Detection of total and hemolysin-producing Vibrio parahaemolyticus in shellfish using multiplex PCR amplification of tl, tdh and trh
Asim K. Bej,Donald P. Patterson,Cynthia W. Brasher,Michael C.L. Vickery,Daniel D. Jones,Charles A. Kaysner +5 more
TL;DR: The multiplex PCR approach was successfully used to detect various strains of V parahaemolyticus in seeded oyster tissue homogenate, and this high level of sensitivity of detection within 8 h of pre-enrichment is well within the action level suggested by the National Seafood Sanitation Program guideline.
Journal ArticleDOI
Detection of Microbial Pathogens in Shellfish with Multiplex PCR
TL;DR: Multiplex PCR amplification, followed by colorimetric GeneComb™ DNA-DNA hybridization, has been shown to be an effective, sensitive, and rapid method to detect microbial pathogens in shellfish.
Evaluation of PCR Amplification-based Detection of Heat-killed Escherichia coli and Cell-free DNA in Shellfish
TL;DR: DNA-PCR detection was positive for uidA using the Chelex™ 100 DNA extraction method on oyster samples with high concentrations of cell-free DNA or heat-killed cells, suggesting that a difference in DNA extraction techniques can eliminate false-positive results by PCR, thus making it a reliable and rapid diagnostic tool to differentiate viable and non-viable cells in shellfish samples.
