An increasing problem in the field of health protection is the emergence of drug-resistant and multi-drug-resistant bacterial strains. They cause a number of infections, including hospital infections, which currently available antibiotics are unable to fight. Therefore, many studies are devoted to the search for new therapeutic agents with bactericidal and bacteriostatic properties. One of the latest concepts is to search for this type of substances among toxins produced by venomous animals. In this approach, however, special attention is paid to snake venom because it contains molecules with antibacterial properties. Thorough investigations have shown that the phospholipases A2 (PLA2) and l-amino acids oxidases (LAAO), as well as fragments of these enzymes, are mainly responsible for the bactericidal properties of snake venoms. Some preliminary research studies also suggest that fragments of three-finger toxins (3FTx) are bactericidal. It has also been proven that some snakes produce antibacterial peptides (AMP) homologous to human defensins and cathelicidins. The presence of these proteins and peptides means that snake venoms continue to be an interesting material for researchers and can be perceived as a promising source of antibacterial agents.

Venoms are a complex cocktail of biologically active molecules, including peptides, proteins, polyamide, and enzymes widely produced by venomous organisms. Through long-term evolution, venomous animals have evolved highly specific and diversified peptides and proteins targeting key physiological elements, including the nervous, blood, and muscular systems. Centipedes are typical venomous arthropods that rely on their toxins primarily for predation and defense. Although centipede bites are frequently reported, the composition and effect of centipede venoms are far from known. With the development of molecular biology and structural biology, the research on centipede venoms, especially peptides and proteins, has been deepened. Therefore, we summarize partial progress on the exploration of the bioactive peptides and proteins in centipede venoms and their potential value in pharmacological research and new drug development.

Bioactive Peptides and Proteins from Centipede Venoms

We developed a bacterial expression system to produce a recombinant disintegrin, vicrostatin (VCN), whose structure is based on a natural disintegrin isolated from southern copperhead snake venom. Our goal is to develop VCN for potential clinical translation as an anti-cancer agent. VCN is a peptide of 69 amino acids with a single tyrosine residue. We have employed VCN as integrin-targeted radionuclide therapy (brachytherapy) for treatment of glioblastoma (GBM, glioma). GBM is a deadly brain cancer that doesn’t discriminate between sexes and knows no age limit. We established that the tyrosine residue in VCN can be radioiodinated with full retention of bioactivity. 131I-VCN was utilized for integrin-targeted radionuclide therapy using mouse models of glioma. The combination of radioiodinated VCN plus temozolomide (a DNA alkylating agent) significantly prolonged survival of glioma-bearing mice. We also obtained similar results using an immunocompetent mouse model and a murine glioma cell line. In summary, as demonstrated in studies reported here we have shown that VCN as targeted radionuclide therapy for GBM has significant translational potential for therapy of this deadly disease.

A New Indole Glucoside and Other Constituents from the Sea Cucumber-Derived Aspergillus fumigatus M580 and Their Antimicrobial, Cytotoxic, and Inhibitory α-Glucosidase Activities

Microorganisms are of particular interest because of their ability to synthesize high-value secondary compounds and provide us with novel and diverse chemical structures. The most common source of antibiotics is Actinomycetes which provide around two-third of naturally occurring antibiotics, including many of medical importance. In this study, 81 strains of actinomycetes were isolated from 145 samples including: sediments, sponges, soft corals, echinoderms and starfish collected from three sea areas of Vietnam: Hue, Da Nang and Quang Nam. The strains were fermented in A+ medium and fermentation broths were extracted 5 times with ethyl acetate. The extracts were evaporated under reduced  pressure to yield crude extracts. Quantitative assay was used to determine MIC (Minimum inhibitory concentration) of extract against 7 reference strains. From the results of screening, Seven strains of actinomycetes that have the highest biological activity (Code: G244, G246, G261, G266, G278, G280 and G290) were chosen to be identified by morphological and phylogenetic based on 16S rRNA gene sequences. The results showed that 6 strains G246, G261, G266, G278, G280 and G290 belonged to the genus Streptomyces; and the strain G244 belonged to the genus Micromonospora. In particular, strains G244, G278, G280 were resistant 5/7 strains of microorganisms test, with values  MICs from 2 µg/mL to 256 µg/mL; and three strains G261, G266, G290 showed the inhibitory effect towards 4/7 strains of microorganisms test, with respective values MICs from 2 µg/mL to 256 µg/mL. Moreover, six of the seven selected strains were highly resistant to yeast Candida albicans ATCC10231 with MIC values from 2 µg/mL to 256 µg/mL. These results indicated that marine Actinomycetes in Vietnam are also a potential source to find bioactive substances.
