Relaxation time measurements at 3.0 T are reported for both gray and white matter in normal human brain. Measurements were made using a 3.0 T Bruker Biospec magnetic resonance imaging (MRI) scanner in normal adults with no clinical evidence of neurological disease. Nineteen subjects, 8 female and 11 male, were studied for T1 and T2 measurements, and 7 males were studied for T2. Measurements were made using a saturation recovery method for T1, a multiple spin-echo experiment for T2, and a fast low-angle shot (FLASH) sequence with 14 different echo times for T2. Results of the measurements are summarized as follows. Average T1 values measured for gray matter and white matter were 1331 and 832 msec, respectively. Average T2 values measured for gray matter and white matter were 80 and 110 msec, respectively. The average T2 values for occipital and frontal gray matter were 41.6 and 51.8 msec, respectively. Average T2 values for occipital and frontal white matter were 48.4 and 44.7 msec, respectively. ANOVA tests of the measurements revealed that for both gray and white matter there were no significant differences in T1 from one location in the brain to another. T2 in occipital gray matter was significantly higher (0.0001 < P < .0375) than the rest of the gray matter, while T2 in frontal white matter was significantly lower (P < 0.0001). Statistical analysis of cerebral hemispheric differences in relaxation time measurements showed no significant differences in T1 values from the left hemisphere compared with the right, except in insular gray matter, where this difference was significant at P = 0.0320. No significant difference in T2 values existed between the left and right cerebral hemispheres. Significant differences were apparent between male and female relaxation time measurements in brain.

NMR field-cycling spectroscopy: principles and a]lications

Since 1/T2 of protons of tissue water is generally much greater than 1/T1 at typical imaging fields, small single-ion contrast agents--such as Gd(DTPA), which make comparable incremental contributions and therefore smaller fractional contributions to 1/T2 compared to 1/T1--are not as desirable for contrast-enhancement as agents that could enhance 1/T2 preferentially. In principle, such specialized agents will only be effective at higher fields because the field dependence (dispersion) of 1/T1 is such that it approaches zero at high fields whereas 1/T2 approaches a constant value. The residual 1/T2 is called the "secular" contribution and arises from fluctuations in time--as sensed by the protons of diffusing solvent or tissue water molecules--of the component of the magnetic field parallel to the static applied field. For solutions or suspensions of sufficiently large paramagnetic or ferromagnetic particles (greater than or equal to 250 A diameter), the paramagnetic contributions to the relaxation rates satisfy 1/T2 much greater than 1/T1 at typical imaging fields. We examine the theory of secular relaxation in some detail, particularly as it applies to systems relevant to magnetic resonance imaging, and then analyze the data for solutions, suspensions, or tissue containing ferritin, erythrocytes, agar-bound magnetite particles, and liver with low-density composite polymer-coated magnetite. In most cases we can explain the relaxation data, often quantitatively, in terms of the theory of relaxation of protons (water molecules) diffusing in the outer sphere environments of magnetized particles. The dipolar field produced by these particles has a strong spatial dependence, and its apparent fluctuations in time as seen by the diffusing protons produce spin transitions that contribute to both 1/T1 and /T2 comparably at low fields; for the larger particles, because of dispersion, the secular term dominates at fields of interest. On the basis of the agreement of theory with data for solutions of small paramagnetic complexes, large magnetite particles, and liver containing low-density polymer-coated magnetite agglomerates, it is argued that the theory is sufficiently reliable so that, e.g., for ferritin--for which 1/T2 is unexpectedly large--the source of its large relaxivity must reside in nonideal chemistry of the ferritin core. For blood, it appears that diffusion through intracellular gradients determines 1/T2.

It is well established that the spin-lattice magnetic relaxation rate 1/T1 of solvent protons in homogeneous protein solutions increases dramatically as the magnetic field is reduced well below the traditional NMR range. For a 5% solution of protein of 10(5) Daltons, for example, 1/T1 increases from about 50% above the pure solvent rate at 20 MHz to five times the solvent rate at 0.01 MHz. At higher fields, the effect of protein on the relaxation rate decreases progressively toward zero. 1/T1 of solvent in erythrocyte suspension behaves similarly, indicating that extracellular water has ready access to intracellular protein. We now report analogous data for samples of various mammalian tissues: we find that the data can be accommodated within the conceptual framework developed earlier for analyzing homogeneous protein solutions. It appears that tissue water probes the macromolecular composition and structure in a tissue-specific fashion. The variation of 1/T1 with field differs for each tissue, and its magnitude at low fields varies by more than a factor of three, far more than does the water content of the tissues. The relevance to contrast in NMR imaging is discussed.
