Abstract: PCR amplification was conducted for genome DNA of Yak, Holstein and pian niu using 80 10bp random primers. 7 primers amplified the clear polymorphic bands, and F 15 (CCAGTACTCC) could be used as an important marker of distinguishing yak and holstein. Similarity index analysis of tested breeds indicated that the smallest (0 5942), the biggest (0.6456), and the moderate (0.6410) genetic distance existed between Holstein and pianniu, Holstein and yak, and pian niu and yak. The population genetic diversity index of yak, Holstein, and pien niu was 0.386, 0.412, and 0.498 respectively.
