Fluorescent molecular rotors (FMRs) can act as viscosity sensors in various media including subcellular organelles and microfluidic channels. In FMRs, the rotation of rotators connected to a fluorescent π-conjugated bridge is suppressed by increasing environmental viscosity, resulting in increasing fluorescence (FL) intensity. In this minireview, we describe recently developed FMRs including push-pull type π-conjugated chromophores, meso-phenyl (borondipyrromethene) (BODIPY) derivatives, dioxaborine derivatives, cyanine derivatives, and porphyrin derivatives whose FL mechanism is viscosity-responsive. In addition, FMR design strategies for addressing various issues (e.g., obtaining high FL contrast, internal FL references, and FL intensity-contrast trade-off) and their biological and microfluidic applications are also discussed.

Fluorescent Molecular Rotors for Viscosity Sensors

For deeper understanding of the coupling of electronic processes with conformational motions, we exploit a tailored strategy to harness the excited-state planarization of N,N′-disubstituted dihydrodibenzo[a,c]phenazines by halting the structural evolution via a macrocyclization process. In this new approach, 9,14-diphenyl-9,14-dihydrodibenzo[a,c]phenazine (DPAC) is used as a prototype, in which the para sites of 9,14-diphenyl are systematically enclosed by a dialkoxybenzene-alkyl-ester or -ether linkage with different chain lengths, imposing various degrees of constraint to impede the structural deformation. Accordingly, a series of DPAC-n (n = 1–8) derivatives were synthesized, in which n correlates with the alkyl length, such that the strength of the spatial constraint decreases as n increases. The structures of DPAC-1, DPAC-3, DPAC-4, and DPAC-8 were identified by the X-ray crystal analysis. Snapshotting the Excited-State Planarization of Chemically Locked N,N′-Disubstituted Dihydrodibenzo[a,c]phenazines

Millirobots that can adapt to unstructured environments, operate in confined spaces, and interact with a diverse range of objects would be desirable for exploration and biomedical applications. The continued development of millirobots, however, requires simple and scalable fabrication techniques. Here, we propose a minimalist approach to construct millirobots by coating inanimate objects with a composited agglutinate magnetic spray. Our approach enables a variety of one-dimensional (1D), 2D, or 3D objects to be covered with a thin magnetically drivable film (~100 to 250 micrometers in thickness). The film is thin enough to preserve the original size, morphology, and structure of the objects while providing actuation of up to hundreds of times its own weight. Under the actuation of a magnetic field, our millirobots are able to demonstrate a range of locomotive abilities: crawling, walking, and rolling. Moreover, we can reprogram and disintegrate the magnetic film on our millirobots on demand. We leverage these abilities to demonstrate biomedical applications, including catheter navigation and drug delivery.

Switchable Adhesives for Multifunctional Interfaces

Luminogens with aggregation-induced emission (AIE) characteristics are nowadays undergoing explosive development in the fields of imaging, process visualization, diagnosis and therapy. However, exploration of an AIE luminogen (AIEgen) system allowing for extremely wide color tunability remains challenging. In this contribution, the facile synthesis of triphenylamine (TPA)–thiophene building block-based AIEgens having tunable maximum emission wavelengths covering violet, blue, green, yellow, orange, red, deep red and NIR regions is reported. The obtained AIEgens can be utilized as extraordinary fluorescent probes for lipid droplet (LD)-specific cell imaging and cell fusion assessment, showing excellent image contrast to the cell background and high photostability, as well as satisfactory visualization outcomes. Interestingly, quantitative evaluation of the phototherapy effect demonstrates that one of these presented AIEgens, namely TTNIR, performs well as a photosensitizer for photodynamic ablation of cancer cells upon white light irradiation. This study thus provides useful insights into rational design of fluorescence systems for widely tuning emission colors with high brightness, and remarkably extends the applications of AIEgens.

Facile synthesis of AIEgens with wide color tunability for cellular imaging and therapy

Liquid crystal (LC) provides a suitable platform to exploit structural motions of molecules in a condensed phase. Amplification of the structural changes enables a variety of technologies not only in LC displays but also in other applications. Until very recently, however, a practical use of LCs for removable adhesives has not been explored, although a spontaneous disorganization of LC materials can be easily triggered by light-induced isomerization of photoactive components. The difficulty of such application derives from the requirements for simultaneous implementation of sufficient bonding strength and its rapid disappearance by photoirradiation. Here we report a dynamic molecular LC material that meets these requirements. Columnar-stacked V-shaped carbon frameworks display sufficient bonding strength even during heating conditions, while its bonding ability is immediately lost by a light-induced self-melting function. The light-melt adhesive is reusable and its fluorescence colour reversibly changes during the cycle, visualizing the bonding/nonbonding phases of the adhesive.

Light-melt adhesive based on dynamic carbon frameworks in a columnar liquid-crystal phase

A variety of fluorescent molecular viscosity probes have been widely used for mapping the local viscosity in cells and for monitoring the microenvironments in materials. However, their viscosity-sensing structural design still relies strongly on molecular rotors featuring intramolecular rotational dynamics. Here we report flapping molecules (FLAP) as a ratiometric viscosity-sensing fluorophore that shows polarity-independent dual fluorescence. Viscosity-sensing mechanism is based on a unique V-shaped-to-planar conformational change in the singlet excited state (S1), in which the flexible motion of an eight-membered ring plays an important role. Fast conformational dynamics have been studied by time-resolved spectroscopies, and the viscochromic properties have been quantitatively analyzed. Application of FLAP to monitoring the curing process of epoxy resins has also been demonstrated, in which other typical environment-sensitive dyes did not work as a local viscosity probe.
