This paper describes the fabrication and the performance of microfluidic paper-based electrochemical sensing devices (we call the microfluidic paper-based electrochemical devices, µPEDs). The µPEDs comprise paper-based microfluidic channels patterned by photolithography or wax printing, and electrodes screen-printed from conducting inks (e.g., carbon or Ag/AgCl). We demonstrated that the µPEDs are capable of quantifying the concentrations of various analytes (e.g., heavy-metal ions and glucose) in aqueous solutions. This low-cost analytical device should be useful for applications in public health, environmental monitoring, and the developing world.

Electrochemical sensing in paper-based microfluidic devices

Unprecedented bioaccumulation and biomagnification of heavy metals (HMs) in the environment have become a dilemma for all living organisms including plants. HMs at toxic levels have the capability to interact with several vital cellular biomolecules such as nuclear proteins and DNA, leading to excessive augmentation of reactive oxygen species (ROS). This would inflict serious morphological, metabolic, and physiological anomalies in plants ranging from chlorosis of shoot to lipid peroxidation and protein degradation. In response, plants are equipped with a repertoire of mechanisms to counteract heavy metal (HM) toxicity. The key elements of these are chelating metals by forming phytochelatins (PCs) or metallothioneins (MTs) metal complex at the intra- and intercellular level, which is followed by the removal of HM ions from sensitive sites or vacuolar sequestration of ligand-metal complex. Nonenzymatically synthesized compounds such as proline (Pro) are able to strengthen metal-detoxification capacity of intracellular antioxidant enzymes. Another important additive component of plant defense system is symbiotic association with arbuscular mycorrhizal (AM) fungi. AM can effectively immobilize HMs and reduce their uptake by host plants via binding metal ions to hyphal cell wall and excreting several extracellular biomolecules. Additionally, AM fungi can enhance activities of antioxidant defense machinery of plants.

Plants are the target of a wide range of pollutants that vary in concentration, speciation, and toxicity. Such pollutants mainly enter the plant system through the soil (Arshad et al. 2008) or via the atmosphere (Uzu et al. 2010). Among common pollutants that affect plants, lead is among the most toxic and frequently encountered (Cecchi et al. 2008; Grover et al. 2010; Shahid et al. 2011). Lead continues to be used widely in many industrial processes and occurs as a contaminant in all environmental compartments (soils, water, the atmosphere, and living organisms). The prominence of environmental lead contamination results both from its persistence (Islam et al. 2008; Andra et al. 2009; Punamiya et al. 2010) and from its present and past numerous sources. These sources have included smelting, combustion of leaded gasoline, or applications of lead-contaminated media (sewage sludge and fertilizers) to land (Piotrowska et al. 2009; Gupta et al. 2009; Sammut et al. 2010; Grover et al. 2010). In 2009, production of recoverable lead from mining operations was 1690, 516, and 400 thousand metric tons by China, Australia, and the USA, respectively (USGS 2009).

Lead Uptake, Toxicity, and Detoxification in Plants

Certain five heavy metals viz. arsenic (As), cadmium (Cd), chromium (Cr)(VI), mercury (Hg), and lead (Pb) are non-threshold toxins and can exert toxic effects at very low concentrations. These heavy metals are known as most problematic heavy metals and as toxic heavy metals (THMs). Several industrial activities and some natural processes are responsible for their high contamination in the environment. In recent years, high concentrations of heavy metals in different natural systems including atmosphere, pedosphere, hydrosphere, and biosphere have become a global issue. These THMs have severe deteriorating effects on various microorganisms, plants, and animals. Human exposure to the THMs may evoke serious health injuries and impairments in the body, and even certain extremities can cause death. In all these perspectives, this review provides a comprehensive account of the relative impact of the THMs As, Cd, Cr(VI), Hg, and Pb on our total environment.

This review paper is focused predominantly on the role of the cell wall in the defense response of plants to trace metals. It is generally known that this compartment accumulates toxic divalent and trivalent metal cations both during their uptake by the cell from the environment and at the final stage of their sequestration from the protoplast. However, from results obtained in recent years, our understanding of the role played by the cell wall in plant defense response to toxic metals has markedly altered. It has been shown that this compartment may function not only as a sink for toxic trace metal accumulation, but that it is also actively modified under trace metal stress. These modifications lead to an increase in the capacity of the cell wall to accumulate trace metals and a decrease of its permeability for trace metal migration into the protoplast. One of the most striking alterations is the enhancement of the level of low-methylesterified pectins: the polysaccharides able to bind divalent and trivalent metal ions. This review paper will present the most recent results, especially those that are concerned with polysaccharide level, composition and distribution under trace metal stress, and describe in detail the polysaccharides responsible for metal binding and immobilization in different groups of plants (algae and higher plants). The review also contains information related to the entry pathways of trace metals into the cell wall and their detection methods.
