Phycobilisomes serve as the primary light-harvesting antennae for Photosystem II in cyanobacteria and red algae. These supramolecular complexes are primarily composed of phycobiliproteins, a brilliantly colored family of water-soluble proteins bearing covalently attached, open-chain tetrapyrroles known as phycobilins. In addition, phycobilisomes also contain smaller amounts ‘linker polypeptides,’ most of which do not bear chromophores. These components are absolutely required for proper assembly and functional organization of the structure. Phycobilisomes are constructed from two main structural elements: a core substructure and peripheral rods that are arranged in a hemidiscoidal fashion around that core. The core of most hemidiscoidal phycobilisomes is composed of three cylindrical subassemblies. The peripheral rods radiate from the lateral surfaces of the core substructure which are not in contact with the thylakoid membrane. Absorbed light energy is transferred by very rapid, radiation-less downhill energy transfer from phycoerythrin or phycoerythrocyanin (if present) to C-phycocyanin and then to allophycocyanin species that act as the final energy transmitters from the phycobilisome to the Photosystem II or (partially) Photosystem I reaction centers.

Phycobilisome and Phycobiliprotein Structures

The molecular architectures of photosynthetic complexes are rapidly becoming available through the power of X-ray crystallography. These complexes are comprised of antenna complexes, which absorb and transfer energy into photochemical reaction centers. Most reaction centers, found in both oxygenic and non-oxygenic species, are connected to transmembrane chlorophyll containing antennas, and the crystal structures of these antennas contain information on the structure of the entire complex as well as clear indications on their modes of functional association. In cyanobacteria and red alga, most of the Photosystem II associated light harvesting is performed by an enormous (3-7 MDa) membrane attached complex called the phycobilisome (PBS). While the crystal structures of many isolated components of different PBSs have been determined, the structure of the entire complex as well as its manner of association with Photosystem II can only be suggested. In this review, the structural information obtained on the isolated components will be described. The structural information obtained from the components provides the basis for the modeled reconstruction of this giant complex.

Elucidation of the molecular structures of components of the phycobilisome: reconstructing a giant.

Post-translational modification of proteins.

Methylation is a post-translational modification that can affect numerous features of proteins, notably cellular localization, turnover, activity, and molecular interactions. Recent genome-wide analyses have considerably extended the list of human genes encoding putative methyltransferases. Studies on protein methyltransferases have revealed that the regulatory function of methylation is not limited to epigenetics, with many non-histone substrates now being discovered. We present here our findings on a novel family of distantly related putative methyltransferases. Affinity purification coupled to mass spectrometry shows a marked preference for these proteins to associate with various chaperones. Based on the spectral data, we were able to identify methylation sites in substrates, notably trimethylation of K135 of KIN/Kin17, K561 of HSPA8/Hsc70 as well as corresponding lysine residues in other Hsp70 isoforms, and K315 of VCP/p97. All modification sites were subsequently confirmed in vitro. In the case of VCP, methylation by METTL21D was stimulated by the addition of the UBX cofactor ASPSCR1, which we show directly interacts with the methyltransferase. This stimulatory effect was lost when we used VCP mutants (R155H, R159G, and R191Q) known to cause Inclusion Body Myopathy with Paget's disease of bone and Fronto-temporal Dementia (IBMPFD) and/or familial Amyotrophic Lateral Sclerosis (ALS). Lysine 315 falls in proximity to the Walker B motif of VCP's first ATPase/D1 domain. Our results indicate that methylation of this site negatively impacts its ATPase activity. Overall, this report uncovers a new role for protein methylation as a regulatory pathway for molecular chaperones and defines a novel regulatory mechanism for the chaperone VCP, whose deregulation is causative of degenerative neuromuscular diseases.

A Newly Uncovered Group of Distantly Related Lysine Methyltransferases Preferentially Interact with Molecular Chaperones to Regulate Their Activity

Cryptophytes are unicellular photosynthetic algae that use a lumenally located light-harvesting system, which is distinct from the phycobilisome structure found in cyanobacteria and red algae. One of the key components of this system is water-soluble phycoerythrin (PE) 545 whose expression is enhanced by low light levels. The crystal structure of the heterodimeric α1α2ββ PE 545 from the marine cryptophyte Rhodomonas CS24 has been determined at 1.63-Å resolution. Although the β-chain structure is similar to the α and β chains of other known phycobiliproteins, the overall structure of PE 545 is novel with the α chains forming a simple extended fold with an antiparallel β-ribbon followed by an α-helix. The two doubly linked β50/β61 chromophores (one on each β subunit) are in van der Waals contact, suggesting that exciton-coupling mechanisms may alter their spectral properties. Each α subunit carries a covalently linked 15,16-dihydrobiliverdin chromophore that is likely to be the final energy acceptor. The architecture of the heterodimer suggests that PE 545 may dock to an acceptor protein via a deep cleft and that energy may be transferred via this intermediary protein to the reaction center.
