Brewer's yeast polysaccharides — A review of their exquisite structural features and biomedical applications

Despite being widely available, Saccharomyces cerevisiae has not been widely explored for direct extraction of chitosan biopolymer for antimicrobial applications. In our study, S. cerevisiae from Baker’s yeast and Aspergillus niger from moldy onion extracts are studied as alternative sources of chitosan; and S cerevisiae chitosan tested for antimicrobial efficacy. The properties of S. cerevisiae chitosan are compared with moldy onion chitosan and shrimp chitosan extracted from shrimp shells. Chitosan extracted from S. cerevisiae is tested for antimicrobial efficacy against Staphylococcus Aureus. The maximum yields of fungal chitosan are 20.85 ± 0.35 mg/g dry S. cerevisiae biomass at 4th day using a culture broth containing sodium acetate, and 16.15 ± 0.95 mg/g dry A. niger biomass at 12th day. The degree of deacetylation (DD%) of the extracted fungal chitosan samples from S. cerevisiae and A. niger is found to be 63.4%, and 61.2% respectively, using Fourier Transform Infrared Spectroscopy. At a concentration of 2 g/L, S. cerevisiae chitosan shows the maximum inhibition zone diameter of 15.48 ± 0.07 mm. Baker’s yeast S cerevisiae biomass and A. niger from moldy onions has not been previously explored as a source of extractible fungal chitosan. This study gives insight that S. cerevisiae and A. niger from agricultural or industrial wastes could be a potential biomass source for production of the chitosan biopolymer. The S. cerevisiae chitosan displayed effective antimicrobial properties against S aureus, indicating the viablitiy of S cerevisae as a resource for extraction of high-quality chitosan.

Saccharomyces Cerevisiae as an Untapped Source of Fungal Chitosan for Antimicrobial Action.

Two experiments were conducted to determine the impact of various combinations of yeast-based direct fed microbials (DFM) in diets fed to nursery pigs weaned from sows fed lactation diets with or without yeast additives. In Exp. 1, 340 weaned pigs, initially 5.1 kg ± 0.02, were used to evaluate previous sow treatment (control vs yeast additives) and nursery diets with or without added yeast-based DFM on growth performance and antimicrobial resistance (AMR) patterns of fecal Escherichia coli. Treatments were arranged in a 2 × 2 factorial with main effects of sow treatment (control vs. yeast-based pre- and probiotic diet; 0.10% ActiSaf Sc 47 HR+ and 0.025% SafMannan, Phileo by Lesaffre, Milwaukee, WI) and nursery treatment (control vs. yeast-based pre- and probiotic diet; 0.10% ActiSaf Sc 47 HR+, 0.05% SafMannan, and 0.05% NucleoSaf from d 0 to 7, then concentrations were decreased by 50% from d 7 to 24) with 5 pigs per pen and 17 replications per treatment. Progeny from sows fed yeast additives had increased (P < 0.05) average daily gain (ADG) from d 0 to 24 and d 0 to 45. However, pigs that were fed yeast additives for the first 24 d in the nursery tended to have decreased d 0 to 45 ADG (P = 0.079). Fecal E. coli isolated from pigs from the sows fed yeast group had increased (P = 0.034) resistance to nalidixic acid and a tendency for increased resistance to ciprofloxacin (P = 0.065) and gentamicin (P = 0.054). Yet, when yeast additives were added in the nursery there was reduced (P < 0.05) fecal E. coli resistance to azithromycin and chloramphenicol. In Exp. 2, 330 weaned pigs, initially 5.8 kg ± 0.03, were used to evaluate diets with two different combinations of DFM on growth performance. Treatments were arranged in a 2 × 3 factorial with main effects of sow treatment (same as described in Exp. 1) and nursery treatment (control; YCW, 0.05% of SafMannan from d 0 to 38 and NucleoSaf at 0.05% from d 0 to 10 and 0.025% from d 10 to 24; or DFM, 0.10% MicroSaf-S from d 0 to 38 and NucleoSaf at 0.05% from d 0 to 10 and 0.025% from d 10 to 24) with 6 pigs per pen and 8 to 10 replications per treatment. From d 0 to 10 post-weaning, progeny of sows fed yeast additives had increased (P < 0.05) ADG and G:F. In conclusion, feeding sows yeast through lactation improved offspring growth performance in the nursery. While feeding live yeast and yeast extracts reduced nursery pig performance in Exp. 1, feeding DFM improved growth later in the nursery period in Exp. 2.

Influence of yeast-based pre- and probiotics in lactation and nursery diets on nursery pig performance and antimicrobial resistance of fecal Escherichia coli.

Spent yeast is one of the main fermentation process by-products and several bioactive ingredients have been extracted from it, such as mannans and β-glucans. These extraction processes generate waste streams rich in protein, that can still be used to produce peptide-rich extracts. This work describes the use of a scalable and low-cost process to obtain yeast peptide extracts in a circular economy concept. Peptide fractions produced contained 48.3 to 86.4% of protein ( w /w) and essential amino acids amounts higher than those defined by FAO and WHO references. Regarding their bioactive properties, extracts exhibited strong antihypertensive and antioxidant activities. Furthermore, an inhibition of 62.0–71.3% in HMG-CoA reductase was observed, a feature hereby described for the first time, thus stating the cholesterol-lowering capacity of yeast peptides. In conclusion, due to their combined nutritional and bioactive characteristics, the produced yeast peptide extracts are good candidates for dietary supplementation and functional foods. • Spent yeast peptides were studied as a potential ingredient for diet supplement. • A sustainable, scalable and low-cost process was used to produce peptide fractions. • Fractions consisted of small peptides with a good essential amino acid balance. • Strong antioxidant and antihypertensive were recorded in the obtained fractions. • For the first time, cholesterol-lowering activities were reported on yeast protein.

Peptide-rich extracts from spent yeast waste streams as a source of bioactive compounds for the nutraceutical market

In the brewing process, the consumption of resources and the amount of waste generated are high and due to a lot of organic compounds in waste-water, the capacity of natural regeneration of the environment is exceeded. Residual yeast, the second by-product of brewing is considered to have an important chemical composition. An approach with nutritional potential refers to the extraction of bioactive compounds from the yeast cell wall, such as β-glucans. Concerning the potential food applications with better textural characteristics, spent brewer’s yeast glucan has high emulsion stability and water-holding capacity fitting best as a fat replacer in different food matrices. Few studies demonstrate the importance and nutritional role of β-glucans from brewer’s yeast, and even less for spent brewer’s yeast, due to additional steps in the extraction process. This review focuses on describing the process of obtaining insoluble β-glucans (particulate) from spent brewer’s yeast and provides an insight into how a by-product from brewing can be converted to potential food applications.

Spent Brewer's Yeast as a Source of Insoluble β-Glucans.

The aim of this study was to develop films based on β-glucans in association with pomegranate juice for its potential in metabolic disorders such as diabetes due to plenty of bioactive compounds from the film composition. Initially, a Box-Behnken design was generated by varying the level of β-glucan content (0.5, 1, 1.5 g), sodium alginate (0.2, 0.4, 0.6 g) and pomegranate juice (10, 20, 30 mL) for development of films. Subsequently, glycerin was added as 25% of the total dry matter. The optimization of the films prepared by the solvent casting method was conducted based on the different responses such as: water vapor transmission rate (WVTR), water vapor permeability (WVP), thickness, density, moisture content, solubility, film opacity and color. The water activity profile and FT-IR analysis were performed in all tests. The model was used to determine the optimal experimental values considering that the optimal film will make a sustained contribution to diabetes. The optimal values of the film sample made of β-glucans, sodium alginate, pomegranate juice and glycerin make it befitting for packaging dry powdered pharmaceuticals. Finally, antimicrobial activity against Gram-negative and Gram-positive bacteria, UV barrier properties and microcrack and pore detections through SEM were also investigated for the optimal film sample.
