Magnetoencephalography (MEG) is a noninvasive technique for investigating neuronal activity in the living human brain. The time resolution of the method is better than 1 ms and the spatial discrimination is, under favorable circumstances, 2-3 mm for sources in the cerebral cortex. In MEG studies, the weak 10 fT-1 pT magnetic fields produced by electric currents flowing in neurons are measured with multichannel SQUID (superconducting quantum interference device) gradiometers. The sites in the cerebral cortex that are activated by a stimulus can be found from the detected magnetic-field distribution, provided that appropriate assumptions about the source render the solution of the inverse problem unique. Many interesting properties of the working human brain can be studied, including spontaneous activity and signal processing following external stimuli. For clinical purposes, determination of the locations of epileptic foci is of interest. The authors begin with a general introduction and a short discussion of the neural basis of MEG. The mathematical theory of the method is then explained in detail, followed by a thorough description of MEG instrumentation, data analysis, and practical construction of multi-SQUID devices. Finally, several MEG experiments performed in the authors' laboratory are described, covering studies of evoked responses and of spontaneous activity in both healthy and diseased brains. Many MEG studies by other groups are discussed briefly as well.

An Introduction to the Event-Related Potential Technique

Motivated by the vast amount of information that is rapidly accumulating about the human brain in digital form, we embarked upon a program in 1992 to develop a four–dimensional probabilistic atlas and reference system for the human brain. Through an International Consortium for Brain Mapping (ICBM) a dataset is being collected that includes 7000 subjects between the ages of eighteen and ninety years and including 342 mono– and dizygotic twins. Data on each subject includes detailed demographic, clinical, behavioural and imaging information. DNA has been collected for genotyping from 5800 subjects. A component of the programme uses post–mortem tissue to determine the probabilistic distribution of microscopic cyto– and chemoarchitectural regions in the human brain. This, combined with macroscopic information about structure and function derived from subjects in vivo , provides the first large scale opportunity to gain meaningful insights into the concordance or discordance in micro– and macroscopic structure and function. The philosophy, strategy, algorithm development, data acquisition techniques and validation methods are described in this report along with database structures. Examples of results are described for the normal adult human brain as well as examples in patients with Alzheimer's disease and multiple sclerosis. The ability to quantify the variance of the human brain as a function of age in a large population of subjects for whom data is also available about their genetic composition and behaviour will allow for the first assessment of cerebral genotype–phenotype–behavioural correlations in humans to take place in a population this large. This approach and its application should provide new insights and opportunities for investigators interested in basic neuroscience, clinical diagnostics and the evaluation of neuropsychiatric disorders in patients.

A probabilistic atlas and reference system for the human brain: International Consortium for Brain Mapping (ICBM)

There has been tremendous advances in our ability to produce images of human brain function. Applications of functional brain imaging extend from improving our understanding of the basic mechanisms of cognitive processes to better characterization of pathologies that impair normal function. Magnetoencephalography (MEG) and electroencephalography (EEG) (MEG/EEG) localize neural electrical activity using noninvasive measurements of external electromagnetic signals. Among the available functional imaging techniques, MEG and EEG uniquely have temporal resolutions below 100 ms. This temporal precision allows us to explore the timing of basic neural processes at the level of cell assemblies. MEG/EEG source localization draws on a wide range of signal processing techniques including digital filtering, three-dimensional image analysis, array signal processing, image modeling and reconstruction, and, blind source separation and phase synchrony estimation. We describe the underlying models currently used in MEG/EEG source estimation and describe the various signal processing steps required to compute these sources. In particular we describe methods for computing the forward fields for known source distributions and parametric and imaging-based approaches to the inverse problem.

Electromagnetic brain mapping

This article examines the role of attention and automaticity in auditory processing as revealed by event-related potential (ERP) research. An ERP component called the mismatch negativity , generated by the brain's automatic response to changes in repetitive auditory input, reveals that physical features of auditory stimuli are fully processed whether or not they are attended. It also suggests that there exist precise neuronal representations of the physical features of recent auditory stimuli, perhaps the traces underlying acoustic sensory (“echoic”) memory. A mechanism of passive attention switching in response to changes in repetitive input is also implicated. Conscious perception of discrete acoustic stimuli might be mediated by some of the mechanisms underlying another ERP component (NI), one sensitive to stimulus onset and offset. Frequent passive attentional shifts might accountforthe effect cognitive psychologists describe as “the breakthrough of the unattended” (Broadbent 1982), that is, that even unattended stimuli may be semantically processed, without assuming automatic semantic processing or late selection in selective attention. The processing negativity supports the early-selection theory and may arise from a mechanism for selectively attending to stimuli defined by certain features. This stimulus selection occurs in the form ofa matching process in which each input is compared with the “attentional trace,” a voluntarily maintained representation of the task-relevant features of the stimulus to be attended. The attentional mechanism described might underlie the stimulus-set mode of attention proposed by Broadbent. Finally, a model of automatic and attentional processing in audition is proposed that is based mainly on the aforementioned ERP components and some other physiological measures.

Retinotopic mapping strategies similar to those used for invasive electrophysiological studies to identify multiple visual areas in monkeys have been adapted for noninvasive studies in humans, using magnetic recordings of brain activity in conjunction with anatomical magnetic resonance imaging. The retinotopic organization of the primary visual area (V1) in the left hemisphere of human subjects was examined by presenting a small patterned stimuli near the vertical and horizontal meridians in the lower right visual field. In contrast with the classical model of V1 retinotopy, our results suggest that the representation of the horizontal meridian does not necessarily correspond in a one-to-one manner with the base of the calcarine fissure and that some lower field stimuli can activate regions in the lower bank of the fissure. The results also indicate significant individual variability in the details of how V1 maps around the calcarine fissure.
