Questions often arise concerning the genetic stability of plant materials stored in liquid nitrogen for long time periods. This study examined the genetic stability of cryopreserved shoot tips of Rubus germplasm that were stored in liquid nitrogen for more than 12 yr, then rewarmed and regrown. We analyzed the genetic stability of Rubus grabowskii, two blackberry cultivars (“Hillemeyer” and ‘Silvan’), and one raspberry cultivar (“Mandarin”) as in vitro shoots and as field-grown plants. No morphological differences were observed in greenhouse-grown cryopreserved plants when compared to the control mother plants. In the field, cryopreserved plants appeared similar but were more vigorous than mother plants, with larger leaves, fruit, and seeds. Single sequence repeats (SSR) and amplified fragment length polymorphism (AFLP) analyses were performed on shoots immediately after recovery from cryopreservation and on shoots subcultured for 7 mo before analysis. Ten SSR primers developed from “Marion” and “Meeker” microsatellite-enriched libraries amplified one to 15 alleles per locus, with an average of seven alleles and a total of 70 alleles in the four genotypes tested. No SSR polymorphisms were observed between cryopreserved shoots and the corresponding mother plants regardless of subculture. Although no polymorphisms were detected in shoots analyzed immediately after recovery from cryopreservation, AFLP polymorphisms were detected in three of the four Rubus genotypes after they were subcultured for 7 mo. Field-grown plants from the polymorphic shoot tips of R. grabowskii and ‘Silvan’ displayed the same AFLP fingerprints as their corresponding mother plants. Only long-cultured in vitro shoot tips displayed polymorphisms in vitro, and they were no longer detected when the plants were grown ex vitro. The transitory nature of these polymorphisms should be carefully considered when monitoring for genetic stability.

Genetic stability of cryopreserved shoot tips of Rubus germplasm

Sugarcane breeding, while key to continued improvement of pest and disease resistance and to increasing sucrose and biomass yields, is constrained by the length of time taken to release a new cultivar (10–14 yr). Forty years of international research into sugarcane in vitro culture has delivered many well-developed systems that are routinely applied to research and commercial activities, namely: (a) micropropagation of genotypes; (b) production of disease-free material from excised apical meristems; (c) international germplasm exchange; (d) generation of somaclones; (e) rapid disease and pest resistance screening; and (f) germplasm conservation. This review outlines several in vitro techniques, discussing how protocols have been tailored to address pertinent research issues and exploring possible commercial applications in the sugar industry to reduce the time frame to release a new cultivar and decrease demands on resources such as land and labour.

Agronomic performance and genetic characterization of sugarcane transformed for resistance to sugarcane yellow leaf virus

Kaempferia galanga L., family Zingiberaceae, is used extensively in the preparation of both traditional and modern medicines. Buds of rhizomes of K. galanga were incubated on Murashige and Skoog (MS) medium supplemented with 1 mg/l benzyladenine and 0.5 mg/l indole-3-acetic acid (IAA) to induce shoot proliferation. Micropropagated plantlets subjected to random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) marker-based molecular profiling revealed uniform banding patterns similar to those of the mother plants. After 2 years of culture in vitro, plantlets were transplanted to the field, evaluated for different agronomic traits, and their rhizomes were subjected to biochemical profiling using quantitative and qualitative assays of essential oils. Gas chromatography and mass spectroscopy analysis of rhizome oil of micropropagated plants showed the presence of 10 major components which were similar to those detected in the mother plants, and accounted for 95.5% of the total compounds. The compound ethyl-p-methoxy cinnamate accounted for 59.5% of the total compounds detected, followed by ethyl cinnamate, 3-carene, pentadecane, borneol, bornyl acetate, delta-selinene, camphor, alpha-piene and immidazole, 5-carbonylvinyl-4-nitro. Biochemical and molecular profiling of micropropagated clones revealed that an in vitro micropropagation protocol could be effectively used for commercial propagation of true-to-type K. galanga for a stable supply of the medicinal compounds present in this plant species.

Biochemical and molecular profiling of micropropagated and conventionally grown Kaempferia galanga

Embryogenic suspension cultures of soybean (Glycine max L. cv. Iroquois) were subjected to mutagenesis using varying concentrations (1, 3, 10, and 30 mM) of ethyl methanesulfonate (EMS). Depending on the concentration of EMS used, the mean survival rate of embryogenic cultures decreased from 74 % (1 mM EMS) to 43 % after 30 mM EMS treatment. Random amplified polymorphic DNA (RAPD) analysis was used to determine whether induction of genetic variability in embryogenic cultures in response to the different EMS treatments may result in identification of polymorphic markers. Two of 35 ‘core’ primers tested revealed polymorphisms. One of the primers, OPO-01/1150, revealed polymorphism in tissue treated with 10 mM EMS, while the other primer, OPO-05/1200, revealed polymorphism in tissue treated with either 1 or 30 mM EMS. These results suggest that RAPD markers are useful in detecting mutations in embryogenic cultures of soybean.

Mutagenesis of Embryogenic Cultures of Soybean and Detecting Polymorphisms Using RAPD Markers

The RADP (Random amplified polymorphic DNA) technique was used to detect tissue-culture-induced variations in sugarcane. Plants of the Brazilian variety RB83-5486 propagated via rhizomes and via meristem cultures were studied. The polymorphism rate for 98 RAPD loci was 6.93% when the plants derived from meristems. Besides, in order to evaluate the influence of the number of subcultures on the generation of somaclonal variation, field-grown RB83-5486 plants derived from 10 meristems were studied after five subcultivations. Although different rates of polymorphism were observed, there was no direct association with the stage of subcultivation. The analysis of plants of two sugarcane varieties cultivated in vitro from meristems showed that variety RB83-5486 was more unstable than variety SP80-185.
