Microbial resistance to metals in the environment.

Interactions of toxic metals with fungi have long been of importance since metal toxicity still is the basis of many fungicidal preparations while in an environmental context, accelerating pollution of the natural environment has led to increased interest because of the sometimes dominant presence of fungi in metal-polluted habitats, the translocation of toxic metals and radionuclides to fruit bodies of edible higher fungi, and the significance of mycorrhizal fungi in the amelioration of metal phytotoxicity (Colpaert and Van Assche, 1987). Furthermore, the use of fungal (and other microbial) biomass for the detoxification of metal/radionuclide-containing industrial effluents is of biotechnological potential (Gadd, 1990, 1992a).

Interactions of fungip with toxic metals

Biosorption may be simply defined as the removal of substances from solution by biological material. Such substances can be organic and inorganic, and in gaseous, soluble or insoluble forms. Biosorption is a physico-chemical process and includes such mechanisms as absorption, adsorption, ion exchange, surface complexation and precipitation. Biosorption is a property of both living and dead organisms (and their components) and has been heralded as a promising biotechnology for pollutant removal from solution, and/or pollutant recovery, for a number of years, because of its efficiency, simplicity, analogous operation to conventional ion exchange technology, and availability of biomass. Most biosorption studies have carried out on microbial systems, chiefly bacteria, microalgae and fungi, and with toxic metals and radionuclides, including actinides like uranium and thorium. However, practically all biological material has an affinity for metal species and a considerable amount of other research exists with macroalgae (seaweeds) as well as plant and animal biomass, waste organic sludges, and many other wastes or derived bio-products. While most biosorption research concerns metals and related substances, including radionuclides, the term is now applied to particulates and all manner of organic substances as well. However, despite continuing dramatic increases in published research on biosorption, there has been little or no exploitation in an industrial context. This article critically reviews aspects of biosorption research regarding the benefits, disadvantages, and future potential of biosorption as an industrial process, the rationale, scope and scientific value of biosorption research, and the significance of biosorption in other waste treatment processes and in the environment. Biosorption: critical review of scientific rationale, environmental importance and significance for pollution treatment

Many algae have immense capability to sorb metals, and there is considerable potential for using them to treat wastewaters. Metal sorption involves binding on the cell surface and to intracellular ligands. The adsorbed metal is several times greater than intracellular metal. Carboxyl group is most important for metal binding. Concentration of metal and biomass in solution, pH, temperature, cations, anions and metabolic stage of the organism affect metal sorption. Algae can effectively remove metals from multi-metal solutions. Dead cells sorb more metal than live cells. Various pretreatments enhance metal sorption capacity of algae. CaCl2 pretreatment is the most suitable and economic method for activation of algal biomass. Algal periphyton has great potential for removing metals from wastewaters. An immobilized or granulated biomass-filled column can be used for several sorption/desorption cycles with unaltered or slightly decreased metal removal. Langmuir and Freundlich models, commonly used for fitting sorption data, cannot precisely describe metal sorption since they ignore the effect of pH, biomass concentration, etc. For commercial application of algal technology for metal removal from wastewaters, emphasis should be given to: (i) selection of strains with high metal sorption capacity, (ii) adequate understanding of sorption mechanisms, (iii) development of low-cost methods for cell immobilization, (iv) development of better models for predicting metal sorption, (v) genetic manipulation of algae for increased number of surface groups or over expression of metal binding proteins, and (vi) economic feasibility.

Use of Algae for Removing Heavy Metal Ions From Wastewater: Progress and Prospects

Bacteria, and other microorganisms, exhibit a number of metabolism-dependent and-independent processes or the uptake and accumulation of heavy metals and radionuclides. The removal of such harmful substances from effluents and waste waters by microbe-based technologies may provide an alternative or additional means of metal/radionuclide recovery for economic reasons and/or environmental protection. Both living and dead cells as well as products derived from or produced by microorganisms can be effective metal accumulators and there is evidence that some biomass-based clean-up processes are economically viable. However, many aspects of metal-microbe interactions remain unexploited in biotechnology and further development and application is necessary, particularly to the problem of radionuclide release into the environment.

Heavy metal accumulation by bacteria and other microorganisms

Cadmium is an important environmental pollutant and a potent toxicant to bacteria, algae, and fungi. Mechanisms of Cd toxicity and resistance are variable, depending on the organism. It is very clear that the form of the metal and the environment it is studied in, play an important role in how Cd exerts its effect and how the organism(s) responds. A wide range of Cd concentrations have been used to designate resistance in organisms. To date, no concentration has been specified that is applicable to all species studied under standardized conditions. Cadmium exerts its toxic effect(s) over a wide range of concentrations. In most cases, algae and cyanobacteria are the most sensitive organisms, whereas bacteria and fungi appear to be more resistant. In some bacteria, plasmid-encoded resistance can lead to reduced Cd2+ uptake. However, some Gram-negative bacteria without plasmids are just as resistant to Cd as are bacteria containing plasmids encoding for Cd resistance. According to Silver and Misra (1984), there is no evidence for enzymatic or chemical transformations associated with Cd resistance. Insufficient information is available on the genetics of Cd uptake and resistance in cyanobacteria and algae. Mechanisms remain largely unknown at this point in time. Cadmium is toxic to these organisms, causing severe inhibition of such physiological processes as growth, photosynthesis, and nitrogen fixation at concentrations less than 2 ppm, and often in the ppb range (Tables 2 and 3). Cadmium also causes pronounced morphological aberrations in these organisms, which are probably related to deleterious effects on cell division. This may be direct or indirect, as a result of Cd effects on protein synthesis and cellular organelles such as mitochondria and chloroplasts. Cadmium is accumulated internally in algae (Table 4) as a result of a two-phase uptake process. The first phase involves a rapid physicochemical adsorption of Cd onto cell wall binding sites, which are probably proteins and (or) polysaccharides. This is followed by a lag period and then a slow, steady intracellular uptake. This latter phase is energy dependent and may involve transport systems used to accumulate other divalent cations, such as Mn2+ and Ca2+. Some data indicate that Cd resistance, and possibly uptake, in algae and cyanobacteria is controlled by a plasmid-encoded gene(s). Although considerable information is available on Cd toxicity to, and uptake in fungi, further work is clearly needed in several areas. There is little information about Cd uptake by filamentous fungi, and even in yeasts, information on the specificity, kinetics, and mechanisms of Cd uptake is limited.(ABSTRACT TRUNCATED AT 400 WORDS)
