Synthesis of transportation fuels from biomass: chemistry, catalysts, and engineering.

Fundamental features of microbial cellulose utilization are examined at successively higher levels of aggregation encompassing the structure and composition of cellulosic biomass, taxonomic diversity, cellulase enzyme systems, molecular biology of cellulase enzymes, physiology of cellulolytic microorganisms, ecological aspects of cellulase-degrading communities, and rate-limiting factors in nature. The methodological basis for studying microbial cellulose utilization is considered relative to quantification of cells and enzymes in the presence of solid substrates as well as apparatus and analysis for cellulose-grown continuous cultures. Quantitative description of cellulose hydrolysis is addressed with respect to adsorption of cellulase enzymes, rates of enzymatic hydrolysis, bioenergetics of microbial cellulose utilization, kinetics of microbial cellulose utilization, and contrasting features compared to soluble substrate kinetics. A biological perspective on processing cellulosic biomass is presented, including features of pretreated substrates and alternative process configurations. Organism development is considered for "consolidated bioprocessing" (CBP), in which the production of cellulolytic enzymes, hydrolysis of biomass, and fermentation of resulting sugars to desired products occur in one step. Two organism development strategies for CBP are examined: (i) improve product yield and tolerance in microorganisms able to utilize cellulose, or (ii) express a heterologous system for cellulose hydrolysis and utilization in microorganisms that exhibit high product yield and tolerance. A concluding discussion identifies unresolved issues pertaining to microbial cellulose utilization, suggests approaches by which such issues might be resolved, and contrasts a microbially oriented cellulose hydrolysis paradigm to the more conventional enzymatically oriented paradigm in both fundamental and applied contexts.

Microbial cellulose utilization: fundamentals and biotechnology.

Lignocellulosic biomass has long been recognized as a potential sustainable source of mixed sugars for fermentation to biofuels and other biomaterials. Several technologies have been developed during the past 80 years that allow this conversion process to occur, and the clear objective now is to make this process cost-competitive in today's markets. Here, we consider the natural resistance of plant cell walls to microbial and enzymatic deconstruction, collectively known as "biomass recalcitrance." It is this property of plants that is largely responsible for the high cost of lignocellulose conversion. To achieve sustainable energy production, it will be necessary to overcome the chemical and structural properties that have evolved in biomass to prevent its disassembly.

Border Gateway Protocol (BGP) is the protocol backing the core routing decisions on the Internet. It maintains a table of IP networks or 'prefixes' which designate network reachability among autonomous systems (AS). Point of concern in BGP is its lack of effective security measures which makes Internet vulnerable to different forms of attacks. Many solutions have been proposed till date to combat BGP security issues but not a single one is deployable in practical scenario. Any security proposal with optimal solution should offer adequate security functions, performance overhead and deployment cost. This paper critically analyzes the deployment issues of best three proposals considering trade-off between security functions and performance overhead.

ABSTRACT A range of game-changing biodigital and biodesign technologies are coming of age all around us, transforming our world in complex ways that are hard to predict. Not a day goes by without news of how data-centric engineering, algorithm-driven modelling, and biocyber technologies—including the convergence of artificial intelligence, machine learning, automated robotics, quantum computing, and genome editing—will change our world. If we are to be better at expecting the unexpected in the world of wine, we need to gain deeper insights into the potential and limitations of these technological developments and advances along with their promise and perils. This article anticipates how these fast-expanding bioinformational and biodesign toolkits might lead to the creation of synthetic organisms and model systems, and ultimately new understandings of biological complexities could be achieved. A total of four future frontiers in wine yeast research are discussed in this article: the construction of fully synthetic yeast genomes, including minimal genomes; supernumerary pan-genome neochromosomes; synthetic metagenomes; and synthetic yeast communities. These four concepts are at varying stages of development with plenty of technological pitfalls to overcome before such model chromosomes, genomes, strains, and yeast communities could illuminate some of the ill-understood aspects of yeast resilience, fermentation performance, flavour biosynthesis, and ecological interactions in vineyard and winery settings. From a winemaker's perspective, some of these ideas might be considered as far-fetched and, as such, tempting to ignore. However, synthetic biologists know that by exploring these futuristic concepts in the laboratory could well forge new research frontiers to deepen our understanding of the complexities of consistently producing fine wines with different fermentation processes from distinctive viticultural terroirs. As the saying goes in the disruptive technology industry, it take years to create an overnight success. The purpose of this article is neither to glorify any of these concepts as a panacea to all ills nor to crucify them as a danger to winemaking traditions. Rather, this article suggests that these proposed research endeavours deserve due consideration because they are likely to cast new light on the genetic blind spots of wine yeasts, and how they interact as communities in vineyards and wineries. Future-focussed research is, of course, designed to be subject to revision as new data and technologies become available. Successful dislodging of old paradigms with transformative innovations will require open-mindedness and pragmatism, not dogmatism—and this can make for a catch-22 situation in an archetypal traditional industry, such as the wine industry, with its rich territorial and socio-cultural connotations.

LL IN ACCORD with Louis Pasteur’s famous dictum that there is not pure science and applied science but only the application of science, useful practical outcomes often start with a twinkle in a basic researcher’s blurred, but alert, eye. Pasteur’s own research reflects an impressive synthesis of goals of understanding to reach the applied goals of preventing spoilage in milk, vinegar, beer and wine, and of conquering flacherie in silkworms, anthrax in sheep and cattle, cholera in chickens, and rabies in animals and humans. The history of science is full of examples where an open mind has made important insights in areas not originally intended at the outset of the research. However, as Albert Einstein once said, in science, chance favours the prepared mind. In addition to the benefits of defined outcomes formulated by appropriate practitioner/researcher interactions (see Hoj et al. 2003), the occasional serendipitous uncovering of phenomena is one of the important but often overlooked benefits of both public and industry investment in R&D. Alexander Flemming’s discovery of penicillin represents a famous and monumental example of such serendipity at play. Another example of famous spin-offs from ongoing fundamental research was the development of the worldwide web which owes much of its existence to the need for physicists from a multitude of nations to communicate effectively in relation to the conduct of experiments at the CERN particle physics facilities in Europe. During The Australian Wine Research Institute’s ongoing research into the causes and potential prevention of protein hazes in wine, unintended discoveries, although on a much less monumental scale than that of penicillin and the Web, have been made in relation to haze-forming proteins. It has been discovered that haze-forming proteins have a potential use in varietal identification of grapes/juices and hence, by implication, also label integrity programs. Varietal authentication of grapes, juices, musts and wines is important, not only to grapegrowers and winemakers, but also to the marketing of wines, since wine labelling laws and trade regulations now demand that varietals be correctly identified (Pinder and Meredith 2003). This article describes a prime example of an effort that brings together informed judgments of scientific promise of basic research and of industry need for varietal identification.

The unfolded protein response (UPR) is a highly conserved cellular response in eukaryotic cells to counteract endoplasmic reticulum (ER) stress, typically triggered by unfolded protein accumulation. In addition to its relevance to human diseases like cancer cell development, the induction of the UPR has a significant impact on recombinant protein production yields in microbial cell factories, including the industrial workhorse Saccharomyces cerevisiae. Being able to accurately detect and measure this ER stress response in single cells, enables the rapid optimisation of protein production conditions and high-throughput strain selection strategies. Current methodologies to monitor the UPR in S. cerevisiae are often temporally and spatially removed from the cultivation stage, or lack updated systematic evaluation. To this end we constructed and systematically evaluated a series of high-throughput UPR sensors by different designs, incorporating either yeast native UPR promoters or novel synthetic minimal UPR promoters. The native promoters of DER1 and ERO1 were identified to have suitable UPR biosensor properties and served as an expression level guide for orthogonal sensor benchmarking. Our best synthetic minimal sensor, SM1, was only 98 bp in length, had minimal homology to other native yeast sequences and displayed superior sensor characteristics. Using this synthetic minimal UPR sensor, we demonstrate its ability to accurately discriminate between cells expressing different heterologous proteins and at varying production levels. Our sensor is thus a novel high-throughput tool for determining expression/engineering strategies for optimal heterologous protein production.
