The renal vasculature of Wistar Kyoto spontaneously hypertensive rats (SHR), prior to (4-5 week) and during established hypertension (21 week) and those of age-matched Wistar Kyoto normotensive rats (WKY) were morphometrically and pharmacologically studied. Under dilated conditions, the vascular resistances (RVR) of the isolated kidneys of young and adult SHR were similar to WKY. Morphometric measurements of renal vasculature indicated that the cross-sectional area of the intima and adventitia and its subcomponents were similar in adult SHR and WKY. With the exception of the preglomerular arterioles, all the renal arteries of adult SHR exhibited elevated cross-sectional quantities of total media, medial smooth muscle cells (SMCs), and extracellular space. Analysis of the SMCs indicated the presence of increased numbers of SMC layers and/or an increase in the SMC volume-to-surface area ratio in arteries sampled from adult SHR. Vascular contraction produced by infusing norepinephrine, BaCl2, angiotensin II, or by stimulating the renal nerves elevated the RVR to a greater degree in adult SHR than in WKY. The sensitivity of the renal vasculature to the various contractile agents was similar in adult SHR and WKY. When compared with WKY, prehypertensive SHR also exhibited increased cross-sectional quantities of arterial media and elevated amplitudes of RVR change in response to norepinephrine and renal nerve stimulation. However, the vascular contractile sensitivity to norepinephrine was reduced. Our results indicate that renovascular wall thickening and the hypercontractile reactivity associated with such a change precedes hypertension in SHR. In prehypertensive SHR, elevations in RVR might be counterbalanced by a decreased norepinephrine sensitivity. An increase in the norepinephrine contractile sensitivity and further vascular thickening with age could elevate the RVR and establish hypertension.

Structural and reactivity alterations of the renal vasculature of spontaneously hypertensive rats prior to and during established hypertension.

The responses of plasma aldosterone and corticosterone to ACTH, angiotensin II (AII), and potassium chloride (KCl) infusion and the aldosterone, corticosterone and PRA responses to immobilization stress were studied in 2-month-old spontaneously hypertensive rats (SHR) and age-matched Wistar-Kyoto (WKY) normotensive controls. Basal levels of plasma aldosterone and corticosterone were greater and PRA was less in the SHR than in the WKY. Aldosterone and corticosterone responses to graded AII were similar in both groups. Aldosterone and corticosterone responses to graded doses of KCl and ACTH, however, were significantly greater in SHR than in WKY normotensive rats. Plasma corticosterone, PRA, and aldosterone responses to immobilization stress were reduced in SHR compared to WKY. Plasma Aldosterone and Corticosterone Responses to Adrenocorticotropin, Angiotensin, Potassium, and Stress in Spontaneously Hypertensive Rats

Male and female spontaneously hypertensive rats (SHR), which develop hypertension spontaneously with maturation, were autopsied at select time intervals from weaning to 28 months. Their blood pressure began to rise steeply at 4–5 weeks, reaching a zenith of 180–240 mm Hg after 4 months. Elevated blood pressures were maintained in both sexes. After 20 months, the male SHR began to die of myocardial infarction and hypotensive crisis. Heart and adrenal gland weight increased progressively not only during the phase of rapidly rising blood pressure but also during the period of plateaued but sustained high blood pressure. Aldosterone, deoxycorticosterone, corticosterone, and prolactin changes during the lifespan of chronically and spontaneously hypertensive rats.

Abnormalities in the hypothalamo-pituitary-adrenal axis in spontaneously hypertensive rats (SHR) during development of hypertension were investigated using in vivo and in vitro methods. Plasma ACTH responses to hemorrhage and ether stress were significantly smaller in 7-week-old SHR than in age-matched Wistar-Kyoto rats (WKY), while plasma corticosterone baseline levels and its response to stress were greater in SHR than in WKY. There was no significant difference in the plasma ACTH response to ether stress between bilaterally adrenalectomized SHR and WKY replaced with a 25% corticosterone pellet for 6 days. Adrenalectomy prevented the development of hypertension in SHR; however, corticosterone replacement restored hypertension. Abnormalities in the hypothalamo-pituitary-adrenal axis in spontaneously hypertensive rats during development of hypertension.

The responses to immobilization, ether, and heat of spontaneously hypertensive rats (SHR) and normotensive Sprague-Dawley rats (S-D) were compared. Hyperglycemia reached a maximum within 5 min of exposure and was most intense in SHR. There were no differences in the pituitary content of GH or PRL between the SHR and S-D. Circulating GH levels were especially depleted in SHR 15 min post exposure, with rebound to high blood levels at 1 h. Circulating PRL levels were slightly higher in quiescent SHR vs. S-D but surged to much higher levels post exposure in SHR vs. S-D. Postexposure blood corticosterone and aldosterone levels rose to much higher levels in SHR vs. S-D. Although the blood glucose, GH, PRL, corticosterone, and aldosterone levels are similar in hypertensive SHR and normotensive S-D under quiescent conditions, SHR appear to be much more sensitive and respond more vigorously to heat, ether, and immobilization. Hypersensitivity of spontaneously hypertensive rats (SHR) to heat, ether, and immobilization.

Adrenal vein catheterizations were done in SHR and control rats at different ages during the development of hypertension. All SHR became hypertensive by 15 weeks of age. The secretion rate of aldosterone was significantly reduced at 8 weeks of age, 18-hydroxy-11-deoxycorticosterone (18-OH-DOC) at 12 weeks of age, deoxycorticosterone (DOC) at twenty weeks of age, and corticosterone (B) at 12 and 20 weeks of age. Secretion data suggest either an enzyme block, or increased conversion of known steroids to an unknown steroid product. Reduced secretion of corticosterone could explain the adrenal hyperplasia observed in SHR which may be important to the development of hypertension in these animals.
