Nature relies on large molecules to carry out sophisticated chemical operations, such as catalysis, tight and specific binding, directed flow of electrons, or controlled crystallization of inorganic phases. The polymers entrusted with these crucial tasks, mostly proteins but sometimes RNA, are unique relative to other biological and synthetic polymers in that they adopt specific compact conformations that are thermodynamically and kinetically stable. These folding patterns generate “active sites” via precise three-dimensional arrangement of functional groups. In terms of covalent connectivity, the groups that comprise the active site are often widely spaced along the polymer backbone. The remarkable range of chemical capabilities that evolution has elicited from proteins suggests that it might be possible to design analogous capabilities into unnatural polymers that fold into compact and specific conformations. Since biological evolution has operated under many constraints, the functional properties of proteins and RNA should be viewed as merely exemplifying the potential of compactly folded polymers. The chemist’s domain includes all possible combinations of the elements, and the biological realm, vast and complex though it may be, is only a small part of that domain. Therefore, realization of the potential of folding polymers may be limited more by the human imagination than by physical barriers. I use the term “foldamer” to describe any polymer with a strong tendency to adopt a specific compact conformation. Among proteins, the term “compact” is associated with tertiary structure, and there is as yet no synthetic polymer that displays a specific tertiary structure. Protein tertiary structure arises from the assembly of elements of regular secondary structure (helices, sheets, and turns). The first step in foldamer design must therefore be to identify new backbones with well-defined secondary structural preferences. “Well-defined” in this case means that the conformational preference should be displayed in solution by oligomers of modest length, and I will designate as a foldamer any oligomer that meets this criterion. Within the past decade, a handful of research groups have described unnatural oligomers with interesting conformational propensities. The motivations behind such efforts are varied, but these studies suggest a collective, emerging realization that control over oligomer and polymer folding could lead to new types of molecules with useful properties. The purpose of this “manifesto” is to introduce a large audience to the broad research horizons offered by the concept of synthetic foldamers. The path to creating useful foldamers involves several daunting steps. (i) One must identify new polymeric backbones with suitable folding propensities. This goal includes developing a predictively useful understanding of the relationship between the repetitive features of monomer structure and conformational properties at the polymer level. (ii) One must endow the resulting foldamers with interesting chemical functions, by design, by randomization and screening (“evolution”), or by some combination of these two approaches. (iii) For technological utility, one must be able to produce a foldamer efficiently, which will generally include preparation of the constituent monomers in stereochemically pure form and optimization of heteropolymer synthesis. Each of these steps involves fascinating chemical challenges; the first step is the focus of this Account.

Helical polymers: synthesis, structures, and functions

A simple technique for precisely controlling the interfacial energies and wetting behavior of polymers in contact with solid surfaces is described. End-functionalized statistical random copolymers of styrene and methylmethacrylate were synthesized, with the styrene fraction f varying from 0 to 1, and were end-grafted onto silicon substrates to create random copolymer brushes about 5 nanometers thick. For f < 0.7, polystyrene (PS) films (20 nanometers thick) rapidly dewet from the brushes when heated well above the glass transition temperature. The contact angle of the resulting polymer droplets increased monotonically with decreasing f . Similar behavior was observed for poly(methylmethacrylate) (PMMA) films but with an opposite dependence on f . The interfacial energies of the random copolymer brushes with PS and PMMA were equal when f was about 0.6. Thus, precise control of the relative surface affinities of PS and PMMA was possible, demonstrating a way to manipulate polymer-surface interactions.

Controlling Polymer-Surface Interactions with Random Copolymer Brushes

In this review, we describe the recent advances in supramolecular helical assemblies formed from chiral and achiral small molecules, oligomers (foldamers), and helical and nonhelical polymers from the viewpoints of their formations with unique chiral phenomena, such as amplification of chirality during the dynamic helically assembled processes, properties, and specific functionalities, some of which have not been observed in or achieved by biological systems. In addition, a brief historical overview of the helical assemblies of small molecules and remarkable progress in the synthesis of single-stranded and multistranded helical foldamers and polymers, their properties, structures, and functions, mainly since 2009, will also be described.

Supramolecular Helical Systems: Helical Assemblies of Small Molecules, Foldamers, and Polymers with Chiral Amplification and Their Functions

Synthetic helical polymers: conformation and function.

Polyisocyanates, long studied as theoretical models for wormlike chains in dilute solution and liquid crystals, differ from their biological helical analogs in the absence of a pre-determined helical sense. These polymers have an unusual sensitivity to chiral effects that arises from a structure in which alternating right- and left-handed long helical blocks are separated by infrequent and mobile helical reversals. Statistical thermodynamic methods yield an exact description of the polymer and the cooperative nature of its chiral properties. Minute energies that favor one of the helical senses drive easily measurable conformational changes, even though such energies may be extremely difficult to calculate from structural theory. In addition, the chiral nature of the polymer can be used to test theoretical ideas concerned with cholesteric liquid crystals, one of which solves the problem of assigning the helical sense.

A Helical Polymer with a Cooperative Response to Chiral Information

This article reviews the following solution properties of liquid-crystalline stiff-chain polymers: (1) osmotic pressure and osmotic compressibility, (2) phase behavior involving liquid crystal phase(s), (3) orientational order parameter, (4) translational and rotational diffusion coefficients, (5) zero-shear viscosity, and (6) rheological behavior in the liquid crystal state. Among the related theories, the scaled particle theory is chosen to compare with experimental results for properties (1)–(3), the fuzzy cylinder model theory for properties (4) and (5), and Doi's theory for property (6). In most cases the agreement between experiment and theory is satisfactory, enabling one to predict solution properties from basic molecular parameters. Procedures for data analysis are described in detail.

The conformation of amylose in DMSO at 25°C is studied by light scattering, sedimentation equilibrium, and viscosity measurements on narrow-distribution samples with molecular weight M w from 342 (the dimer) to 1.7×10 6 . For M w above 10 5 , the z-average mean-square radius of gyration x and the intrinsic viscosity [η] vary as M w 1.2 and M w 0.7 , respectively, indicating that in this molecular weight region, the amylose chain behaves as a random coil expanded by excluded-volume effect. Below M w ∼ 10 4 , [η] exhibits an unusually weak dependence on M w and finally becomes almost independent of M w
