Chemistry with [18F]Fluoride Ion

Tuberculosis is the leading cause of infectious disease mortality in the world by a bacterial pathogen. We previously demonstrated that a bisubstrate inhibitor of the adenylation enzyme MbtA, which is responsible for the second step of mycobactin biosynthesis, exhibited potent antitubercular activity. Here we systematically investigate the structure−activity relationships of the bisubstrate inhibitor glycosyl domain resulting in the identification of a carbocyclic analogue that possesses a KIapp value of 2.3 nM and MIC99 values of 1.56 μM against M. tuberculosis H37Rv. The SAR data suggest the intriguing possibility that the bisubstrate inhibitors utilize a transporter for entry across the mycobacterial cell envelope. Additionally, we report improved conditions for the expression of MbtA and biochemical analysis, demonstrating that MbtA follows a random sequential enzyme mechanism for the adenylation half-reaction.

Conformationally restricted, orthogonally protected 2,4-diaminocarboxylates with a cyclopentane skeleton were efficiently synthesized from β-lactam 6, the syntheses involving strategies of diastereoselective epoxidation of the β-lactam and the corresponding monoprotected amino esters with opposite selectivities followed by regioselective opening of the oxirane ring with sodium azide. The enantiomers were also prepared. This new class of compounds can be regarded not only as conformationally constrained β,γ-diamino acid derivatives but also as potential functionalized carbocyclic nucleoside precursors.

Diastereo- and Enantioselective Synthesis of Orthogonally Protected 2,4-Diaminocyclopentanecarboxylates: A Flip from β-Amino- to β,γ-Diaminocarboxylates

The present review focusses on the recent progress in the reductive grafting of diazotized big molecules and polymers on carbon nanotubes (CNTs). We briefly summarize the essentials on diazonium synthesis and discuss the CNT physical properties. The different routes for CNT covalent functionalization through diazonium salt interface chemistry are reviewed. The main analytical and spectroscopic techniques used to track carbon nanotube surface modification were cited as well as their advantages and limitations in the light of the information they provide. In this review, the emphasis is on big molecules such as dyes, crown ether, calixarene, cyclodextrin, fullerene and Ru-complex, and biomolecules such as biotin, proteins, and antibodies. The attachment of synthetic polymers to CNT via diazonium chemistry, or by preparing CNT-Polymer nanocomposites through: (1) in situ polymerization (controlled radical polymerization and click chemistry). (2) Oxidative polymerization of conjugated monomers. (3) Grafting onto method by Huisgen 1,3-cycloaddition click reaction and epoxy ring-opening were summarized and discussed. Throughout this review, we reported the recent advances using diazonium salt chemistry in numerous research areas (biomedicine, environment, energy conversion, sensors and actuators, structural composites,…).

Depending on the amount of acid used, treating aryl–dialkyl triazenes of general structure 3 with triflic acid resulted in the formation of either the corresponding arenediazonium triflates 4 or aryl triflates 8 apparently by two different pathways, the latter conversion being favoured at high acid concentration.
