A new assay for quantifying endo-β-d-mannanase activity using congo red dye
TL;DR: The gel-diffusion assay for the quantification of endo-β- d -mannanase (EC 3.2.1.78) activity was used to quantify the enzyme activity present in seeds, fruit, bulbs and fungi; repeatability and sensitivity were excellent.
Journal ArticleDOI
Primary dormancy in tomato (Lycopersicon esculentum cv. Moneymaker): studies with the sitiens mutant
Henk W. M. Hilhorst,Bruce Downie +1 more
TL;DR: The ability of intact, ABA-deficient sit w mutant tomato seeds to complete germination at much lower external osmotic conditions relative to the intact wild-type Moneymaker cultivar was solely dependent on a structural alteration in the mutant testa, making it more delicate and lesssening its resistance to penetration by the radicle.
Journal ArticleDOI
Multiple isozymes of endo-β-D-mannanase in dry and imbibed seeds
TL;DR: A gel overlay method for detecting isozymes of endo-β- d -mannanase separated by isoelectric focusing has been developed for use with plant extracts and found that more isoz enzymes were present in the endosperm/megagametophyte than in the embryo regardless of whether the seed was intact or separated.
Journal ArticleDOI
Upgrading germinability and vigour of jack pine, lodgepole pine, and white spruce by the IDS technique
Bruce Downie,B. S. P. Wang +1 more
TL;DR: The IDS (incubation–desiccation–separation) technique, designed to upgrade germinability and vigour of conifer seed lots, was tested on various seed lots of white spruce, lodgepole pine, and jack pine, finding its effect was not predictable.
Journal ArticleDOI
Dormancy in white spruce (Picea glauca [Moench.] Voss.) seeds is imposed by tissues surrounding the embryo
Bruce Downie,J. Derek Bewley +1 more
TL;DR: The germination of intact, moist-chilled white spruce (Picea glauca [Moench.] Voss.) seeds and liquid-nitrogendecoated (LN2), unchilled seeds is significantly more than of intact and unchilled seed exposed to LN2.
