti-inflammatory drugs (NSAIDs) and hepatic disease were the two most common predisposing factors for ulcer disease. Dogs with liver disease tended to have duodenal ulcers, dogs receiving NSAID treatment tended to have pyloroantral ulcers, and dogs with mastocytosis had ulcers in multiple locations. Three dogs with duodenal ulcers receiving NSAID treatment had an additional predisposing factor. Surgical treatment (with or without medical treatment) had a good outcome when the predisposing factors could be controlled or eliminated. Only 3 of 27 dogs in both the surgical and medically treated groups died from ulcer disease, whereas 6 of 16 dogs in the untreated group died from ulcer disease.

Gastroduodenal ulceration in dogs. Retrospective study of 43 cases and literature review.

Nonsteroidal anti-inflammatory drugs (NSAIDs) are a group of heterogeneous compounds extensively used in both human and veterinary medicine for their antipyretic, anti-inflammation, and analgesic properties. NSAIDs consist of a wide range of pharmacologically active agents with different chemical structures, with similar therapeutic and adverse effects. The ASPCA Animal Poison Control Center received 22,206 NSAID incidents in dogs and cats (3% of total cases; dogs [15,823] and cats [1244]) during 2005 to 2010. This is roughly equivalent to 4% NSAID incidents reported in humans. The most common NSAID involved was ibuprofen, followed by aspirin, naproxen, deracoxib, meloxicam, diclofenac, piroxicam, indomethacin, nabumetone, and etodolac. This article provides a brief overview of classification, mechanism of action, pharmacologic and toxicologic properties, and treatment information involving frequently encountered human and veterinary NSAIDs in dogs and cats.

Toxicology of Frequently Encountered Nonsteroidal Anti-Inflammatory Drugs in Dogs and Cats

Nonsteroidal antiinflammatory drugs (NSAIDs) constitute a class of pharmacologically active agents with similar therapeutic actions and side effects, despite diverse chemical structures. NSAIDs inhibit the enzyme cyclo-oxygenase and, thus, decrease inflammation mediated by prostaglandins. Toxicoses due to NSAIDs are manifested primarily by gastrointestinal upset and hemorrhage and by renal damage. Management consists of detoxification measures, in addition to supportive and symptomatic care. The article outlines the incidence, clinical toxicity, mode of action, and pharmacokinetics of nonsteroidal antiinflammatory drugs. The toxic effects of NSAIDs on individual organ systems are described, and general management recommendations for the treatment ofNSAID toxicoses are presented.

Toxicology of Nonsteroidal Antiinflammatory Drugs

A double-blind, randomised, controlled, multicentre field study was conducted to compare the safety and efficacy of firocoxib chewable tablets and carprofen tablets in 218 dogs with osteoarthritis. Firocoxib is a non-steroidal anti-inflammatory drug with more than 350-fold selectivity in dogs for the inducible isoform of the enzyme cyclo-oxygenase-2. The efficacy, tolerance and ease of administration of firocoxib (5 mg/kg/day) and carprofen (4 mg/kg/day) were assessed by the owners and the attending veterinarians during 30 days of treatment. The efficacy was assessed in terms of the dogs' overall scores at the end of the treatment, based on the veterinarians' assessment of lameness, pain on manipulation/palpation, range of motion, and joint swelling; 92.5 per cent of the dogs treated with firocoxib and 92.4 per cent of the dogs treated with carprofen had improved. The reduction in lameness in the dogs treated with firocoxib was significantly greater than in the dogs treated with carprofen. The owners' evaluations were that 96.2 per cent of the dogs treated with firocoxib and 92.4 per cent of the dogs treated with carprofen had improved, and this difference was statistically significant.

Clinical evaluation of firocoxib and carprofen for the treatment of dogs with osteoarthritis

Naproxen, a nonsteroidal anti-inflammatory drug, induced gastric ulceration and bleeding in a dog. Clinical signs included vomiting, anemia, melena, and weakness. The dog responded to a blood transfusion and supportive treatment (lactated Ringer's solution with added K+ and B vitamins, an antacid, and cimetidine). Naproxen had been given to the dog by the owner, as treatment for shoulder stiffness.
