So far in this course we have dealt entirely with the evolution of characters that are controlled by simple Mendelian inheritance at a single locus. There are notes on the course website about gametic disequilibrium and how allele frequencies change at two loci simultaneously, but we didn’t discuss them. In every example we’ve considered we’ve imagined that we could understand something about evolution by examining the evolution of a single gene. That’s the domain of classical population genetics. For the next few weeks we’re going to be exploring a field that’s actually older than classical population genetics, although the approach we’ll be taking to it involves the use of population genetic machinery. If you know a little about the history of evolutionary biology, you may know that after the rediscovery of Mendel’s work in 1900 there was a heated debate between the “biometricians” (e.g., Galton and Pearson) and the “Mendelians” (e.g., de Vries, Correns, Bateson, and Morgan). Human biochemical genetics

Speciation is characterized by the evolution of reproductive isolation between two groups of organisms. Understanding the process of speciation requires the quantification of barriers to reproductive isolation, dissection of the genetic mechanisms that contribute to those barriers and determination of the forces driving the evolution of those barriers. Through a comprehensive analysis involving 19 pairs of plant taxa, we assessed the strength and patterns of asymmetry of multiple prezygotic and postzygotic reproductive isolating barriers. We then reviewed contemporary knowledge of the genetic architecture of reproductive isolation and the relative role of chromosomal and genic factors in intrinsic postzygotic isolation. On average, we found that prezygotic isolation is approximately twice as strong as postzygotic isolation, and that postmating barriers are approximately three times more asymmetrical in their action than premating barriers. Barriers involve a variable number of loci, and chromosomal rearrangements may have a limited direct role in reproductive isolation in plants. Future research should aim to understand the relationship between particular genetic loci and the magnitude of their effect on reproductive isolation in nature, the geographical scale at which plant speciation occurs, and the role of different evolutionary forces in the speciation process.

The strength and genetic basis of reproductive isolating barriers in flowering plants

Pollination commonly limits seed production, as addition of pollen to stigmas often increases fecundity. This response is usually interpreted as evidence that plants' stigmas receive too few pollen grains to maximize ovule fertilization (quantity limitation); however, many genetic studies demonstrate that poor-quality pollen can also reduce seed production (quality limitation). We explore both aspects of pollen limitation theoretically with a dose-response model that incorporates a saturating negative-exponential relation of seed production to pollen receipt. This relation depends on aspects of ovule production, pollen import, pollen-pistil interactions and seed development, all of which can contribute to pollen limitation. Our model reveals that quantity limitation is restricted to the lowest range of pollen receipt, for which siring success per pollen grain is high, whereas quality limitation acts throughout the range of pollen receipt if plants do not import the highest-quality pollen. In addition to pollinator availability and efficiency, quantity limitation is governed by all post-pollination aspects of seed production. In contrast, quality limitation depends on the difference in survival of embryos sired by naturally delivered pollen vs. by pollen of maximal quality. We briefly illustrate the distinction between these two components of pollen limitation with results from the mistletoe Tristerix corymbosus. Our model also shows that the standard pollen-supplementation technique neither estimates the total intensity of pollen limitation nor distinguishes between its quantity and quality components. As an alternative, we propose a methodological protocol that requires both measurement of seed production following excess pollination with only outcross pollen and quantification of the dose-response relation of seed output to pollen receipt. This method estimates both the total extent of pollen limitation and its two components. Finally, we consider the influences on quantity and quality limitation, which reveals that quantity limitation probably occurs much less often than has been inferred from pollen-supplementation experiments. These interpretations suggest that an expanded perspective that recognizes the fecundity consequences of pollination with poor-quality pollen would promote ecological understanding of pollen limitation.

Expanding the limits of the pollen-limitation concept: effects of pollen quantity and quality.

Accurate phylogenies are crucial for understanding evolutionary processes, especially species diversification. It is commonly assumed that “good” species are sufficiently isolated genetically that gene genealogies represent accurate phylogenies. However, it is increasingly clear that good species may continue to exchange genetic material through hybridization (introgression). Many studies of closely related species reveal introgression of some genes without others, often with more rapid introgression of maternally inherited chloroplast or mitochondrial DNA (cpDNA, mtDNA). We seek a general explanation for this biased introgression using simple models of common reproductive isolating barriers (RIBs). We compare empirically informed models of prezygotic isolation (for pre- and postinsemination mechanisms of both female choice and male competition) with postzygotic isolation and demonstrate that rate of introgression depends critically upon type of RIB and mode of genetic inheritance (maternal versus biparental versus paternal). Our frequency-dependent prezygotic RIBs allow much more rapid introgression of biparentally and maternally inherited genes than do commonly modeled postzygotic RIBs (especially maternally inherited DNA). After considering the specific predictions in the context of empirical observations, we conclude that our model of prezygotic RIBs is a general explanation for biased introgression of maternally inherited genomic components. These findings suggest that we should use extreme caution when interpreting single gene genealogies as species phylogenies, especially for cpDNA and mtDNA.

Leaky prezygotic isolation and porous genomes: rapid introgression of maternally inherited dna

Healthy males are likely to have higher mating success than unhealthy males because of differential expression of condition-dependent traits such as mate searching intensity, fighting ability, display vigor, and some types of exaggerated morphological characters. We therefore expect that most new mutations that are deleterious for overall fitness may also be deleterious for male mating success. From this perspective, sexual selection is not limited to influencing those genes directly involved in exaggerated morphological traits but rather affects most, if not all, genes in the genome. If true, sexual selection can be an important force acting to reduce the frequency of deleterious mutations and, as a result, mutation load. We review the literature and find various forms of indirect evidence that sexual selection helps to eliminate deleterious mutations. However, direct evidence is scant, and there are almost no data available to address a key issue: is selection in males stronger than selection in females? In addition, the total effect of sexual selection on mutation load is complicated by possible increases in mutation rate that may be attributable to sexual selection. Finally, sexual selection affects population fitness not only through mutation load but also through sexual conflict, making it difficult to empirically measure how sexual selection affects load. Several lines of enquiry are suggested to better fill large gaps in our understanding of sexual selection and its effect on genetic load.

Purging the genome with sexual selection : reducing mutation load through selection on males

Sexual selection has traditionally been used to explain exaggerated sexual traits in male animals. Today the concept has been developed and various other sexually related traits have been suggested to evolve in the same manner. In nearly all new areas where the theory of sexual selection has been applied, there has been an intense debate as to whether the application is justified. Is it the case that some scientists are all too ready to employ fashionable ideas? Or are there too many dogmatic researchers refusing to accept that science develops and old ideas are transformed? Maybe the controversies are simply a reflection of the difficulty of defining a theory under constant re-evaluation. Thus, we begin by summarizing the theory of sexual selection in order to assess the influence of sexual selection on the evolution of plant morphology. We discuss empirical findings concerning potentially affected traits. Although we have tried to address criticisms fairly, we still conclude that sexual selection can be a useful tool when studying the evolution of reproductive traits in plants. Furthermore, by including the evidence from an additional kingdom, a fuller understanding of the processes involved in sexual selection can be gained.

Plants have evolved many mechanisms to increase the chance of gene dispersal mainly through pollen and environmental factors play an important role. Understanding the mechanism behind gene dispersal is therefore crucial in the correct evaluation of the use of genetically modified crops for cultivation. In this paper we address the question of weather nutrient availability for the female affects the outcome of pollen competition between two pollen donor cultivars of Cucumis sativus. We do this by carrying out controlled crosses of female plants grown at three different nutrient levels. We separated the effect of a specific donor from the effect of pollen tube growth rate by using reversed crosses of fast and slow pollen. Our results show that female effects on siring ability vary with nutrient level. Pollen with a high pollen tube growth rate was more successful when nutrient availability for the female was high. This could be the result of selection on the female to adjust preference according to environmental circumstances. Pollen tube growth rate was measured under nutrient rich circumstances, thus high performers possessed traits adapted to a nutrient rich situation. Due to trade-off effects, these traits might not be advantageous in poor environments. Instead, individuals adapted to low nutrient circumstances will have a higher pollen tube growth rate. If siring ability varies with the environment of the recipient plant, this means that assessments of gene flow must account for this variation and include both pollen donors and recipient plants subjected to a range of environmental circumstances. In risk assessments of transgenic plants, plants are often kept under experimental, homogenous conditions. If our results also apply to other species, estimates of gene flow under constant conditions may be misleading. Selection on siring ability and female preference have fundamental effects on gene flow and need to be considered in risk assessments of transgenic plants.

Effects of Nutrient Level on Maternal Choice and Siring Success in Cucumis sativus (Cucurbitaceae)

We investigated whether resistant pollen genotypes can be selected for when the maternal plants are infected and whether infection can result to changes in the pistil nutrient level influencing reproductive outcome. Both resistance and susceptibility come with costs that may affect pre- and post-fertilization reproductive functions. We performed the study by crossing zucchini yellow mosaic virus resistant and susceptible pollen (from a hybrid donor) to infected and healthy maternal plants. We also analysed the pistil nutrient content in both treatments and found an increase of the susceptible and not resistant genotypes when maternal plants were infected. The level of nutrients K, P and S was higher in the pistils of the infected maternal plants than the healthy ones. Pistil nutrient level did not affect pollen tube growth rates, as indicated by seed siring patterns along the fruit. We propose that the effect on the siring ability of pollen carrying the susceptible and resistant alleles occurred at the post-fertilization stage, possibly as an indirect result of different growth rates of the two embryo genotypes under elevated nutrient conditions. We discuss our results with respect to possibilities of differential selection, costs and reproductive implications.

Pathogen infection and selection on fertilization success in Cucumis sativus

The environment in which a pollen donating plant grows is very important as it may influence pollen performance. Here we examine effects of herbivory on pollen performance in two cultivars of Nicotiana alata with red and white flowers. Plants used as pollen donors, were subjected to varying degrees of simulated herbivory and were compared to control plants. We grew pollen from all individuals in vitro to evaluate inherent male competitive ability. Hand pollinations were made to study the effects of simulated herbivory on in vivo siring ability, which in turn is the result of interactions between both the male and the female reproductive functions. Herbivory had an impact on both in vitro and in vivo pollen performance. In both cultivars, in vitro pollen tube growth rate from the control plants was higher than that in pollen from moderately and highly treated plants. The siring ability of the red cultivar decreased with increase in herbivory intensity, while the white cultivar was positively affected. The differences found between the two cultivars suggest that the genotype of the pollen grain influences how pollen from individual plants reacts to different environmental factors.
