Journal ArticleDOI
Fluorination of 3-(3-(Piperidin-1-yl)propyl)indoles and 3-(3-(Piperazin-1-yl)propyl)indoles Gives Selective Human 5-HT1D Receptor Ligands with Improved Pharmacokinetic Profiles
M. B. Van Niel,Ian Collins,Margaret S. Beer,Howard B. Broughton,Susan Koon-Fung Cheng,Simon Charles Goodacre,Anne Heald,Karen L Locker,Angus Murray Macleod,Denise Morrison,Christopher Moyes,Desmond O'Connor,Andrew Pike,Michael Rowley,Michael G. N. Russell,Bindi Sohal,Josephine A. Stanton,Steve Thomas,Hugh M. Verrier,A. P. Watt,Castro Jl +20 more
TL;DR: The incorporation of fluorine was found to significantly reduce the pKa of the compounds, and this reduction of basicity was shown to have a dramatic, beneficial influence on oral absorption, although the effect on oral bioavailability could not always be accurately predicted.
Journal ArticleDOI
Total Synthesis and Determination of the Absolute Configuration of Epibatidine
Stephen Robert Fletcher,Raymond Baker,Mark Stuart Chambers,Richard H. Herbert,Sarah Christine Hobbs,Steven R. Thomas,Hugh M. Verrier,Alan P. Watt,Richard G. Ball +8 more
TL;DR: In this paper, the synthesis of (+)- and (-)-epibatidine (exo-2-(2-chloropyridin-5-yl)-7-azabicyclo[2.2.1]heptane) via reaction of 5-lithio-2-loropyridine with (+)-and (-)-N-BOC-7-AZA-1-one is described.
Journal ArticleDOI
Resolution of Synthetic (+)- and (-)-Epibatidine by Chiral High Performance Liquid Chromatography and Identification of the Natural Enantiomer
TL;DR: In this paper, the enantiomers (+)- and (-)-epibatidine and (−)-N-acetylepibatinidine were separated by high performance liquid chromatography using Chiral-AGP and Chiralcel OD chiral stationary phases respectively.
Journal ArticleDOI
An efficient protocol for the preparation of primary alcohols bearing a beta-chiral center via an oxazolidinone auxiliary mediated resolution, and application to the synthesis of 4,4-disubstituted piperidine substance P antagonists.
Richard Thomas Lewis,Tamara Ladduwahetty,Kevin John Merchant,Linda Elizabeth Keown,Laure Hitzel,Hugh M. Verrier,Graeme Irvine Stevenson,Angus Murray Macleod +7 more
Journal ArticleDOI
Crystallisation induced resolution of 4-fluorophenylglycine
TL;DR: In this article, a crystallization-induced resolution of Fluorophenylglycine (FluorPHG) was used to achieve 65% overall yield via its methyl ester.
