An instrument for microspectrofluorimetry, based on the Leitz microspectrograph, with important features are monochromatic epi‐illumination, a reference channel and a measuring system using photon counting with automatic recording of data in digital form.
Abstract
SUMMARY This paper describes an instrument for microspectrofluorimetry, based on the Leitz microspectrograph. Important features are monochromatic epi-illumination, a reference channel and a measuring system using photon counting with automatic recording of data in digital form. To minimize fading, measurements of fluorescence intensity can be made within as little as 200 msec from the commencement of irradiation.

Fluorescently labelled molecules as probes of the structure and function of living cells.

TL;DR: A new approach to cell biology has been created by combining the techniques of micromanipulation of cells, fluorescence spectroscopy and low level light detection to study the spatial and temporal distribution, interaction and activity of specific molecules in living cells.

Demonstration of the neural crest origin of type I (APUD) cells in the avian carotid body, using a cytochemical marker system.

TL;DR: The dopamine phenotype in cells of quail origin provides a cytochemical marker which may be used for other allograft experiments and confirms earlier findings, using a biological (nuclear chromatin) marker, which showed the avian carotid body to be of neural crest origin.

The genesis of apudamyloid in endocrine polypeptide tumours: histochemical distinction from immunamyloid

TL;DR: It is suggested that in the case of insulinomas the fibril protein is derived from C-peptide chains and that similar products (prohormone derivatives or amine-binding granule protein derivatives) are responsible for other endocrine tumour amyloids.

A microspectrofluorometer with epi-illumination operated under computer control.

TL;DR: A new type of microspectrofluorometer with epi-illumination has been developed, operated on line with a digital computer permitting automatic operation of monochromator motors, electrical shutters and correction of spectra.

A method for differentiating dopamine from noradrenaline in tissue sections by microspectrofluorometry

TL;DR: Fluorophores induced from noradrenaline and dopamine in tissue sections by treatment with formaldehyde can be separated by their different behavior upon HCl treatment by using a microspectrofluorometer.

The melanocyte: its cytochemical and immunological relationship to cells of the endocrine polypeptide (APUD) series.

TL;DR: Normal and neoplastic melanocytes are shown to have cytochemical characteristics of the APUD1 cell series: uptake of L-dopa, high α-glycerophosphate dehydrogenase, histochemically demonstrable non-specific cholinesterase and non- specific esterases, and masked metachromasia.

Fluorescence metachromasia in polypeptide hormone-producing cells of the APUD series, and its significance in relation to the structure of the precursor protein

TL;DR: It is suggested that the conformation of the protein precursors of polypeptide hormones, in the storage granules of endocrine cells, is predominantly random-coil.

Fluorescence microscopy and microspectrofluorimetry of malignant melanomas, naevi and normal melanocytes.

TL;DR: It has been found that the same fluorescence can be observed in formalin-fixed “routine” paraffin sections and it is suggested that fluorescence microscopy of formalin fixed sections is a simple and valuable diagnostic method.
