Journal ArticleDOI
Systemic RNA delivery to dendritic cells exploits antiviral defence for cancer immunotherapy
Lena M. Kranz,Mustafa Diken,Heinrich Haas,Sebastian Kreiter,Carmen Loquai,Kerstin C. Reuter,Martin Meng,Daniel Fritz,Fulvia Vascotto,Hossam Hefesha,Christian Grunwitz,Mathias Vormehr,Yves Hüsemann,Abderraouf Selmi,Andreas Kuhn,Janina Buck,Evelyna Derhovanessian,Richard Rae,Sebastian Attig,Jan Diekmann,Robert A. Jabulowsky,Sandra Heesch,Jessica C. Hassel,Peter Langguth,Stephan Grabbe,Christoph Huber,Özlem Türeci,Ugur Sahin +27 more
TL;DR: It is shown that DCs can be targeted precisely and effectively in vivo using intravenously administered RNA-lipoplexes (RNA-LPX) based on well-known lipid carriers by optimally adjusting net charge, without the need for functionalization of particles with molecular ligands.
Journal ArticleDOI
Local delivery of mRNA-encoded cytokines promotes antitumor immunity and tumor eradication across multiple preclinical tumor models.
Christian Hotz,Timothy R. Wagenaar,Friederike Gieseke,Dinesh S. Bangari,Michelle Callahan,Hui Cao,Jan Diekmann,Mustafa Diken,Christian Grunwitz,Andy Hebert,Karl Hsu,Marie Bernardo,Katalin Karikó,Sebastian Kreiter,Andreas Kuhn,Mikhail Levit,Natalia Malkova,Serena Masciari,Jack Pollard,Hui Qu,Sue Ryan,Abderaouf Selmi,Julia Schlereth,Kuldeep Singh,Fangxian Sun,Bodo Tillmann,Tatiana Tolstykh,William Weber,Lena Wicke,Sonja Witzel,Qunyan Yu,Yu-an Zhang,Gang Zheng,Joanne Lager,Gary J. Nabel,Ugur Sahin,Dmitri Wiederschain +36 more
TL;DR: Local immunotherapy ideally stimulates immune responses against tumors while avoiding toxicities associated with systemic administration as mentioned in this paper. But, current strategies for tumor-targeted, gene-based delivery are limited and ineffective.
Journal ArticleDOI
HPV16 RNA-LPX vaccine mediates complete regression of aggressively growing HPV-positive mouse tumors and establishes protective T cell memory.
Christian Grunwitz,Nadja Salomon,Fulvia Vascotto,Abderaouf Selmi,Thomas Bukur,Mustafa Diken,Sebastian Kreiter,Özlem Türeci,Ugur Sahin +8 more
TL;DR: It is shown in mice that HPV16 E7 RNA-LPX, an intravenously administered cancer vaccine based on immuno-pharmacologically optimized antigen-encoding mRNA, efficiently primes and expands antigen-specific effector and memory CD8+ T cells and mediates complete and durable remission of progressing tumors.
Journal ArticleDOI
Selective Glucocorticoid Receptor Agonists for the Treatment of Inflammatory Bowel Disease: Studies in Mice with Acute Trinitrobenzene Sulfonic Acid Colitis
Kerstin C. Reuter,Christian Grunwitz,Bettina M. Kaminski,Dieter Steinhilber,Heinfried H. Radeke,Jürgen Stein +5 more
TL;DR: Although CpdA revealed a narrow therapeutic window, both GR agonists might be seen as a starting point for a future IBD treatment option and both SEGRAs acted as potent anti-inflammatory agents with a significantly improved profile compared with classic GCs.
Book ChapterDOI
mRNA Cancer Vaccines—Messages that Prevail
Christian Grunwitz,Lena M. Kranz +1 more
TL;DR: This review summarizes the principles of mRNA vaccines and highlights the importance of in vivo mRNA delivery and recent advances in harnessing their therapeutic potential.
