The epithelia of a number of glands and cavitary organs of the rat and guinea pig have been surveyed, and in all cases investigated, a characteristic tripartite junctional complex has been found between adjacent cells. Although the complex differs in precise arrangement from one organ to another, it has been regularly encountered in the mucosal epithelia of the stomach, intestine, gall bladder, uterus, and oviduct; in the glandular epithelia of the liver, pancreas, parotid, stomach, and thyroid; in the epithelia of pancreatic, hepatic, and salivary ducts; and finally, between the epithelial cells of the nephron (proximal and distal convolution, collecting ducts). The elements of the complex, identified as zonula occludens (tight junction), zonula adhaerens (intermediary junction), and macula adhaerens (desmosome), occupy a juxtaluminal position and succeed each other in the order given in an apical-basal direction. The zonula occludens (tight junction) is characterized by fusion of the adjacent cell membranes resulting in obliteration of the intercellular space over variable distances. Within the obliterated zone, the dense outer leaflets of the adjoining cell membranes converge to form a single intermediate line. A diffuse band of dense cytoplasmic material is often associated with this junction, but its development varies from one epithelium to another. The zonula adhaerens (intermediate junction) is characterized by the presence of an intercellular space ( approximately 200 A) occupied by homogeneous, apparently amorphous material of low density; by strict parallelism of the adjoining cell membranes over distances of 0.2 to 0.5 micro; and by conspicuous bands of dense material located in the subjacent cytoplasmic matrix. The desmosome or macula adhaerens is also characterized by the presence of an intercellular space ( approximately 240 A) which, in this case, contains a central disc of dense material; by discrete cytoplasmic plaques disposed parallel to the inner leaflet of each cell membrane; and by the presence of bundles of cytoplasmic fibrils converging on the plaques. The zonula occludens appears to form a continuous belt-like attachment, whereas the desmosome is a discontinuous, button-like structure. The zomula adhaerens is continuous in most epithelia but discontinuous in some. Observations made during experimental hemoglobinuria in rats showed that the hemoglobin, which undergoes enough concentration in the nephron lumina to act as an electron-opaque mass tracer, does not penetrate the intercellular spaces beyond the zonula occludens. Similar observations were made in pancreatic acini and ducts where discharged zymogen served as a mass tracer. Hence the tight junction is impervious to concentrated protein solutions and appears to function as a diffusion barrier or "seal." The desmosome and probably also the zonula adhaerens may represent intercellular attachment devices.

Neurogenesis in the dentate gyrus of the hippocampus persists throughout life in many vertebrates, including humans. The progenitors of these new neurons reside in the subgranular layer (SGL) of the dentate gyrus. Although stem cells that can self-renew and generate new neurons and glia have been cultured from the adult mammalian hippocampus, the in vivo primary precursors for the formation of new neurons have not been identified. Here we show that SGL cells, which express glial fibrillary acidic protein and have the characteristics of astrocytes, divide and generate new neurons under normal conditions or after the chemical removal of actively dividing cells. We also describe a population of small electron-dense SGL cells, which we call type D cells and are derived from the astrocytes and probably function as a transient precursor in the formation of new neurons. These results reveal the origins of new neurons in the adult hippocampus.

a)

Tanycyten. Die mit der Goldsublimatmethode nach Cajal dargestellte Faserglia im Gehirn geschlechtsreifer Scylliorhinusarten wird vorwiegend von Gliazellen gebildet, die wenige lange, gestreckt verlaufende Fasern besitzen. Sie stellen einen besonderen Gliazelltyp dar, den ich als „Tanycyt“ bezeichne. Die Kerne der Tanycyten liegen im Ependym oder mitten in der Hirnsubstanz.

Die Gliafasern sind nicht starr und besitzen nur eine geringe Zugfestigkeit. Ihre radiare Anordnung macht eine statische Funktion unwahrscheinlich.

Wo die Hirnwand dunn ist, besteht die Tanycytenglia nur aus Zellen, deren Perikarya im Ependym liegen. Auch diese einfachste ependymale Tanycytenglia bildet verschiedene Gliaarchitekturen aus, indem Dichte, Verlauf und Bundelung variieren.




b)

Astrocyten. Neben den Tanycyten gibt es bei Scylliorhinus auch Astrocyten, besonders haufig in den kernhaltigen Zellagen. In der Schicht der Glomera olfactoria und im Ganglion interpedunculare sind sie die einzige Gliazellform.




c)

Glia und Gefaβe. Zu den Gefasen nehmen die Gliafasern entweder durch Anlagerung oder durch Fuschenbildung Beziehungen auf. Von den Gliakontakten entspringen sehr feine Gliafasern, die sich in der nervosen Substanz verteilen. An den groseren Gefasen, Arterien und Venen, sind nur wenige Gliaanlagerungen und keine Fuschen zu finden. Die Beziehungen der Astrocyten zu den Gefasen fallen weniger auf.




d)

Glia und Hirnoberflache. Die Gliafasern bilden an der Hirnoberflache Verbreiterungen mit sternformigen Auslaufern, die einander beruhren. Von diesen Verbreiterungen ziehen rucklaufig feine Faserchen in die Gehirnsubstanz.

Virchow-Robinsche Raume gibt es nur an tief einschneidenden Furchen und an einem Bindegewebsstrang, der aus einer solchen Furche entstanden ist.

Trypanblau tritt von den Ventrikeln her, nicht aus der Perilymphe in das Gehirn.




e)

Glia und nervose Anteile. Es werden regelmasige Beziehungen der Gliastruktur zu den verschiedenen Bestandteilen des Zentralnervensystems (Kerngebiete, Kornermassen, Nervenfaserstrange) aufgezeigt.




f)

Zellteilungen der Gliocyten kommen im Gehirn vor.

The fiber glia of selacean brain

The first part of the paper deals with the fine histology of the CNS as revealed by electron microscopy. The results of electron microscopic examinations of the thin ventricular walls of Scylliorhinus are described. They are in accordance with the findings in the nervous tissue of other animals. It could be shown that in the CNS of vertebrates:

1.

There is only a very small extracellular space of about 200 A width between two adjacent cells or cell processes. The close packing of cells and the absence of any gaps is made possible by a mutual distortioning and flattening of adjacent cell processes as well as by bundling of small fibres.




2.

The percentage of extracellular space increases with decreasing diameter of fibres. If the mean diameter of the fibres is about 0,08–0,1 μ, the percentage of extracellular space is about 30% of the total volume, whereas if the mean diameter of the fibres is about 1 μ, it is only 3,5%. According to the calculated relationship between fibre diameter and amount of extracellular space the measurements in electron micrographs show, that in nervous tissue the mean percentage of extracellular space does not exceed 5% of the total volume.




3.

The diameter of the smallest fibres, which could be found, was about 0,2 μ.




4.

It is not yet possible to discriminate between unmyelinated neurites and dendrites in electron micrographs. Most of the numerous lamellae-like and very small processes seem to be final branchings of glial cell processes.

Im Oberflachenrelief der Zellen nehmen die Desmosome charakteristische Orte ein.

Die Zellmembranen hangen in bestimmten Arealen, den Bizzozeroschen Knotchen (Desmosome), inniger zusammen. Im Bereich der Desmosome ist der Zellularspalt von besonderer Kittsubstanz erfullt.




4.

Tonofilamente, Einzelfaden, die eine Dicke von etwa 80 A besitzen, sind in Strahnen verschiedener Dicke, den Tonofibrillen, zusammengefast.




5.

Die Tonofilamentstrahnen setzen an den Bizzozeroschen Knotchen (Desmosome) an.




6.

Im Stratum corneum weist die Kittsubstanz der Desmosome eine komplizierte Schichtung auf. Hier sind die Zellen an ihrer ganzen Oberflache durch eine interzellulare Substanz von durchschnittlich 100 A E Dicke getrennt.




7.

Die Keratohyalinbildung geht allmahlich vor sich. Das Keratohyalin bedeckt die fibrillaren Elemente der Rattenepidermis nie vollstandig.




8.

Zellkerne und Zellorganelle — Mitochondrien, Golgisubstanz und Paladeschen Kornchen — bleiben bis zum Stratum corneum erhalten. Beim Sprung in die Verhornung verschwinden alle diese Elemente plotzlich. In der Zelle bleibt nur eine fibrillierte Masse ubrig, die den Zelleib vollstandig fullt.

Nach Injektion einer schwach alkoholischen, alkalischen Dithizonlosung farbt sich bei Igel, Meerschweinchen, Kaninchen, Ratte, Goldhamster, Katze und Hund die Rindenplatte der Fascia dentata und des Feldes h 3 (Ammonshorn) rot, wahrend die Felder h 2 und h 1 sowie das Subiculum ungefarbt bleiben. Neben der elektiven Hervorhebung homologer Abschnitte der Ammonsformation findet sich regelmasig eine Rotung im N. amygdalae. Bei manchen Tieren ist auserdem im Bereich des Striatum sowie in der basalen Rinde eine Rotfarbung festzustellen; doch handelt es sich hierbei um einen inkonstanten Befund. Neocortex, Mittel- und Kleinhirn bleiben stets ungefarbt. — Die Befunde zeigen, das cytoarchitektonisch homologe Felder sich auch in chemischer Hinsicht gleichartig verhalten konnen. Auf die Bedeutung der Befunde fur die Neuropathologie der Ammonsformation wird hingewiesen.

Die Bedeutung des Nucleolus ffir den Eiweil~stoffwechseI der Zelle ist dutch die Untersuchungen CASPERSSONS (1950) mit Hilfe der Ultraviolettabsorption hervorgehoben worden. Die im Kernk6rperchen deponierten Nucleotide sollen dutch das Karyoplasma lind die Kernmembran in das Cytoplasma geschleust werden, wobei die Kernmembran vorfibergehend breit erSffnet werde (ALTMANN 1955). Im Folgenden werden einige elektronenmikroskpoische Beobachtungen am Nucleolus und am Karyoplasma mitgeteilt, die in diesem Zusammenhang yon Interesse sein dfirften. Im KernkSrperchen sind gelegentlich lichtmikroskopisch darstellbare f/idige Strukturen beschrieben worden (G. HERTW]G1929). ESTABLE und SOTELO (1951, 1955) haben erneut auf diese Strukturen hingewiesen und sie als ,,Nucleolonemata\" besonders herausgestellt. Nach ihren yon BERNHARD, GAUTIER und OBERLING (1951), BERNHARD, GAUTIER, OBERLING und HAGUENAU (1952) sowie OBERLING und BERNHARD (1955) besti~tigten Beobachtungen besteht der Nucleolus aus plumpen F~den eines kSrnigen osmiophilen Materials und einer weniger osmiophilen amorphen Substanz (,,Pars amorpha\", ESTABLE und SOTELO). Die runden bis ovalen KSrnchen, die das Xetzwerk der ,,Nucleolonemata\" bilden, sind 100--150 A dick (BERNItARD und Mitarbeiter 1953). Eine Ordnung der Granula wurde nicht entdeckt. BERNHARD und Mitarbeiter sowie R. LETTR~ und SIEBS (1955) nehmen an, dab die F~den ehromosomale Teile darstellen. BERNHARD und Mitarbeiter finden KSrnchen der gleichen GrSi~e aueh im Karyoplasma wieder. Im Lichtmikroskop heben sich die Nueleolen meist scharf gegen das fibrige Karyoplasma ab, doch kann man im Elektronenmikroskop an ihrer Oberfls keine Membran feststellen.
