Abstract: Histoenzymorphologic investigations on β-D-glucuronidase in cortical granules in oocytes of some representative vertebrates revealed interesting differences in localisation of the enzyme in the two techniques employed using 8-hydroxyquinoline β-D-glucosiduronic acid and naphthol AS-BI β-D-glucosiduronic acid as substrates. In the former a nonspecific localisation in the entire cytoplasm of the oocytes excepting nucleus was evident whereas in the latter the cortical granules alone exhibited intense activity. To some extent this difference could be accounted for on the basis of yolk content of the eggs. β-D-glucoronidase-rich lysosomal cortical granules were found in the amphibian, reptilian and avian oocytes whereas mammalian oocyte cortical granules were devoid of this enzyme. β-D-glucuronidase in cortical granules was sensitive to low concentrations of Hg++, Cu++, glucuronic and mucic acids. Other histochemical evidence for the lysosomal nature of the cortical granules is presented and discussed. β-D-glucuronidase seems to have a role in ovum-nutrition and activation which, thus, make a new addition to the existing knowledge of physiological role of β-D-glucuronidase and lysosomes.

Abstract: Treatment of fixed frozen sections of rat kidney with 40–60% ethanol for 2 minutes at 0° C improved the cytologic localisation ofβ-glucuronidase in naphthol AS-BI post-coupling technique. Concentration of ethanol, duration of treatment and temperature are critical. Higher concentrations of ethanol, longer treatment and higher temperatures deteriorate the normal localisation of the enzyme. Such pretreatment can he introduced as a step in the original histochemical technique.

Abstract: A comparative and comprehensive report is given on the distribution of chromaffin and nonchromaffin cells in the lower vertebrates by means of iodate and chromaffin tests. Whatever be the distribution of these two types of cells, there is a gradual tendency for a close proximity between adrenaline cells from the chromaffin tissue and the nonchromaffin cells. This seems to support the claim that the nonchromaffin cells provide glucocorticoids to the adrenaline synthesizing mechanism in adrenaline cells. The increase and decrease respectively in adrenaline and noradrenaline from fishes to mammals is supported by the corresponding increase and decrease in adrenaline and noradrenaline cells. The possible evolutionary implications of these findings are discussed.

Abstract: An interesting case of a colloidal iron (CI) positive basophilic substance in the adrenal medullary cells of amphibia and reptilia is reported here. The substance, however, does not stain by alcian blue (AB). It is negative to PAS, Azure A, aldehyde fuchsin, AB at pH 1 and MgCl2 — AB though orthochromatically stained by toluidine blue at pH 3. More work is needed to establish the exact nature of the CI positive material.

Abstract: A puzzling feature of Colloidal Iron positive and Alcian Blue negative substance is encountered in yolk sac of young larvae of a fish —Tilapia mossambica. This yolk material is PAS positive (proved to be due to neutral mucopolysaccharide) and negative to Toluidine Blue, Azure A (pH 2 to 4.5), Aldehyde Fuchsin and AB pH 1. More work is necessary to establish the exact chemical nature of the CI positive material.
