中国拥有数目庞大、种类繁多的动植物,也有很多珍贵、濒危生物,这些更是异常宝贵的科学资源。DNA条形码技术的出现可以更好的帮助鉴别这些物种,了解其分支来源,甚至可以预知其进化方向。笔者简述了DNA条形码的技术原理与操作步骤及这项技术的产生与发展情况;简要列出了DNA条形码发展过程中的重要进步与研究相对集中的物种;概述了DNA条形码的应用途径及目前DNA条形码研究中存在的主要问题、矛盾、研究思路与发展方向,并对其发展前景做出展望。

On the inappropriate use of Kimura-2-parameter (K2P) divergences in the DNA-barcoding literature.

Nucleotide sequences of a partial cytochrome c oxidase subunit I gene were used to assess the manner in which historical processes and geomorphological effects may have influenced genetic structuring and phylogeographic patterns in Channa striata. Assaying was based on individuals from twelve populations in four river systems, which were separated into two regions, the eastern and western, of the biodiversely rich state of Perak in central Peninsular Malaysia. In 238 specimens, a total of 368-bp sequences with ten polymorphic sites and eleven unique haplotypes were detected. Data on all the twelve populations revealed incomplete divergence due to past historical coalescence and the short period of separation. Nevertheless, SAMOVA and FST revealed geographical structuring existed to a certain extent in both regions. For the eastern region, the data also showed that the upstream populations were genetically significantly different compared to the midand downstream ones. It is inferred that physical barriers and historical processes played a dominant role in structuring the genetic dispersal of the species. A further inference is that the Grik, Tanjung Rambutan and Sungkai are potential candidates for conservation and aquaculture programmes since they contained most of the total diversity in this area.

An annotated checklist of the chondrichthyan fishes (sharks, batoids and chimaeras) of the world is presented. As of 7 November 2015, the number of species totals 1188, comprising 16 orders, 61 families and 199 genera. The checklist includes nine orders, 34 families, 105 genera and 509 species of sharks; six orders, 24 families, 88 genera and 630 species of batoids (skates and rays); one order, three families, six genera and 49 species of holocephalans (chimaeras). The most speciose shark orders are the Carcharhiniformes with 284 species, followed by the Squaliformes with 119. The most species-rich batoid orders are the Rajiformes with 285 species and the Myliobatiformes with 210. This checklist represents the first global checklist of chondrichthyans to include information on maximum size, geographic and depth distributions, as well as comments on taxonomically problematic species and recent and regularly overlooked synonymizations. Furthermore, a detailed analysis of the biogeographical diversity of the species across 10 major areas of occurrence is given, including updated figures for previously published hotspots of chondrichthyan biodiversity, providing the detailed numbers of chondrichthyan species per major area, and revealing centres of distribution for several taxa.

Annotated checklist of the living sharks, batoids and chimaeras (Chondrichthyes) of the world, with a focus on biogeographical diversity

DNA barcoding has been adopted as a global bio-identification system for animals in recent years. A major national programme on DNA barcoding of fish and marine life was initiated in India by the authors during 2006 and 115 species of marine fish covering Carangids, Clupeids, Scombrids, Groupers, Sciaenids, Silverbellies, Mullids, Polynemids and Silurids representing 79 Genera and 37 Families from the Indian Ocean have been barcoded for the first time using cytochrome c oxidase I gene (COI) of the mtDNA. The species were represented by multiple specimens and a total of 397 sequences were generated. After amplification and sequencing of 707 base pair fragment of COI, primers were trimmed which invariably generated a 655 base pair barcode sequence. The average Kimura two parameter (K2P) distances within species, genera, families, orders were 0.30%, 6.60%, 9.91%, 16.00%, respectively. In addition to barcode-based species identification system, phylogenetic relationships among the species have also been attempted. The neighbour-joining tree revealed distinct clusters in concurrence with the taxonomic status of the species.

Endemic fish diversity of Western Ghats

Quercetin (3,3',4',5,7-penthydroxyflavone)-induced biological effects have been beneficial in various disease conditions. In this study, wound healing potential of quercetin was evaluated in a time-dependent manner in open excision wounds in adult Wistar rats. Experimentally-wounded rats were divided into two groups namely, control and quercetin-treated. Wounds were photographed and the area was measured on the day of wounding and on days 3, 7, 11 and 14 post-wounding. The granulation/healing tissue was collected on days 3, 7, 11 and 14 post-wounding for cytokine/growth factor measurements and histology/immunohistochemistry studies. There was significant time-dependent increase in wound closure in quercetin-treated rats. Vascular endothelial growth factor and transforming growth factor-β1 expressions were significantly upregulated in quercetin-treated rats, whereas tumor necrosis factor-α level was markedly reduced. Interleukin- 10 levels and CD31 stained vessels were markedly higher on day 3 and on day 7, respectively, in quercetin-treated rats. In H & E stained sections, quercetin-treated group showed less inflammatory cells, more fibroblast proliferation, increased microvessel density, better reepithelialization and more regular collagen deposition, as compared to control. The results suggest that topical application of quercetin promotes wound healing by effectively modulating the cytokines, growth factors and cells involved in inflammatory and proliferative phases of healing.

Conservation, management and sustainable utilisation of biological resources depend on the accurate identification of exploited taxa, which emphasises the need for systematic taxonomic research. Chondrichthyans (sharks, rays, skates and chimaeras) are considered to be one of the most vulnerable exploited marine resources, however, the basic taxonomic study of these groups in Indian waters needs improvement to achieve better management for their sustainable exploitation. We discuss issues concerning chondrichthyan taxonomic research in India and provide an extended, updated checklist of chondrichthyans listed/reported from Indian waters, together with comments on their occurrence.
